秀珍菇子实体发育相关消减杂交cDNA文库的构建与分析

摘要: 秀珍菇(Pleurotus pulmonarius)经冷刺激后可以诱发子实体的形成。为了更好地了解秀珍菇在冷刺激后子实体发育的机理,本文利用抑制性消减杂交技术构建了cDNA文库。基于该文库,共检测到185条高质量的EST差异序列。根据序列比对分析,136条序列与已知基因同源,49条序列属功能未知。生物信息学分析显示136条序列按功能分属于23个大类。这些研究为秀珍菇变温结实的分子机理探究提供理论基础。

Abstract: A transient cold condition can induce the fruiting body formation of P. pulmonarius. A subtractive cDNA library using suppressive subtractive hybridisation (SSH) was constructed for a better understanding of the molecular basis of fruitingbody formation of P. pulmonarius after cold stimulation. Based on the SSH library,a total of 185 highquality ESTs were identified by upregulated expression. BLAST searching against the Basidiomycota genomes from JGI showed that 136 EST sequences were homologous to previously known genes,and there were 49 sequences with unknown function. A total of 139 ESTs were categorized into 23 functional groups. This information can help us better understand the molecular basis underlying the fruiting induced by cold stimulation in P. pulmonarius.
