    Molecular cloning and characterization of BnPPT1 gene from Brassica napus

    Phosphoenolpyruvate (PEP)/phosphate translocator 1 (PPT1) gene is best known for its role as the translocator in importing the PEP into plastids as substrates for fatty acid biosynthesis and more important for the shikimate pathway. In order to investigate the function of PPT1 from Brassica napus, we genetically isolated a full-length cDNA encoding PPT1 with total RNA of developing seeds from Brassica napus, and the corresponding genomic DNA region from DNA of young leaves with specific primers designed according to sequences of AtPPT1 and ESTs from Brassica napus by searching the BBSRC BrassicaDB. This gene, designated BnPPT1, is 2 075 bp in length of the genomic DNA with eight introns and is 1 224 bp in length of the complete coding sequences encoding 407 amino acids. Multiple sequence alignment of protein sequences showed BnPPT1 shared high homology with AtPPT1 of Arabidopsis thaliana. Furthermore,semi-quantitative RT-PCR analysis carried out with RNA isolated from tissues and various stages of developmental seeds from Brassica napus demonstrated that BnPPT1 was preferentially expressed in seeds at middle developmental stages,which indicated that BnPPT1 gene might play an important role in carbohydrate metabolism of developing seeds in Brassica napus.

    In order to explore the diversity of rice varieties, a total of 42 SSR markers were used to identify the diversity of 137 japonica rice varieties including some landraces and cultivars grown in different periods. Of the 42 SSR markers, 33 were polymorphic, and 107 alleles were present in the 33 polymorphic loci.The mean number of alleles per locus (Na) was 3.2, with a range from 2 to 7. The average genetic diversity index of Nei was 0.290, with a range from 0.162 to 0.419. The differentiation degree of indica-japonica showed obvious difference in 137 japonica varieties, and Tdj value ranged from 0.3620-0.9445. Most varieties differentiated thoroughly, and a few japonica landraces and cultivars grown in early period showed indica type. UPGMA cluster analysis showed that all varieties could be differentiated easily, and the result was consistent with classifying based on morphological index. The japonica variety groups could be further classified into several groups. SSR analysis showed the genetic diversity of varieties popularized recently was low, exploring new germplasms and expanding genetic diversity was necessary.

    Field experiment was carried out on five olive varieties (i.e. Moraiolo, Frantoio, Leccino, Pendolino, Maurino) introduced from Italy to test their adaptability and productivity on costal land in Wenzhou, Zhejiang Province. The average yield per tree of Moraiolo was 21.97 kg at the age of six to eight years, being the highest among the five varieties; the yield of Frantoio was 12.57 kg and was the second among them. The trees of Moriolo and Frantoio grew strong with good vegetative growth, early fruitage and high setting percentage compared to the other three olive varieties (Leccino, Pendolino, Maurino). Thus, the adaptability of Moraiolo and Frantoio to the tested area was better than that of the other tested varieties, which indicated that Moraiolo and Frantoio had a potential value for practical utilization. However, the economic character of Leccino, Pendolino and Maurino was not well under the tested condition.

    The genomic DNA of 24 main wheat cultivars in Huai'an Region were amplified by 86 SSR primers located on 42 chromosomes of wheat.The results showed that 26 out of 86 SSR primers could amplify stable polymorphism among 24 wheat cultivars. Genetic similarity of 24 wheat cultivars were analyzed by 124 alleles detected from 26 pairs of primers. Six out of 26 pairs of polymorphic SSR primers were selected for their clear bands and high polymorphism to construct the fingerprinting maps of the 24 wheat cultivars. All the 24 wheat cultivars could be distinguished from each other by the fingerprint map constituted by 6 pairs of SSR primers. The genetic similarity coefficients of the 24 wheat cultivars ranged from 054 to 092. Furthermore, all the cultivars were classified into 4 groups, at the level of genetic similarity coefficient 064 according to cluster analysis by an un-weighted pair-group average method with arithmetic mean, which revealed the genetic relationship of cultivars to a degree.

    The parameter of the curve fitting process was analyzed on grain filling process with different rowed barley in the field. The experimental results indicated that barley grain filling process showed S-curve of “slow-fast-slow”, and the Logistic equation was at significant level (R2>0.9906). During the grain filling period, the duration of slow increasing stage was the longest, the duration of rapid increasing stage was followed, and the duration of gradual increasing stage was the shortest, in which the average filling rate of rapid increasing stage was the highest, and the final dry matter accumulation was significantly higher than the other two stages. The average rate of grain filling (except for V1 of Zhu 3 barley variety) and the final dry matter accumulation for two-rowed barley were higher than those for six-rowed barley. The actual 1 000-grain weight of different rowed barley was lower than the theoretical 1 000-grain weight. To achieve super high yield, the two-rowed barley should have high grain number per spike and the six-rowed barley should have high 1 000-grain weight.

