杏鲍菇的菌株筛选与原生质体制备条件优化

摘要: 杏鲍菇是一种极具开发潜力的优质食用菌。通过筛选获得一株生长较快的杏鲍菇菌株,经条件优化,获得了最佳的原生质体制备条件。制备原生质体的最佳条件为:pH 55、酶解时间3 h、酶解温度37℃、6 mol·L-1甘露醇为稳渗剂、3%蜗牛酶和1%纤维素酶、菌龄3 d。在最优条件下,原生质体产量达到67×104个·mL-1。研究结果为进一步开展杏鲍菇菌株的紫外诱变和原生质体融合选育工作奠定了重要的技术基础,可为杏鲍菇高产菌株选育和应用提供参考依据。

Abstract: Pleurotus eryngii is a promising edible fungus. In this study, a Pleurotus eryngii strain was screened for rapid growth. The best protoplast preparation conditions were obtained via optimization. The optimum conditions for the preparation of protoplasts are: snailase of 3% and cellulase of 1% pH 55, enzymatic hydrolysis temperature of 37℃, cell age of 3 days, and mannitol of 6 mol·L-1 as osmotic stabilizer. The results presented here lay an important foundation of UV mutation and fusion protoplast of Pleurotus eryngii strains, which has importantly practical values for the breeding of Pleurotus eryngii strains.
