鸭疫里默氏杆菌原生质体制备与再生的研究

摘要: 以耐药标记的鸭疫里默氏杆菌TXRAl(CAd ,Elf)为亲本菌株,在DNase I始终存在的条件下,采用Lysozyme-EDTA法制备原生质体,然后在高渗琼脂平板上再生。系统地优化了影响原生质体制备及再生的各种条件,摸索出鸭疫里默氏杆菌TXRA1以Lysozyme 0.1 g/L作用20 min后补加EDTA至终浓度为0.01 mol/L继续作用45 min为最佳试验条件,并成功获得了92%的原生质体制备率和38.77%的再生率。

Abstract: Using Riemerella anatipestifer TXRA1(Chl r,Ery s)strain with drug resistance label as the parental strain,protoplasts were pfe with lysozyme and EDTA at the presence of DNase I,then the protoplasts were regenerated on hypertonic
medium.Conditions of protoplast formation and regeneration were optimized.indicating that the optimum Riemerella anatipestifer TXRA1(Chl r,Ery s)added with 0.1 g/L lysozyme and reacted for 20 min and then EDTA Was added slowly to a final concentration of O.O1 mol/L and reacted for another 45 min, in which the protoplast formation rate was92% and the regeneration rate was 38.77% .