    The fluorescein is a common fluorescent dye. In present experiment, seeds of two tobacco cultivars, MS Yunyan 85 and Honghua Dajinyuan, were directly mixed with fluorescein powder. Then, seed germination performance, seedling growth and protective enzyme activities in tobacco seedlings were investigated. The results showed that fluorescein coating had no adverse effect on seed germination and seedling growth of tobacco. When the ratio of fluorescein powder was 100∶10, there was no significant difference in growth and physiological characteristics of two tobacco cultivars, MS Yunyan 85 and Honghua Dajinyuan, which indicated that the ratio of 100∶10 was optimal.

Based on the geographical indication system, the regional scope of GI for Longjing tea and the essential elements of its industry were defined in this paper. In addition, the competitive advantage of Longjing tea industry and some difficulties it faced were discussed, and accordingly some strategy measures for its further development were proposed as well.

    The effects of liquid medium pollen, including pure pollen of Yunyan 85, Yunyan 87 and K326 and boric acid, sucrose, calcium chloride, Put, Spd and Spm solution, on the fruit setting, seed vigor and seedling quality of MS Yunyan 85, MS Yunyan 87 and MS K326 were investigated, with the pure pollen as the control. The results showed that the effects of artificial pollination with the liquid medium pollen which contained 0.01 g calcium chloride, 100 g sucrose and 100 g pure pollen per liter was better for MS Yunyan 85, the fruit setting percentage increased by 19.98%, the 1 000-grain weight increased 2 mg, seedling dry weight increased 1.7 mg, the amount of pure pollen used decreased by 37.6%, the liquid medium pollen had no significant effect on seed number per capsule, seed weight per capsule, germination energy, germination percentage, germination index and mean germination time. For MS Yunyan 87 and MS K326, the effects of artificial pollination with liquid medium pollen containing 0.5 mmol Spd and 150 g pure pollen per liter was better, the 1 000-grain weight increased 7.4 mg and 8.4 mg respectively, dry weight of seedlings increased 0.95 mg and 0.15 mg respectively, and fruit setting percentage of MS K326 increased by 26.09%, the pure pollen amount decreased by 40.1% and 39.3%, and had no significant effect on other indexes. The results indicated that the liquid medium pollen used in tobacco seed production could save the application amount of pure pollen on the basis of ensuring the yield and quality of seed, thus reducing the seed production costs.

    The effects of potassium fertilizer application on main quality traits and starch RVA characters of sweetpotato was studied in a long\|term located fertilizer experiment base established in 1980 in Xuzhou. The results showed that potassium treatment (NPK) enlarged the differences of RVA characters and quality among different sweetpotato genotypes, improved carbohydrates and protein synthesis and accumulation, and reduced the dry matter rate of storage root. The peak viscosity (PKV)and breakdown viscosity(BDV) were significantly different (P<0.01), which proved the potassium of the genetic diversity in sweetpotato. No\|potassium treatment (NP) showed significant correlation between main quality characteristics and RVA characters, which showed the feasibility of reflecting quality characteristics of sweetpotato starch in use of RVA characters.

    We collected 100 clinical samples from suspected Porcine reproductive and respiratory syndrome virus(PRRSV) infection occurred in pig farms for virus isolation. The positive samples of PRRSV detected by RT-PCR were divided into two groups by serum and tissue samples. PRRSV were isolated and cultured with Marc-145 and PAM cell. The results showed that 29 PRRSV, including 21 serum samples (72.5%) and 8 tissue samples (27.5%), were obtained from the 45 positive samples with cell culture. Nineteen and 24 PRRSV strains were respectively isolated with Marc-145 cell and PAM cell, in which 10 PRRSV strains were isolated with both Marc-145 and PAM cell. Data showed that serum was the ideal sample of isolating PRRSV, while the virus had no significant difference in sensitivity effect on Marc-145 and PAM cells.

    Under low N and P, low N and suitable fertilizer treatments, the changes of heading dates for 148 accessions of different barley genotypes were studied. The results showed as follows: (1) There was comparatively significant difference in the heading date, which of most barley under low N and P condition was later than under suitable fertilizer condition, and difference in delay days was significantly larger than barley genotype.(2) Under low N stress, the origin and row of barley would significantly influence heading date. Under suitable fertilizer, the origin of barley would significantly influence heading date.

