摘要: 研究旨在分析谷氨酰胺(Gln)对基础状态乳腺上皮细胞生长、增殖及热休克蛋白70(HSP70)表达的影响。取对数生长期的奶牛乳腺上皮细胞用于试验,细胞在含有不同浓度(0,05,10,20,40,80和160 mmol·L-1)Gln的DMEM培养液中培养24 h,收取细胞及培养液;细胞在含8 mmol·L-1 Gln的DMEM培养液中培养不同时间(0,3,6,12,24,48,72 h)后,收取细胞及细胞培养液。采用MTT法检测细胞存活率,比色法检测LDH活性,SYBR GreenⅠ荧光定量PCR检测细胞内热休克转录因子1(HSF\|1)和HSP70的表达水平。结果显示:Gln能促进奶牛乳腺上皮细胞生长增殖,Gln浓度为20 mmol·L-1时细胞存活率最高,40~80 mmol·L-1时仍维持较高水平;细胞内HSP70与HSF\|1的表达趋势一致,80 mmol·L-1的Gln诱导24 h,二者表达量均达峰值,分别为细胞活性最佳剂量20 mmol·L-1表达量的849倍(P<001)和590倍(P<001),表明Gln能促进奶牛乳腺上皮细胞的生长、增殖,并通过启动HSF\|1 mRNA的转录活性诱导基础状态乳腺上皮细胞高表达HSP70 mRNA。

Abstract: The present study was performed to investigate the effects of glutamine on the growth, proliferation and expression of HSP70 gene in bovine mammary epithelial cells. Bovine mammary epithelial cells in logarithmic growth phase were used for test. The cells were cultured in DMEM with different concentrations (0, 05, 10, 20, 40, 80 and 160 mmol·L-1) of glutamine for 24 h, then cells and supernatant were collected. The cells were cultured in DMEM with 8 mmol·L-1 glutamine at 37℃ for 0, 3, 6, 12, 24, 48, 72 h,respectively, then cells and supernatant were collected. The survival rate of cells was detected by MTT assay, LDH activity was detected by colorimetric assay, and the expression levels of HSF\|1 and HSP70 were tested by SYBR GreenⅠFQ\|PCR. The results showed that glutamine promoted the growth and proliferation of bovine mammary epithelial cells. The survival rate of cells was the highest at the glutamine concentration of 20 mmol·L-1, and it maintained higher level at the glutamine concentration of 40-80 mmol·L-1. The expression trend of HSP70 was the same with HSF\|1 and both reached the peak with the glutamine concentration of 80 mmol·L-1 at 24 h. Compared with the optimal dosage of 20 mmol·L-1, it was 849 (P<001) and 590 times (P<001). These results suggested that glutamine could improve the growth and proliferation of bovine mammary epithelial cells in vitro, and it could up\|regulate the expression of HSP70 through promoting the transcriptional activity of HSF\|1 mRNA.
