麒麟火焰兰叶片及根类原球茎的诱导研究

摘要: 为了建立新品种麒麟火焰兰(Renanthera Tom Thumb ‘Qilin’)的高效离体再生体系,以其组培苗的叶片及根作为外植体,比较不同生长调节剂配比以及根的不同处理方式对诱导类原球茎(PLBs)的影响。试验结果显示,以叶片作为外植体,在不同浓度的6\|BA与NAA配比下,均能诱导形成类原球茎,诱导率最高可达63%,最佳培养基配方为:1/2 MS+015 g·L-1柠檬酸+6 mg·L-1 6\|BA+02 mg·L-1 NAA;而以其根作为外植体,较难诱导出类原球茎。表明在火焰兰类原球茎再生体系的建立中,宜选用叶片作为外植体。

Abstract: In order to establish the high efficient regeneration system of Renanthera Tom Thumb “Qilin”, its leaves and roots were used as explants to investigate the effects of different growth regulators combinations and different treatments of roots on protocorm\|like bodies(PLBs)formation. The results showed that leaves of Renanthera Tom Thumb “Qilin” cultivated in different combinations of concentrations of 6\|BA and NAA could induce PLBs easily. The highest induction rate reached 6333% and the best culture medium was 1/2 MS+015 g · L-1 citric acid+6 mg · L-1 6\|BA+02 mg · L-1 NAA. While on the other hand, it was difficult for roots to induce PLBs. It was indicated that leaves were more suitable for rapid propagation in vitro as explants.
