Effect of sertoli cells apoptosis in piglets induced by single or combined administration of zearalenone and deoxynivalenol

Abstract: The aim of this study was to investigate the mechanism of apoptosis of sertoli cells in piglets induced by single or combined administration of zearalenone and deoxynivalenol. Sertoli cells were treated with 10 μg·mL-1 ZEA and 0.2 μg·mL-1 DON for 24 h. Cell viability was detected by CCK-8 method, ultrastructural changes were observed by electron microscope, apoptosis rate and mitochondrial membrane potential were detected by flow cytometry and immunofluorescence. The expression of apoptosis related genes was detected by fluorescence quantitative PCR. The results showed that single or combined administration of ZEA and DON could decrease the viability of sertoli cells, damage the ultrastructure of sertoli cells and increase the apoptosis rate. In addition, ZEA and DON could down regulate the mRNA expression of Bcl-2, Bax, Bid, Caspase-3 and Caspase-9. Moreover, the combined effect of ZEA and DON was stronger than single effect. These results showed that ZEA and DON could induce apoptosis of sertoli cells of piglets through mitochondrial pathway, and the combined toxicity was greater than that of single toxicity, showing additive effect.
