紫杉醇在鸡肉中的沉积及对Bcl\|2基因表达的影响

Deposition of paclitaxel in chicken and its effects on the Bcl\|2 gene expression

摘要: 试验旨在研究不同红豆杉添加水平对肉鸡鸡肉紫杉醇含量及组织Bcl\|2 mRNA表达水平的影响,以确定红豆杉在肉鸡饲粮中的适宜添加水平。试验选取日龄、体重相近(819±50)g且健康状况良好的生长肉鸡180只,随机分为3组:对照组饲喂基础饲粮,试验组在基础饲粮的基础上分别添加1%,3%红豆杉枝叶干粉。试验期为67天,结果表明:1)饲喂红豆杉前期(第1~30天),3%红豆杉极显著降低了肉鸡平均日增重及平均日采食量(P<001);但在后期(第31~60天),1%(P<005)、3%(P<001)红豆杉显著提高了肉鸡平均日增重与平均日采食量;从整个采食期来看,肉鸡平均日增重、平均日采食量随红豆杉添加剂量的增加分别呈升高、降低趋势,但影响不显著(P>005)。饲料增重比随红豆杉添加剂量的增加呈降低趋势,但在不同时期各组之间均表现差异不显著(P>005)。2)肉鸡采食红豆杉后,红豆杉中的紫杉醇可以沉积到鸡肉中,由原来的0水平上升到062 ng·g-1。3)3%红豆杉添加水平可使肉鸡肝脏Bcl\|2 mRNA表达量显著升高(P<001),肺脏Bcl\|2 mRNA表达量显著降低(P<001),对脾脏无显著影响(P>005)。由此可见,饲粮红豆杉中的紫杉醇能够沉积到鸡肉中,但沉积量很低;该试验条件下,红豆杉适宜添加水平为3%。

Abstract: To determine the optimum supplemental level of Taxus chinensis for broiler diet, this experiment was conducted to investigate the effects of paclitaxel on deposition in chicken and the Bcl\|2 gene expression. One hundred and eighty healthy broilers with similar weight (819±50) g were randomly allocated to 3 groups with 3 replicates per group and 20 chickens per replicate. Control group was fed with a basal diet, and test groups were fed with the basal diet supplemented with 1%, 3% dry powder of Taxus chinensis, respectively. The experiment lasted for 67 days. The results showed as follows: 1) The average daily gain and average daily feed intake of broilers fed diets with 3% Taxus chinensis were significantly lower than that in the control group during 1-30 day (P<001). However, Taxus chinensis supplementation significantly improved the average daily gain and average daily feed intake of broilers during 31-60 day. The 1% group (P<005) and 3% group (P<001) all had higher average daily gain and average daily feed intake. The average daily gain and average daily feed intake of broilers were not significantly different between the control group and test group in the whole experiment period (P>005). The ratio of feed to gain in all test groups were lower than that in the control group to some extent, while there were no significant differences among them (P>005). 2) The paclitaxel in Taxus chinensis could deposit in breast muscle of broilers after feeding the diet supplemented with Taxus chinensis. The contents of paclitaxel in chicken increased from 0 to 062 ng·g-1. 3) Diet supplementation with 3% Taxus chinensis significantly up\|regulated the Bcl\|2 mRNA expression in liver (P<001), but significantly down\|regulated the Bcl\|2 mRNA expression in lungs (P<001). The Bcl\|2 mRNA expression in thymus and spleen were not significantly different between diets with 0% and 3% Taxus chinensis (P>005). It is concluded that the paclitaxel in Taxus chinensis can be deposited in breast muscle of broilers after feeding the diet supplemented with Taxus chinensis, but the deposit amount is very low. The effects of 3% Taxus chinensis dry powder supplementation are better.
