Analysis of differential expression of TLR4-MyD88 independent pathway genes under resistin-induced bovine alveolar macrophages

Abstract: In order to explore the mechanism of bovine resistin through TLR4-MyD88 independent signal pathway in bovine alveolar macrophages, bovine alveolar macrophages were exposed to resistin(100 ng·mL-1) for 0, 1.5, 3.0, 6.0, 12.0, 24.0 h, respectively. The relative mRNA expression of TLR4-MyD88 independent signal pathway genes (TLR4, TRAM, NF-κB) were detected by qRT-PCR, the expression of proinflammatory cytokine IL-6, 1L-1β and TNF-α were tested by ELISA methods. The results showed that resistin(100 ng·mL-1) sharply upregulated the expression of TLR4-MyD88 independent signal pathway genes at 6.0 h(P<0.01). The expression of related genes reached the highest at 12.0 h. The expression of proinflammatory cytokines IL-6, IL-1β and TNF-α significantly increased at 1.5 h (P<0.01). Through the signal transmission, a large number of proinflammatory cytokines were released, causing pulmonary inflammation.
