固相萃取—高效液相色谱—荧光检测法测定泥蚶中的苯并[a]芘含量

摘要: 采用固相萃取—高效液相色谱—荧光(HPLC\|FLD)联合测定滩涂贝类泥蚶中苯并[a]芘的含量。将泥蚶样品用正己烷超声振荡提取,用苯并[a]芘(B[a]P)专用固相萃取柱(SPE)净化,荧光检测器定量检测。苯并[a]芘在001~1 μg·mL-1浓度范围内线性相关系数达0999 2,本方法平均回收率为8147%~8471%,相对标准偏差(RSD)为48%~88%(n=6)。在优化的试验条件下,该方法苯并[a]芘检出限为03 μg·kg-1;荧光检测器的检出限为10 ng·mL-1。总体来看,用该方法检测泥蚶中苯并[a]芘含量前处理简单、分析时间短、具有良好的灵敏度和准确性。

Abstract: A method of solid phase extraction\|high performance liquid chromatography with fluorescent detector (HPLC\|FLD) was developed for the determination of benzo[a]pyrene(B[a]P) in Tegillarca granosa. The analytical procedure included extraction using n\|hexane by ultra\|sonic oscillation, purification using a B[a]P special SPE column and detection by a fluorescent detector. The method exhibited a good linearity over the range of 001-1 μg·mL-1 for benzopyrene with r=0999 2. The average recovery rate was 8147%~8471% and the relative standard deviation was 48%~88% (n=6). Under optimized conditions, the detection limit of this method was 03 μg·kg-1, and the detection limits of the detector was 10 ng·mL-1. In conclusion, the proposed method exhibited advantages of easy to operate, fast to perform, and high sensitivity and accuracy.