    Ninety crossbred weaning piglets were randomly divided into 3 groups with 30 piglets each. In the weaned pig period, growing pig period and finishing pig period, the pigs in control group were fed with weaned pig diet, growing pig diet and finishing pig diet, respectively. The pigs in trial group 1(Chinese herbal medicine group) were fed with diets added with 3 500, 3 000 and 2 500 mg·kg-1 Chinese herbal medicine additives into relevant control diets. The pigs in trial group 2 (plant extract group) were fed with diets added with 800, 600 and 500 mg·kg-1 plant extract, a foreign patent plant extract additive. It was found that the average daily gains of control group, trial group 1 and trial group 2 were (587.47±91.93), (604.78±91.06) and(601.44±72.26) g in total trial period,respectively. The feed conversion ratios of above groups were 3.42±0.11, 3.23±0.26 and 3.41±0.22,respectively. Antibody positive rates of classical swine fever (CSF) in trial group 1, trial group 2 and control group were 83.3%, 41.7% and 41.7%, respectively. Antibody positive rate of CSF in trial group 1 was significantly higher than those in trial group 2 and control group (P<0.005), in addition, the elimination rate decreased by 33.3% and 33.3%, in comparison to those of control group and trial group 2, respectively. The economic benefit of trial group 1 increased by 40.96% and 95.89% more than those of control group and trial group 2. The results suggested that the Chinese herbal medicine additives show improving effects on pig's productive performances, disease resistance and economic benefits.

    Effects of Enterococcus faecium on growth performance, immune and antioxidant function of piglets

    The experiment was conducted to study the effects of Enteroccus faecium on the growth performance, immune and antioxidant function of piglets. One hundred and twenty Duroc×Landracex×Jinhua crossbred piglets with average body weight of about 19.7 kg were randomly distributed in two treatment groups with three replicates of twenty pigs each. The control group was fed only with basal diet and the trial group was fed with basal diet supplemented with Enteroccus faecium(E. faecium 1×106 colony forming unit (CFU)·g-1 of feed). The results showed that compared with the control, supplementation of E. faecium significantly improved the growth performance of piglets, with the ADG increased by 11.46% (P<0.05) and F/G decreased by 6.20% (P<0.05). The immune function and antioxidant ability of the treated piglets were improved i.e. the concentration of albumin, the levels of IgA, IgG, the complement component 3 (C3), the complement component 4 (C4) and the lysozyme activity in serum increased by 28.06%(P<0.05), 16.48%(P<0.05), 25.43%(P<0.05), 37.84%(P<0.05), 25.00%(P<0.05) and 76.08%(P<0.05), respectively. The level of serum glutathione increased by 46.80%(P<0.01), whereas the total superoxide dismutase activity decreased by 10.38%(P<0.05). In addition, supplementation with E. faecium decreased the counts of Salmonella and Escherichia coli by 15.37% (P<0.01) and 11.79% (P<0.05) in piglets feces respectively, and increased the lactic acid bacteria (LAB) counts by 18.78% (P<0.05). This study indicated that supplementation of E. faecium could significantly improve the growth performance of piglets, and this may due to enhanced specific and non-specific immunity function, and improved intestinal flora composition.

    In this study, a pair of special primers,according to the complete genome of Duck hepatitis virus Ⅰ(DHV-Ⅰ) , was designed to amplify VP3 gene of DHV-Ⅰ. VP3 was subcloned into pET-28a(+), then the recombinant plasmids were transformed into Escherichia coli BL21 (DE3) and induced by 0.1 mmol·L-1 IPTG. The bacteria containing pET-28a(+)VP3 were collected and examined by SDS-PAGE and Western-blotting. The results showed that VP3 protein was well expressed in E. coli. The molecular weight of the recombinant protein was 27 kD. The protein could be recognized by the specific antibody against DHV-Ⅰ.

    Optimization of high efficient in vitro regeneration system of vegetable soybean

    To select the easily regenerating variety and set up a better regeneration system for transgenic work, effects of explant type, hormone concentration, inoculation time, inoculation method and genotype on shoot regeneration were studied based on six varieties of vegetable soybean, including Chunfengzao, Zhenong No.6, Zhenong No.8, Huning 95-1, Liaoxian No.1 and Taiwan 75. The results showed that the regeneration frequency of cotyledon\|cotyledon petiole was the highest among different explant types. The best medium of inducing adventitious buds was B5+05 mg·L-1 TDZ+005 mg·L-1 NAA+50 mg·L-1 AgNO3. The best inoculation time was 10 days after seed germination. Cotyledon-cotyledon petiole vertical insertion showed higher regeneration frequency than horizontal insertion. Regeneration frequency of Zhenong No.8 was the highest among six varieties, which reached 8089%. And the best medium of rooting was 1/2 B5+05 mg·L-1 NAA.

