猪源结肠小袋纤毛虫培养、分子鉴定和显微观察

Cultivation, molecular identification and microscopic observation of Balantidium coli isolated from pig

摘要: 小袋纤毛虫病是一种人畜共患寄生虫病。本研究从浙江地区病猪盲肠内分离到了一株结肠小袋纤毛虫,通过RPMI1640培养基对其进行了体外培养,结肠小袋纤毛虫在28℃和37℃均能良好生长,在培养后第5天和第 4天分别达到高峰值,虫体密度分别为160×104和231×104 个·mL-1。显微镜下滋养体胞口、胞肛和食物泡结构明显,DAPI染色后大核明显呈哑铃型,且滋养体有大小差别明显的2种类型。18S rRNA基因PCR扩增、测序和聚类分析结果显示,猪源结肠小袋纤毛虫分离株存在2条明显不同的18S rRNA基因序列,序列间趋异性为06%,且该分离株序列与国外分离株间存在显著差异,在进化树上形成一独立的亚枝。在高倍显微镜下,滋养体内常见有许多生物活动,种类不明,16S rRNA基因PCR扩增到3条未知细菌序列,未知细菌16S rRNA基因序列与结肠小袋纤毛虫内微生物间的确切关系有待研究。

Abstract: Balantidiosis is a cosmopolitan parasitic zoonosis. In the study, a Balantidium coli was isolated from the cecum of a severely diarrheal pig in Zhejiang province of China. RPMI1640 medium was used to cultivate the trophozoites of B.coli. Trophozoites grew well in the RPMI1640 medium at 28℃ or 37℃ and the maximum growth density of 818×103 and 231×104cells·mL-1 was gained on the fifth and fourth day post the inoculation, respectively. Cytostome, cytopyge and food vacuole were obviously observed under the light microscopy in the trophozoites. Macronucleus obviously showed dumbbell\|like after DAPI staining. Two different types of trophozoites, large one and small one, existed in B.coli. And two different sequences of 18s rRNA gene were gained for the B.coli after the PCR and DNA sequencing. Divergence between the two sequences was 06%. The two sequences formed a single branch separated from the reference 18s rRNA gene sequences of B.coli in the phylogenetic tree. In addition, a lot of microorganisms were seen at high magnification of microscopy in the B.coli. Species for these microorganisms were unknown. Three novel sequences of 16s rRNA gene was gained after the PCR with B.coli culture as the template. Relationship between the three novel sequences and the endocytobionts was needed to be further confirmed.
