    2019, 31(3):  345-354.  DOI: 10.3969/j.issn.1004-1524.2019.03.01
    Abstract ( 1035 )   HTML ( 129 )   PDF (1971KB) ( 1758 )
Endophytic bacteria DZSY21 can stably colonize in maize and enhance the maize resistance to disease. Therefore, this study predicted the molecular mechanism of maize disease resistance induced by endophytic bacteria DZSY21 at the genetic level. The transcriptome sequencing technology was used to analyze the differential expression pattern of total genes in different periods of maize leaves inoculated with DZSY21. Cufflinks software was used to process the data, and the fold change≥2 and FDR<0.01 were set as standards to select DEGs. The results showed that there were 2 413 DEGs after inoculated with DZSY21 at 12 h compared with the control group inoculated with DZSY21 at 0 h, including 1 278 up-regulated DEGs and 1 135 down-regulated DEGs. There were 737 DEGs after inoculation at 24 h, including 538 up-regulated DEGs and 199 down-regulated DEGs. According to the results of functional annotation, 218 DEGs were obtained involved in 26 resistance pathways after inoculation at 12 h, including bifunctional inhibitor/lipid-transfer protein, MATE efflux family protein and LysM domain receptor-like kinase. There were 71 DEGs involved in 30 resistance pathways after inoculation at 24 h, including isoflavone reductase, probable cellulose synthase, phenylalanine ammonia-lyase, L-ascorbate peroxidase, probable transcription factor GLK and ACC oxidase. There were 23 DEGs duplicated in different treatment periods. The above results showed that the introduction of endophytic bacteria DZSY21 into maize could regulate the expression of resistance-related genes in maize leaves. This work provided a theoretical basis for further searching for resistance genes and their function identification.

YE Zipiao, YIN Jianhua, CHEN Xianmao, AN Ting, DUAN Shihua
    2019, 31(3):  355-364.  DOI: 10.3969/j.issn.1004-1524.2019.03.02
    Abstract ( 972 )   HTML ( 0 )   PDF (1454KB) ( 1988 )
In order to investigate the photosynthetic characteristics of flag leaves of hybrid rice in dough stage, the gas exchange data of four hybrid rice cultivars (Shen95you1326, Wufengyou1326, Ganxiangyou1326 and restorer line JR1326) were performed by LI-6400 portable photosynthesis measurement system. The results showed that the chlorophyll content of Wufengyou1326 was 33.41% and 16.83% higher than that of Shen95you1326 and Ganxiangyou1326. There was no difference between the maximum electron transport rate of Wufengyou1326 and Shen95you1326 or the maximum net photosynthetic rate of Wufengyou1326 and Shen95you1326 (P>0.05). However, there was a significant difference between the two rice cultivars and Ganxiangyou1326 in the maximum electron transport rate and the maximum net photosynthetic rate (P<0.05). The minimum average life-time of the Wufengyou1326 of pigment molecules in the excited state was 14.89% longer than Shen95you1326 and Ganxiangyou1326, and the intrinsic cross-section of pigment molecules of Shen95you1326 was 28.71% higher than Wufengyou 1326, and the instantaneous water-use efficiency of Shenyou1326 was 34.99% and 16.71% more than that of Wufengyou1326 and Ganxiangyou1326. The results revealed that photosynthetic capacity of rice cultivars was not only related to chlorophyll content, but also to intrinsic absorption cross-section and minimum average life time in the lowest excited state. The interaction of these factors determined the maximum electron transport rate and the maximum net photosynthetic rate of rice. Therefore, the selected rice cultivars should have some special characteristics such as higher intrinsic absorption cross-section, shorter minimum average life time in the lowest excitation state and higher chlorophyll content.

