Tissue culture and rapid propagation technology of Dioscorea alata

Abstract: With the stem segments as explants, the tissue culture and rapid propagation of purple Dioscorea alata from Taizhou, Zhejiang Province were investigated. The results showed that the sterilization efficacy of 70% alcohol for 30 s and 84 disinfector for 15-20 min was the best. The optimal axillary buds inducing medium was MS+05 mg·L-1 6BA+01 mg·L-1 NAA, in which the buds number was the largest and the height was maximum. After breeding for 25 d,the average buds number was 158 and the height was 23 cm. The optimal rooting medium was 1/2MS+01 mg·L-1 6BA+20 mg·L-1 NAA+002% activated carbon, in which the average rooting days was 6 d, rooting rate was 100%, aerial roots rate was low and roots were thick and strong. After 30 d culture in rooting medium, the lateral branch cuttings with the axillary buds were propagated in the rooting medium, and the multiplication coefficient reached 30.
