A high\|throughput method for determination of apparent amylase content in rice

Abstract: A rapid and simplified method was developed to determine apparent amylose content in rice based on the standard method (NY/T 83—1988) in this study. A significant positive correlation was found between the modified method and standard method. In addition, the modified method was simple, and less sample and time were needed. The differences in I2\|KI staining could be easily distinguished by eyes (without spectrophotometer) when the difference of apparent amylose content between two samples was above 5%. This modified method could be applied in high throughput screening of the breeding populations with little amount of sample in the future.