    2019, 31(3):  372-378.  DOI: 10.3969/j.issn.1004-1524.2019.03.04
    Abstract ( 477 )   HTML ( 0 )   PDF (1115KB) ( 1516 )
Litopenaeus vannamei was adopted as the research object in this study, which was treated with different doses of T-2 toxin. T-2 toxin dose was adopted as the independent variable, along with morphological indicators of shrimp as the dependent variable. To investigate the correlation within the dose of T-2 toxin and various morphological indexes of treated shrimp, common statistical analysis methods were used including gray correlation analysis method, path analysis and regression analysis. The most convenient and simple biological statistics were filtered out. The experimental results indicated that the body weight, body length, carapace length and the first abdominal segment width of toxic shrimp morphological index were the most influential indexes of T-2 toxin as analyzed by gray correlation and path analysis. The significance test performed by regression analysis showed that the effects of T-2 toxin on body weight, body length, carapace length and the first abdominal segment width of treated shrimp were significant (P<0.05). The results of the three statistical methods were consistent. Body weight, body length, carapace length and the first abdominal segment width were the important morphological indicators of toxic shrimp, and they were negatively correlated with the exposure dose of T-2 toxin. And gray correlation analysis was the most convenient and easiest biological statistics.

    2019, 31(3):  379-383.  DOI: 10.3969/j.issn.1004-1524.2019.03.05
    Abstract ( 897 )   HTML ( 0 )   PDF (1425KB) ( 1750 )
In order to explore the mechanism of bovine resistin through TLR4-MyD88 independent signal pathway in bovine alveolar macrophages, bovine alveolar macrophages were exposed to resistin(100 ng·mL-1) for 0, 1.5, 3.0, 6.0, 12.0, 24.0 h, respectively. The relative mRNA expression of TLR4-MyD88 independent signal pathway genes (TLR4, TRAM, NF-κB) were detected by qRT-PCR, the expression of proinflammatory cytokine IL-6, 1L-1β and TNF-α were tested by ELISA methods. The results showed that resistin(100 ng·mL-1) sharply upregulated the expression of TLR4-MyD88 independent signal pathway genes at 6.0 h(P<0.01). The expression of related genes reached the highest at 12.0 h. The expression of proinflammatory cytokines IL-6, IL-1β and TNF-α significantly increased at 1.5 h (P<0.01). Through the signal transmission, a large number of proinflammatory cytokines were released, causing pulmonary inflammation.

    2019, 31(3):  401-407.  DOI: 10.3969/j.issn.1004-1524.2019.03.08
    Abstract ( 771 )   HTML ( 2 )   PDF (1807KB) ( 1994 )
Stilbenes, such as resveratrol, are postulated to be involved in the health benefits for human and associated with stresses resistance in plant. Stilbene synthase (STS) is key enzyme and responsible for resveratrol biosynthesis. To explore the expression characteristics of STS gene family in grape, candidate STS genes were screened by bioinformatics, expression patterns of STS genes were detected during the development of fruits and leaves. The results showed that there were 48 STS genes identified in whole grape genomics and showed cluster distribution on chromosomes 10 and 16. Thirty-two out of these 48 STS genes could code intact amino acid sequence for STS proteins with 392 aa. These 32 potentially functional STS genes were divided into 3 groups base on the similarity analysis. Although diverse expression patterns of selected STS genes were displayed during the development of berries, a majority of them indicated higher expression level during 20-40 and 70-80 days after full bloom. Most of STS genes had higher expression level in mature leaves or senescent leaves than that of in young leaves. These results provided a basis for functional study of STS gene family in grape during the development of berries and leaves.

