Cloning and analysis of OsCSP2 gene promoter in rice (Oryza sativa L.)

Abstract: To understand functional expression profiling of cold shock protein gene OsCSP2, the promoter sequence and expression of OsCSP2 in rice were investigated. The promoter of OsCSP2, named Poscsp2, was cloned from the salt tolerant rice cultivar Shanyou 10 using PCR. An expression vector containing the Poscsp2::GUS fusion gene was constructed and introduced into rice. The expression patterns of Poscsp2 under low temperature and high salt stress were analyzed by GUS histochemical staining and qRT-PCR. The results showed that Poscsp2 was predicted to contain a large number of cis elements associated with low temperature, high salinity and other stresses. Expression levels of GUS reporter gene and OsCSP2 were significantly increased after low temperature and high salt stress treatment, indicating that Poscsp2 can improve expression levels of downstream genes under abiotic stresses.
