‘桐乡槜李’及4个授粉品种的S基因型分析

The S-genotype analysis of Tongxiang Zui Li (Prunus salicina Lindl) and 4 pollination varieties

摘要: 利用李属S基因特异性引物, 对‘桐乡槜李’及目前桐乡生产中应用的4个授粉品种进行PCR扩增, 在‘槜李’中获得500 bp与300 bp左右两个目的条带,授粉品种中只扩增出300 bp左右的一个条带。目的条带的序列比对结果表明,500 bp与300 bp左右条带的DNA序列分别与S-h和S-7达到100%的同源性。田间授粉试验表明,4个授粉品种分别与‘槜李’杂交及‘槜李’自交的坐果率无显著差异。

Abstract: PCR was conducted with Prunus S-RNase-gene-specific primers using genome templates of Tongxiang Zui Li and 4 pollination varieties. Two target bands (about 300 bp and 500 bp, respectively) were amplified in Zui Li, only one band (about 300 bp) was amplified in each of the 4 varieties. These fragments were cloned and sequenced and two types of S-RNase-genes were identified. The longer band in Zui Li was corresponding to S-h, and the other one was corresponding to S-7. The bands in other varieties were all corresponding to S-7. There was no significant difference in the fruit setting percentage between Zui Li self\|pollination and hybridizations with related pollination varieties.
