Construction and analysis of a suppressive subtractive hybridisation cDNA library related to fruiting body formation of Pleurotus pulmonarius after cold stimulation

Abstract: A transient cold condition can induce the fruiting body formation of P. pulmonarius. A subtractive cDNA library using suppressive subtractive hybridisation (SSH) was constructed for a better understanding of the molecular basis of fruitingbody formation of P. pulmonarius after cold stimulation. Based on the SSH library,a total of 185 highquality ESTs were identified by upregulated expression. BLAST searching against the Basidiomycota genomes from JGI showed that 136 EST sequences were homologous to previously known genes,and there were 49 sequences with unknown function. A total of 139 ESTs were categorized into 23 functional groups. This information can help us better understand the molecular basis underlying the fruiting induced by cold stimulation in P. pulmonarius.