    Comparative studies on 2-DE protein patterns of shrimp meats

    Using Penaeus vannamei, Metapenaeus ensis and Macrobrachium rosenbergii as materials, the shrimp meat electrophoretic patterns with sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and two-dimensional gel electrophoresis (2-DE) which showed high resolution and good repetition was established. The comparison study was conducted to analyze the differentially expressed proteins among three breeds. The result indicated that 37 spots showed a significant difference among three shrimp species by using statistical and quantity analysis. The 2-DE maps of Penaeus vannamei, Metapenaeus ensis and Macrobrachium rosenbergii meats demonstrated 15,3,7 spots respectively as characteristic protein spots.

    To determine the effect of frequency\|vibration trap lamp (FTL) on controlling TLB, tea longhorn beetle (Aeolesthes induta Newman), a case study was carried out on the relationship between TLB larvae damage degree and the distances of tea clumps to trap lamp (about 0-120 m) and to road side (about 0-250 m) in a tea garden in Cangnan County, Zhejiang Province. The result showed that 9.0% tea clumps, about 5 m close to FTL was damaged by TLB larvae, which was significantly lower than that (17.5%) of tea clumps about 20-50 m away from trap lamp; the clumps, about 50 m near to road side, was damaged by TLB larvae more seriously than that of far away from the road, the average injured rate was more than 20.0%; the clumps about 70 m away from road side was damaged lightly, the injured rate was less than 10.0%; approximately 30% injured rate was observed in the tea clumps where about 20 m farther away from trap lamp and about 50 m nearer to road side. The effect of FTL on controlling TLB was affected by two kinds of distances, in which the distance of tea clump to road side was major impact factor. FTL could control the damage of TLB larvae down to 10.0% where the clumps were 70 m farther away from road side and 5 m nearer to FTL.

    Establishment of SRAP marker and cultivar identification in Salvia splendens

    The concentrations of primers, dNTP, DNA template, Taq DNA polymerase and annealing temperature were optimized in order to establish SRAP (sequence-related amplified polymorphism) marker system in Salvia splendens and 17 cultivars were identified using the SARP markers established. The suitable SRAP-PCR system was established for Salvia splendens. Totally 25 μL reaction mixture contained PCR buffer (containing 2.0 mmol·L-1 MgCl2), DNA template 60 ng,dNTP mixture 0.2 mmol·L-1,each primer 0.4 μmol·L-1 and Taq DNA polymerase 1 U. Gradient temperature test showed that SRAP-PCR was not sensitive to the variation of annealing temperature (44-56℃) at second stage. Based on the reaction system established, 17 varieties of Salvia splendens were analyzed using 44 SRAP primer combinations and 37 polymorphic primer combinations were screened in total. The PIC (polymorphism information content) value of these markers varied from 0.215 to 0.941, averaging 0.672. The genotypes revealed by each marker ranged from 2 to 17 with average of 6.76. The 17 varieties could be distinguished completely only using a single primer combination F1/R1 and even two varieties quite similar in morphology, Shenzhouhong and Diwang, could be discriminated by several primer combinations, suggesting that SRAP is powerful for variety identification.

    Usage of fuzzy reasoning technology in cucumber greenhouse environment decision

    Considering the influence of greenhouse environment on cultivation of cucumber, this article proposed a method of cucumber greenhouse environment decision based on fuzzy reasoning technology. Through collecting information of the main environmental factors in greenhouse such as temperature, humidity, light intensity and CO2 concentration, the system quantified their degrees of membership based on expert knowledge, calculated their conclusion value for each conclusion of knowledge in the knowledge base, and communicated with users to give the last conclusion. The system helped users to manage the cucumber greenhouse environment.

    Effect of different concentrations of ZnSO4 (0, 500, 1 000, 2 000, 4 000 μmol·L-1)on the physiological characters in three vegetables of rape, lettuce and crown daisy was studied to investigate Zn toxicity mechanism. After 5 days stress, the results showed that SOD activity of rapeseed increased with the increase of Zn concentration, but SOD activity of lettuce decreased and SOD activity of crown daisy firstly decreased and then increased. POD activity of three vegetables firstly decreased and then increased. But the MDA content showed a pattern of “increase-decrease-increase”, and the highest MDA content was observed when Zn concentration was 500 μmol·L-1.Vitamin C content of rape decreased, but vitamin C content of lettuce firstly decreased and then increased and crown daisy showed contrary trend.

