PCR-DGGE技术分析不同地区野生锯缘青蟹肠道菌群多样性

Analysis of the intestinal bacterial communities in wild Scylla serrata from different districts by PCR-DGGE

摘要: 采用PCR-DGGE技术对7个地区的锯缘青蟹肠道菌群进行了群落结构分析和比较。结果显示7个地区的锯缘青蟹肠道菌群的结构多样性存在较大差异,相似性指数在30%~70%之间,且同一地区雌雄样本的肠道菌群相似性也存在差异。对DGGE图谱中的12条主条带进行回收、测序,结果经Blast比对,发现12个条带代表的细菌分别属于3个细菌类群:变形细菌(Proteobacterium)、厚壁菌(Firmicutes)和棍状厌氧菌(Anaerorhabdus),其中有7个条带代表的细菌为不可培养的种类。

Abstract: PCR-DGGE of 16S rDNA was used to analyze and compare intestinal microbial communities in wild Scylla serrata from seven different districts. The total DNA of the intestinal bacteria in wild S. serrata was exacted and V3 region of 16S rDNA was amplified with the BV3-F and BV3-R primers. Then, the technology of denaturing gradient gel electrophoresis (DGGE) was used to analyze the amplified fragments. The results showed that there was a high diversity of bacterial community compositions among different samples and the similarity indices were only from 30% to 70%, and even the female and male samples from the same district also showed different diversity and similarity indices. Twelve main bands were discovered in DGGE fingerprint and re\|amplified, sequenced and aligned with Blast. The result showed that the bacteria belonged to three major groups: Proteobacteria, Firmicutes and Anaerorhabdus, and seven of the fragments represented the uncultivated bacteria.
