Optimization of high efficient in vitro regeneration system of vegetable soybean

Abstract: To select the easily regenerating variety and set up a better regeneration system for transgenic work, effects of explant type, hormone concentration, inoculation time, inoculation method and genotype on shoot regeneration were studied based on six varieties of vegetable soybean, including Chunfengzao, Zhenong No.6, Zhenong No.8, Huning 95-1, Liaoxian No.1 and Taiwan 75. The results showed that the regeneration frequency of cotyledon\|cotyledon petiole was the highest among different explant types. The best medium of inducing adventitious buds was B5+05 mg·L-1 TDZ+005 mg·L-1 NAA+50 mg·L-1 AgNO3. The best inoculation time was 10 days after seed germination. Cotyledon-cotyledon petiole vertical insertion showed higher regeneration frequency than horizontal insertion. Regeneration frequency of Zhenong No.8 was the highest among six varieties, which reached 8089%. And the best medium of rooting was 1/2 B5+05 mg·L-1 NAA.
