Abstract: Lesion mimic mutant is a powerful tool to study the mechanisms of cell death and disease resistance. This study established a suspension-cell line of spl5， a rice lesion mimic mutant. For spl5 mutant， the optimum concentration of 2，4-D was 2 mg·L-1 in callus induction and the ideal embryogenic suspension-cell line， from osteoporosis type Ⅱ embryogenic callus， was achieved after cultured 3-4 months in suspension medium. After FDA-PI staining， the suspension-cell line of spl5 mutant showed high viability， and could be used in the further study on the mechanism of cell death and interaction with pathogen in spl5 mutant.
