Synchronous determination of various aflatoxins in coarse fodder by HPLC

Abstract: The present study was conducted to establish a determination method for various aflatoxins in coarse fodder by HPLC. Aflatoxins were synchronously extracted from samples, cleaned up with immunoaffinity column, separated by elution in C18 column and analyzed by fluorescence detector after the photochemical derivation. Four kinds of aflatoxins were separated within 35 min. The recovery rate of added aflatoxin AFB1, AFB2, AFG1 and total aflatoxins from rice straw, Chinese wildrye and bamboo shell silage were all above 800%. But, the recovery rate of AFG2 was relatively low. The relative standard deviations (RSD) for aflatoxins in parallel samples were less than 100%. The limit of detection (LOD) for aflatoxin AFB1, AFB2, AFG1, AFG2 and total aflatoxins were 04,02,04, 02 and 12 μg·kg-1, respectively. Thus, the established determination method could realize simultaneous extraction, clean up, separation, and detection of aflatoxins in coarse fodder. The method is accurate, simple, high sensitive and well repeatable, which is suitable for the rapid determination of aflatoxin in coarse fodder.
