Combining response surface analysis and principal component analysis to optimize priming condition in tobacco seeds

Abstract: Response surface analysis (RSA) and principal component analysis (PCA) were used to optimize the combination of priming time, priming temperature and the pH values of priming solution in tobacco seeds of cv. Honghua Dajinyuan and Yunyan 97. According to the RSA design, the tobacco seeds were treated by 20 combinations. After that, the germination test of treated seeds was conducted; seedling height, dry weight, fresh weight and vigor index were measured and analyzed by RSA and PCA. Finally, the optimal combination of priming time, priming temperature and the pH values in priming solution were obtained, priming time was 49 h and 45 h, priming temperature was 30℃ and 21℃ and pH values was 5 and 6 for Honghua Dajinyuan and for Yunyan97, respectively.