    2019, 31(3):  420-427.  DOI: 10.3969/j.issn.1004-1524.2019.03.11
    Abstract ( 833 )   HTML ( 13 )   PDF (1465KB) ( 1999 )
Wild eggplant Solanum torvum is very popular as an eggplant rootstock in graft seedling production. There are several issues of Solanum torvum in the current seed market, such as too many supply channels and uneven seed quality. In the present experiment, ten sources of Solanum torvum were used as materials to study the effect of GA3 concentration and GA3 soaking duration on seed germination potential, seed germination rate (GR) and seed germination index (GI). It was found that seed germination potential and seed germination rate were significantly different among these ten Solanum torvum seeds without GA3 treatment, and there were only two sources of Solanum torvum seeds whose germination rate were more than 90%. The source of Solanum torvum and GA3 concentration had significant effects on seed germination rate and seed germination index, and GA3 soaking duration had significant effect on seed germination index. There were significant effects of interaction on seed germination rate in Solanum torvum seed source×GA3 concentration, GA3 concentration×GA3 soaking duration, and Solanum torvum seed source×GA3 concentration×GA3 soaking duration. Moreover, there were significant interactive effects on GI in the source of Solanum torvum seed×GA3 concentration, the source of Solanum torvum seed×GA3 soaking duration, GA3 concentration×GA3 soaking duration, and Solanum torvum seed source×GA3 concentration×GA3 soaking duration. It suggested that each source of Solanum torvum demanded respective GA3 concentration and GA3 soaking duration to obtain the best germination performance. In addition, the germination rate of most Solanum torvum seed sources were lower than that mentioned on the seed packing bag.

    Abstract ( 644 )   HTML ( 3 )   PDF (1456KB) ( 1983 )
Citrus Huanglongbing (HLB) is one of the most devastating diseases to citrus production, and can spread to various citrus species. It is necessary to establish a more efficient way to detect HLB disease and then timely remove the infection source in latent period. This research was aimed to investigate the impact of bacterium on host carbohydrate metabolism and the potential of hyperspectral imaging for citrus HLB disease in latent period. The results showed that starch, sucrose, glucose and fructose were excessively accumulated in HLB infected leaves with 3.58, 2.16, 3.41 and 1.70 times, respectively as high as to those in healthy samples. In addition, an increasing reflectance of citrus leaves in near-infrared region was observed when subjected to HLB infection. Six sensitive wavelengths (1 015, 1 331, 1 065, 1 334, 1 022 and 951 nm) were selected by Random Frog, and Naïve Bayes (NB) classifier was used for differentiating asymptomatic HLB infected leaves from healthy ones with the overall classification accuracy of 97.5% and the false negative rate of 0. These results demonstrated the feasibility of near-infrared hyperspectral imaging for HLB disease detection in the asymptomatic period.

    2019, 31(3):  436-443.  DOI: 10.3969/j.issn.1004-1524.2019.03.13
    Abstract ( 627 )   HTML ( 3 )   PDF (1663KB) ( 2024 )
To verify the pathogens that lead to stem canker on blueberry, and provide a theoretical basis for field diagnosis and integrated control of the disease, the pathogens causing stem canker of blueberry were isolated and purified by tissue separation and single spore separation. Identification of pathogen were carried out using Koch's postulates, morphological characteristics and inter transcriptional spacer of ribosome DNA (rDNA-ITS). In vivo inoculation of pathogen and spot length measurement were performed to detect the resistance of five blueberry cultivars to stem canker. The test results showed that both strains JA4 and JB2 could cause stem canker on blueberries. ITS sequences analyses and colony identification revealed that strains JA4 and JB2 were Phomopsis sp. and Botryosphaeria dothidea, respectively. The inoculation results indicated that there was no significant difference in the resistance to stem canker among Brigitta, Bluecrop and Bluebelle. Both Phomopsis sp. and Botryosphaeria dothidea could cause stem canker on blueberry. Phomopsis sp. could be invaded by flower buds, and Botryosphaeria dothidea could be infected by the wound. The blueberry cultivars Brigitta, Bluecrop and Bluebelle showed higher resistance than Sharpblue, which was susceptible to infection by the pathogen and showed weaker resistance than other four cultivars.

    2019, 31(3):  450-457.  DOI: 10.3969/j.issn.1004-1524.2019.03.15
    Abstract ( 800 )   HTML ( 1 )   PDF (1603KB) ( 1722 )
To reveal the effects of different straw returning methods on cropland soil fauna community, an experiment with four straw returning methods, i.e., drop the rice straw after burning over the surface of the soil (FS), cover with chopped rice straw on top of the soil (FG), bury the chopped rice straw (TM), and no straw returning (CK), was carried out in the Puyang Town of Dujiangyan in June and September in 2016. The soil fauna collected were classified into 50 taxonomic groups, among which Nematoda was the dominant species. The soil fauna density under FG was significantly (P<0.05) increased than CK, but there was no significant difference in the quantity of soil fauna communities. Two months after straw returning, there was no significant difference in biodiversity indexes within treatments. However, 5 months after straw returning, the dominance index of FG and TM treatments was significantly (P<0.05) higher than that of CK. These results indicated that proper straw returning could improve soil fauna community.

