摘要:
为研究血耳(Tremella sanguinea Y.B. Peng)及其宿主菌的分类地位,利用担孢子弹射法和耳木组织培养法从椴木栽培的血耳中分离得到1株酵母状菌株XE01和1株丝状真菌菌株XE02,分别对这两株菌株进行了菌落形态和显微形态观察及基于rDNA-ITS序列的系统发育分析。结果显示,XE01在PDA培养基上形成湿润、光滑的白色菌落;细胞为椭圆形,可见芽体;其ITS序列与褐银耳属(Phaeotremella)聚为一支,与P. neofoliacea 和P. eugeniae的遗传距离最小,亲缘关系最近,初步推测为血耳的酵母态型。菌株XE02在PDA培养基上形成白色绒毛状菌落;菌丝分枝少,有隔,可见单生的球状孢子和锁状联合;培养基中可见红褐色分泌物;其ITS序列与韧革菌属(Stereum)聚为一支,与皱韧革菌(S. rugosum)的遗传距离最小,亲缘关系最近,初步推测为血耳的宿主菌。研究为厘清血耳及其宿主菌的分类地位及种质资源鉴定提供了分子依据。
