Isolation and identification of halophilic and halotolerant fungi from roots of Suaeda plants

Abstract: In the present study, the traditional tissue-isolation method was used to isolate fungi from suaeda roots collected from Inner Mongolian for investigation of fungi composition structure and their salinity tolerance. Salinity tolerance of fungi were evaluated by fungal incubating method on PDA media with different concentrations of NaCl. The halophilic and halotolerant fungi were screened out and identified via both morphological characters and ITS rDNA sequencing. A total of 40 halotolerant fungi strains were isolated in this study, and were preliminary classified as Fusarium sp., Alternaria sp., Aspergillus sp., Pythium sp., etc. Among these fungi, Aspergillus aculeatus, Fusarium oxysporum were newly recorded halotolerant fungi. Three halophilic fungi were isolated and all were classified as Aspergillus sydowii. Results of the present research indicated that thediversity of salinity-tolerance fungi was high in the halotolerant plant rhizosphere.
