TCDD对建鲤肝脏的影响

摘要: 研究2,3,7,8\|四氯二苯并\|p\|二噁英(TCDD)对建鲤肝组织的影响。将建鲤随机分为5个处理组和1个空白对照组,每组30尾鱼。采用一次性腹腔注射染毒,处理组注射剂量分别为01,03,06,12,24 μg·kg-1,空白组注射同体积二甲基亚砜(DMSO)。分别于给药后24,48,72,96,120 h采集血液及肝组织,测定谷草转氨酶(GOT)、谷丙转氨酶(GPT)、乳酸脱氢酶(LDH)、谷胱甘肽过氧化物酶(GSH\|Px)、谷胱甘肽S\|转移酶(GST)活力和总抗氧化能力(T\|AOC)的变化。结果表明:TCDD染毒72 h,各染毒组损伤程度较严重,GOT,LDH,GPT活性值显著升高(P<001或P<005),GST,GSH\|Px,T\|AOC活力显著降低(P<001或P<005)。这说明TCDD急性染毒后可以造成建鲤肝组织的损伤,引起建鲤肝组织的氧化应激反应。

Abstract:  In order to evaluate the toxic effect of TCDD on fish liver, Jian carp were randomly divided into 6 groups (30 fish for each group) including a control group. The treated fish were injected with TCDD(01, 03, 06, 12, 24 μg·kg-1, respectively). The control group had no TCDD injection, but was injected the same volume of DMSO. The blood and fish liver were collected at 24, 48, 72, 96, 120 h after injection, respectively, and the expression levels of LDH, GPT, GOT in Jian carp serum and GSH\|Px, GST, T\|AOC activity in liver homogenates were determined. It was shown that, after injection with TCDD for 72 h, GOT, GPT, LDH activities were significantly increased (P<001 or P<005), while GST, GSH\|Px, T\|AOC activities were significantly decreased (P<001 or P<005), indicating that TCDD could induce serious damage on fish liver and cause oxidative stress reaction in Jian carp after acute exposure.
