高产色素红曲霉菌株的筛选、鉴定和固体发酵条件优化

摘要: 从全国各地民间收集的红曲米中分离筛选到1株高产红曲色素的红曲霉Mp\|41菌株,依据形态和ITS序列鉴定该菌株为紫色红曲霉(Monascus purpureus)。对Mp\|41菌株的固体发酵基质和发酵条件进行响应面优化。结果表明,Mp\|41菌株产生红曲色素的适宜条件为:以籼米为固体发酵基质,初始含水量4481%,初始pH 60,培养温度32℃,接种量为265 mL·30g-1,发酵时间为1019 d。在此条件下,Mp\|41菌株发酵生产红曲米色价最高,可达5 3404 U·g-1,为市售红曲米色价的2~3倍。

Abstract:  One Monascus strain Mp\|41 with high pigment production ability was isolated from red kojic rice collected all over China. According to its morphology and ITS sequences, the strain Mp\|41 was identified as Monascus purpureu. The solid fermentation conditions of Mp\|41 were optimized by response surface method. It was shown that the optimal fermentation conditions were as follows: long rice as substrate, initial moisture content of 4481%, initial pH of 60, culture temperature of 32℃, inoculation amount of 265 mL·30g-1, and fermentation time of 1019 d. Under this condition, the color value of red kojic rice after fermentation was the highest (5 3404 U·g-1), which was 2-3 times as much as those sold in the market.
