摘要:
基于HPLC进行多成分含量测定和指纹图谱技术,结合化学模式识别法对生稻芽的质量水平进行评价,为生稻芽后续深入研究与开发提供依据。采用HPLC法建立18批不同批次生稻芽指纹图谱,通过对照品指认出生稻芽中3种主要酚酸类成分,并测定了样品中3种酚酸的含量。通过中药色谱指纹图谱相似度评价系统(2012版)和IBM SPSS Statistics 23进行相似度评价、聚类分析、主成分分析和因子载荷分析。18批生稻芽指纹图谱共标定了6个共有峰,指认的3个共有峰为香草酸、对香豆酸和阿魏酸,指纹图谱相似度范围为0.868~0.995,具有较好的一致性。样品中香草酸、对香豆酸和阿魏酸含量分别为16.70~54.10 μg·g-1、27.00~49.54 μg·g-1、23.09~50.81 μg·g-1。聚类分析将样品分为3类,18批样品中16批聚为一类,来自浙江杭州(S14)、浙江杭州(S17)的样品分别聚为一类,主成分分析用两个主成分进行综合评价,综合得分大于1的前2位分别为S14、S17,18批样品中3种酚酸成分的总含量与主成分分析综合得分存在明显的相关性。建立的生稻芽质量评价方法操作简便、结果可靠,香草酸、对香豆酸和阿魏酸3个主要酚酸可以作为生稻芽质量评价的指标性成分。
