不同羽色斑嘴野鸭毛囊中Tyr,Tyrp1及C\|kit基因的表达及调控分析

Expression and regulation of Tyr, Tyrp1 and C\|kit gene in feather bulbs of spot\|billed ducks

摘要: 斑嘴野鸭羽毛呈褐色,少数个体会呈现出黄白羽及白羽表型。Tyrosinase(Tyr)、Tyrosinase\|related protein\|1(Tyrp1)以及C\|kit(The tyrosine kinase receptor)基因与黑色素合成密切相关,研究利用Real\|time PCR对这3个基因在褐羽、黄白羽以及白羽3种表型斑嘴野鸭毛囊组织中进行表达分析;同时利用在线软件分别预测调控这3个基因的microRNA。结果发现,Tyr,Tyrp1在褐羽斑嘴野鸭中表达量最高,黄白羽中表达量显著下降(P<001),而在白羽中则完全不表达。C\|kit在褐羽及黄白羽中表达量类似,而在白羽中表达则显著下降(P<001)。同时,预测到调控Tyr,Tyrp1和C\|kit基因的microRNA数分别为9,25和18个。结果表明,毛囊组织中Tyr,Tyrp1以及C\|kit基因的表达下调可能导致了斑嘴野鸭出现了黄白羽及白羽两种表型,这些候选microRNA可能参与了这3个基因的表达调控。

Abstract: The feathers of wild spot\|billed ducks are brown, but a few individuals with yellow\|white or white plumage were found when they are domesticated. Tyrosinase (Tyr), Tyrosinase\|related protein\|1 (Tyrp1) and The tyrosine kinase receptor (C\|kit) are genes associated with melanogenesis. In this study, the relative expressions of these genes were analyzed in the three types of feather bulbs using Real\|time PCR, and microRNAs were predicted for these genes. The results showed that Tyr and Tyrp1 were strongly expressed in brown feather bulbs, weakly expressed in yellow\|white feather bulbs, and not expressed in white feather bulbs. C\|kit showed same expression level in brown feather bulbs and yellow\|white bulbs, but not expressed in white feather bulbs. In addition, 9, 25 and 18 microRNAs were predicted to regulate the expression of Tyr, Tyrp1 and C\|kit, respectively. These results suggested that the down regulation of Tyr, Tyrp1 and C\|kit might induce yellow\|white or white plumage of spot\|billed ducks.
