Cloning and analysis of full length sequence of STAT1 gene in Chinese Holstein dairy cow

Abstract: The full length sequence of STAT1(signal transducers and activators of transcription 1) gene in Chinese Holstein dairy cow was cloned by SMART technology. The primers used for RACE were designed from the sequence of mammalian orthologs of STAT1 genes deposited in GenBank. Sequence analysis revealed that STAT1 gene was selectively spliced in the 5' and 3' untranslated regions. The Chinese Holstein dairy cow STAT1 gene contains the 2250 bp open reading frame, encoding a putative protein with 749 amino acids. Phylogenetic tree analysis showed that the protein exhibits a high homology with that from sheep (99.6%), rhesus monkey (96.4%), human (95.87%), pongo (95.73%) and pig (95.64%). The results have laid a foundation for further analysis of structure, expression and regulation of STAT1 gene in cow.
