Callus induction, differentiation and plant regeneration of bulbs of different cultivars of Fritillaria thunbergii

Abstract: The bulbs of narrow-leaf, broad-leaf and multi-seed cultivars of Fritillaria thunbergii Miq were used to investigate the effect of different sterilized methods and culture media with different hormone combinations on the callus induction, differentiation and plantlets regeneration. The results showed that the optimum sterilized method was treated with 0.1% mercuric chloride for 20 min; the callus induction rate was the highest on the medium with MS+NAA 1.5 mg ·L-1+KT 0.5 mg ·L-1 , and the induction rate was 36.7%; the callus differentiation rate was the highest on the medium with MS+NAA 4.0 mg ·L-1+KT 1.0 mg ·L-1 , and the differentiation rate was 80.7%. The results also demonstrated that the genotype had significant effects on callus induction, differentiation and plant regeneration of different cultivars of Fritillaria thunbergii.