Identification of soil hydraulic parameters based on HYDRUS-2D software and simulation of soil water movement under indirect subsurface drip irrigation
    YU Mingtao, ZHANG Kefeng
    2019, 31(3):  458-468.  DOI: 10.3969/j.issn.1004-1524.2019.03.16
    Abstract ( 707 )   HTML ( 15 )   PDF (1899KB) ( 1896 )
Indirect subsurface drip irrigation is a new drip irrigation method, which can effectively reduce surface evaporation and improve water transport efficiency in soil. Although numerical modeling of soil water movement has become an important tool for optimizing irrigation, the predictions are often not sufficiently accurate due to the uncertainty of soil hydraulic property values. In this study, based on the laboratory data from the experiments on indirect subsurface drip irrigation, it was attempted to infer the soil hydraulic parameters by using HYDRUS-2D software. Also, the difference in soil water content distribution was numerically explored for drip irrigation devices with different specifications (diameter and height). The results showed that the HYDRUS-2D software could effectively infer the soil hydraulic parameters. The simulated soil water content and wetting distance using the inferred soil hydraulic parameters agreed well with the measured values. The differences in simulated results based on the 3 and 4 inferred parameters were not significantly different. The calculated Nash-Sutcliff model performance coefficient was 0.716 and 0.714, respectively. The irrigation device with the same permeable boundary height but different diameter had little effect on soil water content distribution around the water guide device, while the device with the same diameter but different permeable boundary height had a bigger effect on water content distribution. The results obtained in this study could provide a scientific basis for the selection of water guide device specifications and the precision management of agricultural water for indirect subsurface irrigation.

In the present study, the effect of cook temperature (100, 105, 110, 115 ℃) or cook time (30, 60, 90, 120 min) on the release rate of egg products, and the effect of starch type (potato starch, sweet potato starch, corn starch) or addition amount (1%, 2%, 3%) on gel strength of salted eggs white were studied by single factor experiments. Besides, cook temperature, cook time, starch type and addition amount were selected as factors to explore processing technologies, which would improve the properties of salted egg white by orthogonal test based on sensory evaluation. It was shown that under proper cook temperature (110 ℃) and cook time (90 min), ideal release rate of egg products could be achieved. With the same addition amount, the gel strength of products by adding corn starch was the highest. But, the gel strength of products by addition 1% potato starch was comparable to that of fresh egg white, which showed no significant difference. Among all the factors, cook temperature showed the highest influence on the quality of egg product in the orthogonal test. The theoretical ideal technology was as follows: cook temperature of 115 ℃, cook time of 120 min, addition of 3% potato starch.

    2019, 31(3):  474-479.  DOI: 10.3969/j.issn.1004-1524.2019.03.18
    Abstract ( 686 )   HTML ( 0 )   PDF (1029KB) ( 1709 )
The peel and pomace of Gannan navel orange were fermented to produce dietary fiber by mixed strains combined with two-stage temperature control in order to promote the utilization efficiency of citrus peel and pomace. The fermentation mode of inoculating mixed strains in sequence combined with two-stage temperature control was adopted. Specifically, Lactobacillus plantarum was inoculated firstly in the solid medium based on navel orange peel and pomace, and was cultured at 28 ℃ for 2 days. Then, Penicillium sp. CIs14 was inoculated in the medium containing L. plantarum, and was cultured at 28 ℃ for 5 days. The culture medium after fermentation was mixed with 1.5 times volume of sterile water and was placed in the shaker with 100 r·min-1 speed at 45 ℃ for 24 h. The contents of soluble dietary fiber (SDF) and total dietary fiber (TDF) in the citrus peel and pomace after fermentation were more than 42.0% and 90.8% respectively, and SDF/TDF reached 46.2%. The contents of SDF, TDF and SDF/TDF were increased by 3.08 folds, 50.1% and 1.72 folds, respectively, compared with those in the original navel orange peel and pomace. The content of SDF and SDF/TDF were increased by 33.8% and 30.1%, respectively, compared with those in the navel orange peel and pomace fermented by Penicillium sp. CIs14 only at constant temperature (28 ℃). Therefore, the fermentation mode of inoculating mixed strains in sequence combined with two-stage temperature control could improve the content of SDF in navel orange peel and pomace effectively. In addition, this fermentation mode had good universality for other kinds of citrus peel and pomace to produce dietary fiber with high quality.

