Rapid culture method for barley anther and effect of genotype difference in anther culture response

Abstract: Barley anther culture is one of the technologies for haploid breeding, and is also the foundation for studying barley gene transformation. In this paper, barley anthers were pretreated by mannitol and then were cultured in the induction medium and regeneration medium, and a lot of regenerated plants were formed. Using this method, the effect of anther culture with pretreatment for 4 d and 5 d on 10 barley hybrid combinations was compared, the results showed there was no significant difference in pretreatment 4 d and 5 d, the high frequency of green plants yield was all obtained. We also compared anther culture response of 25 barley hybrid combinations using this method, and we found there was significant difference in different hybrid combinations, the frequency of responding anther and the frequency of regenerated green plants were highly correlated with the frequency of green plants yield.
