Abstract: Effect of three kinds of extraction buffers on preparation of strawberry nuclei suspension flow cytometry of Hongjia， Chuanjiu No.1 and Jilin was investigated. The results showed that extraction bufferⅢ had clear and focused peak particle， little background debris， simple and easytoprepare formula and apparent differences distinguished among three kinds of strawberries materials， which was better than the other two kinds of extraction buffers. Comparison of testing effect on strawberry Hongjia materials with different parts or different sources was performed. Detecting effects of plantlets were superior to field testing materials. Leaves of plantlets were the best test materials due to easy material availability， clear peak， less miscellaneous peak and less cellular debris．