    2019, 31(3):  480-486.  DOI: 10.3969/j.issn.1004-1524.2019.03.19
    Abstract ( 660 )   HTML ( 255 )   PDF (1064KB) ( 2083 )
Mulberry fruit was extracted with deionized water using ultrasonic-assisted method and inhibition rate to alpha-amylase and alpha-glucosidase activities by the extracts was monitored. The extraction process was optimized by L9 (34) orthogonal test. The results showed that, under the optimum conditions, i.e. 1.5 h of extraction time, 1∶40 (g·mL-1) in the ratio of material to liquid, 50 ℃ in ultrasonic temperature and 350 W in ultrasonic power, the inhibition rate of the mulberry fruit extracts to alpha-amylase and alpha-glucosidase was 76.4% and 81.4%, respectively.

    2019, 31(3):  487-495.  DOI: 10.3969/j.issn.1004-1524.2019.03.20
    Abstract ( 871 )   HTML ( 0 )   PDF (2006KB) ( 2134 )
In order to solve the problem that it is difficult for the users to identify the disease accurately in the domestic hydroponic cucumber, a cucumber leaf spot recognition system based on image processing was designed. Adaptive wavelet was applied to the original image noise reduction processing. The ideal cucumber leaf segmentation image was obtained by threshold segmentation combined with morphological operation in HSV space. The disease spots of cucumber leaves were obtained by adaptive threshold segmentation, and the morphology, color and texture original characteristic parameters were extracted from spots. The sensitivity of original characteristic parameters was defined based on GA-BP network, and the optimal combination of features was realized by recursive elimination of some features with low sensitivity. According to the optimized combination of characteristic parameters, the support vector machine was used to identify the anthrax and powdery mildew of cucumber. The experimental results showed that the method effectively improved the recognition rate of cucumber diseases and provided a reference for other crop diseases intelligent identification.

Influence of confirmation of entitlement on farmers' homesteads transfer based on evidence from Wenzhou

    2019, 31(3):  503-511.  DOI: 10.3969/j.issn.1004-1524.2019.03.22
    Abstract ( 604 )   HTML ( 0 )   PDF (1061KB) ( 1554 )
The key to promoting the separation of the three powers of the homestead was to release the right to use the homestead effectively and to promote the formation of the homestead circulation market. In order to identify the transfer decision of the farmer's homestead use right, this paper chose Wenzhou as the reform pilot zone. Theoretical and empirical analysis was carried out from the perspective of homestead confirmation. The research results showed that the right of homestead had significantly promoted the transfer of the right to use the homestead. Relatively, the homestead with no power was confirmed. On average, the probability of the homestead entering the transfer market after the confirmation of power was 13% higher. The sub-sample test showed that for those sub-samples that agreed that the right of homestead qualification belonged only to the farmers, the confirmation of rights would significantly promote circulation, but for those sub-samples that did not agree that the right to homestead qualification belonged only to the farmers, the confirmation of rights did not significantly promote the role. Further analysis showed that the confirmation of homesteads would make farmers more eager for the state to relax restrictions on the transfer of homesteads. Therefore, the research in this paper clarified that the power certification was conducive to the activation of the homestead circulation market, thus promoting the realization of the “three powers separation” of the homestead.
