单叶铁线莲Clematis henryi愈伤组织诱导与植株再生

摘要: 以单叶铁线莲老枝、嫩茎、叶片和叶柄为外植体,在附加各种激素的MS培养基上进行诱导培养,以探讨愈伤组织诱导和植株再生的条件。结果表明,嫩茎为最佳外植体,适用于愈伤组织的诱导,在MS+1.2 mg·L -1 6-BA+0.35 mg·L -1 NAA培养基中,嫩茎的出愈率为88.6%;MS+4.0 mg·L -1 KT+0.4 mg·L -1 NAA为最佳不定芽诱导培养基,不定芽发生率为90.0%;MS+0.04 mg·L -1 NAA为最佳生根培养基,生根率达95.0%。

Abstract: To develop a procedure for callus induction and plant regeneration of Clematis henryi, experiments were performed on Murashige and Skoog medium supplemented with different hormones using old vines, tender stems, leaves and leafstalks as explants. The results showed that tender stems were the best explants for differentiating callus, and gave the highest induction rate of 88.6% on MS+1.2 mg·L -1 6-BA+0.35 mg·L -1 NAA. MS+4.0 mg·L -1 KT+0.4 mg·L -1 NAA, which was proved to be the best adventitious shoots formation medium with a induction rate of 90.0%. The best rooting medium was obtained while Clematis henryi was cultured on MS+0.04 mg·L -1 NAA, resulting in a rooting percentage of 95.0%.
