摘要:
为优化桑树桑黄胞外多糖液态发酵培养基配方并探究其体外抗氧化活性,利用组织分离法纯化并鉴定1株桑树桑黄菌株JM-1,通过Box-Behnken响应面法优化菌株胞外多糖液态培养基配方,并考察其体外抗氧化活性。结果表明,优化后的液体培养基配方为玉米粉5.4 g·L-1、葡萄糖17.0 g·L-1、蛋白胨5.8 g·L-1、维生素B1(VB1),10.0 mg·L-1、KH2PO4 3.0 g·L-1、MgSO4·7H2O 5.0 g·L-1,该条件下菌株JM-1产胞外多糖含量为5.918 g·L-1,比基础培养基提高了13.63%。抗氧化活性试验结果表明,在质量浓度为5 mg·mL-1时,JM-1菌株产胞外多糖的还原能力为3.187 mmol·L-1,对DPPH自由基和羟自由基的清除率分别为63.9%、54.3%。研究结果可为桑黄资源的开发利用提供科学参考。
