免疫胶体金法快速检测水产品中硝基呋喃类代谢物的研究

Rapid determination of nitrofuran metabolites in aquatic products by colloidal gold method

摘要: 对水产品中硝基呋喃类代谢物(AOZ, SEM, AMOZ, AHD)快速衍生化方法的研究发现:理想的衍生化条件是在酸性条件下,加入4\|羰基苯甲醛(4\|CBA)或2\|硝基苯甲醛(2\|NBA)经60℃恒温水浴60 min;进一步对样品的洗涤、提取和净化方法进行了优化,在此基础上建立了水产品中硝基呋喃类代谢物免疫胶体金快速检测方法,研制出免疫胶体金快速检测试剂盒,对AOZ 最低检出限为05 μg·kg-1, 对SEM, AMOZ 和AHD最低检出限均为1. 0 μg·kg-1,最短检测时间约为90 min·个-1样品。胶体金法与液相—串联质谱法的检测结果进行对比,两者的符合率约为98%。胶体金法快速、简便、成本低,有很好的应用价值。

Abstract: The fast methods of the derivatization of nitrofuran metabolites(AOZ, SEM, AMOZ, AHD) in aquatic products were investigated. The results showed that the optimal derivation condition was adding 4\|carbonyl\|benzaldehyde (4\|CBA) or 2\|nitrobenzaldehyde (2\|NBA) solution, then insulating in 60℃ constant temperature water bath for 60 min in acid medium. By comparing and selecting the clean\|up, extraction, and purification methods of the samples,the colloidal gold rapid test methods for the determination of nitrofuran metabolites in aquatic products had been developed and the colloidal gold rapid test kit had been successfully produced. Their lowest detection limits for AOZ, SEM, AMOZ and AHD were 05, 1. 0, 10 and 10 μg·kg-1 respectively, and the shortest test time for one sample was 90 min. These test results were confirmed by the methods of liquid chromatography\|tandem mass spectrometry and the coincidence rate of two methods was 98%. The colloidal gold rapid test methods were fast, easy to handle, inexpensive and of great application value for rapid detection.
