Callus induction and plant regeneration of Clematis henryi

Abstract: To develop a procedure for callus induction and plant regeneration of Clematis henryi, experiments were performed on Murashige and Skoog medium supplemented with different hormones using old vines, tender stems, leaves and leafstalks as explants. The results showed that tender stems were the best explants for differentiating callus, and gave the highest induction rate of 88.6% on MS+1.2 mg·L -1 6-BA+0.35 mg·L -1 NAA. MS+4.0 mg·L -1 KT+0.4 mg·L -1 NAA, which was proved to be the best adventitious shoots formation medium with a induction rate of 90.0%. The best rooting medium was obtained while Clematis henryi was cultured on MS+0.04 mg·L -1 NAA, resulting in a rooting percentage of 95.0%.
