Preparation and identification of monoclonal antibodies against the nucleocapsid protein of porcine epidemic diarrhea virus

Abstract: In order to prepare the monoclonal antibodies against porcine epidemic diarrhea virus (PEDV) N protein, BALB/c mice were immunized with purified recombinant N protein emulsified in Freunds adjuvant. The monoclonal antibodies (Mabs) were prepared by fusing mouse myloma cells(SP2/0) with the spleen cells from BALB/c mice immunized with the purified recombinant N protein. Three hybridoma cell lines secreting Mabs were screened by indirect enzyme\|linked immunosorbent assay (ELISA) and named as 5B2, 3H1 and 4H5, respectively. The results of Western blot and immunofluorescence assay analyses showed that the three Mabs could react specifically with the recombinant N protein or PEDV. The isotypes of three Mabs belong to IgGl subtype with Kappa chain. It was indicated that these Mabs may be useful for developing the diagnosis methods and investigating the pathogenicity of PEDV.
