摘要/Abstract

摘要： 为了探讨兔出血症病毒(RHDV)衣壳蛋白的细胞内定位,利用杆状病毒表达系统构建了表达RHDV结构蛋白(VP60)的重组杆状病毒。间接免疫荧光(IFA)和Western blot检测以及电子显微镜观察结果表明,VP60不仅能在昆虫细胞中正确表达,而且能自组装形成病毒样颗粒(VLPs)。进一步对表达蛋白进行细胞内定位观察发现,表达蛋白定位于Sf9细胞的细胞膜上。

Abstract:

A recombinant baculovirus expressing VP60 of Rabbit hemorrhagic disease virus (RHDV) was constructed with bac\|to\|bac baculovirus expression system to study the expression and intracellular localization of the structural protein VP60 of RHDV in Sf9 cells. The results of indirect immnuofluorescence assay (IFA) and Western blot analysis showed that the proteins were successfully expressed in Sf9 cells. The results of electron micrographs showed that there were some virus\|like particles (VLPs) formed in Sf9 cells, and further results of confocal microscopy indicated that VP60 proteins were localized at the cellular membrane of Sf9 cells．
