狼蛛对黑腹果蝇捕食数量的PCR检测

摘要: 为探究狼蛛对黑腹果蝇的捕食能力,以黑腹果蝇和狼蛛为研究对象,分别提取喂食过1,2,3,4,5,6只黑腹果蝇的狼蛛DNA,以合成黑腹果蝇线粒体COI基因通用引物作为引物进行PCR扩增。结果表明,PCR扩增结果受模板中DNA含量的影响,并且捕食量与扩增结果呈正线性相关关系,线性方程为E(y)=-29111+18212 x,P=001,相关系数R2=095,说明狼蛛捕食黑腹果蝇数量越多,其体内含有的黑腹果蝇DNA残留就越多。

Abstract: In order to study the predation capacity of tarantula to Drosophila melanogaster, the genomic DNA of tarantulas which had been fed with 1, 2, 3, 4, 5 and 6 Drosophila melanogasters were used as templates, and PCR amplifications were carried out by utilizing the universal primers for mitochondrial synthetic gene COI of Drosophila melanogaster as the specific primers. Results showed that the PCR amplifications were influenced by the content of templates, a linear positive correlation was found between feeding amount and PCR products, and the linear equation was E(y)=-29111+18212x, P=001, R2=095. Thus, DNA residues of Drosophila melanogaster in tarantula increased with the addiation of predation.