    Acute toxicity and safety of four insecticides to aquatic organisms

    In order to assess the safety of pesticides used in paddy field to aquatic organisms, the acute toxicity of acetamiprid, profenofos, diazinon and malathion were studied using Scenedesmus obliquus, Daphnia magna Straus and Brachydanio rerio as test organisms by static test, static test and semi\|static test respectively. Safety evaluation had also been conducted. The results showed that acetamiprid, profenofos, diazinon and malathion had low toxicity (55.3 mg·L-1, 72h-EC50), high toxicity (0.00509 mg·L-1, 72h-EC50), high toxicity (0.249 mg·L-1, 72h-EC50), and moderate toxicity (1.69 mg·L, 72 h-EC50) to Scenedesmus obliquus, respectively. It could be seen that acetamiprid, profenofos, diazinon and malathion had low toxicity (49.2 mg·L-1, 48h-EC50), extremely high toxicity (0.00392 mg·L-1, 48h-EC50), extremely high toxicity (0.0308 mg·L-1, 48h-EC50), and extremely high toxicity (0.00165 mg·L-1, 48h-EC50) to Daphnia magna Straus, respectively. As for Brachydanio rerio, acetamiprid, profenofos, diazinon and malathion had low toxicity (49.8 mg·L-1, 96h-LC50), high toxicity (0.706 mg·L-1, 96h-LC50), modreate toxicity (5.12 mg·L-1, 96h-LC50) and low toxicity (10.9 mg a.i.·L-1, 96 h-LC50) , respectively.

    Effects of varieties and major cultivation factors, including plant density, seeding time and amount of nitrogen fertilizer, on the wheat scab in Ninghai county of Zhejiang Province during 2009 to 2010 were studied. The results showed that ten selected wheat varieties showed significant difference in resistance to wheat scab, and the variety Ning71737 had the lowest disease index and the highest resistance level. With Yangmai 12 as research material, the wheat seeded in early and middle November in 2009 were most susceptible to wheat scab, the percentage of scab spikes and disease index were significantly higher than others. And there were no marked difference among different treatments of seeding quantity and amount of nitrogen fertilizer, respectively.

    To compare the removal effects of N, P from eutrophication water by wetland plants. The uptake dynamics characteristics of NO3-N, NH4-N and P by four types of wetland (Water hyacinth, Acorus calamus, Ipomoea aquatica, Ceratophyllum demersum) were studied. The results showed that the four wetland plants showed significant difference in the water N, P uptake , and the maximum absorption rate of NH4-N showed as Water hyacinth> Ipomoea aquatica> Acorus calamus> Ceratophyllum demersum while the maximum absorption rate of NO3-N was Water hyacinth> Ipomoea aquatica> Ceratophyllum demersum> Acorus calamus. Meanwhile, the maximum absorption rate of P was Water hyacinth> Ipomoea aquatica > Acorus calamus> Ceratophyllum demersum. Furthermore, the affinity of nitrogen and phosphorus uptake rate of four wetland plants showed opposite trend with the absorption rate.

    Thirty two rice sheath blight pathogen strains (Rhizoctonia solani) were isolated from the infected rice samples from seven distinctive areas of Anhui Province, and their pathogenicity and genetic diversity were analyzed. Pathogenic differentiation analysis indicated that the pathogenicity among the strains was significantly different. The pathogenicity of the majority isolates was moderate in Anhui province. The genetic diversity of 32 Rhizoctonia solani strains was analyzed by RAPD. The results showed that the pathogenic and genetic diversity among the tested isolates were high, the polymorphic rate was 98.2%, and the genetic similarity coefficients of the isolates were 0.62-0.84. All the strains could be classified into 4 groups at the similarity coefficient of 0.653, and most of the isolates from the same area were consistent, but the pathogenicity showed no significant correlation within the RAPD groups.

    In order to promote accurate management and proper fertilization of tobacco soil, spatial analysis on tobacco soil nutrients was conducted by GIS and geostatistics in the tobacco region of Wulong. The results showed that soil fertility factors were all at moderate levels in the whole research region except available phosphorus. The semivariograms for soil organic matter, and available nitrogen, phosphorus and potassium contents were all well fitted by spherical, exponential, exponential and spherical models. Soil available nitrogen and potassium showed strong spatial correlation, while soil organic matter and available phosphorus showed moderate spatial correlation. Ordinary Kriging method was applied to interpolate soil available elements in the research region. The results showed that available phosphorus was widely deficient in the research region, and the contents of soil organic matter, available nitrogen and potassium were at moderate levels, indicating all apparent discrepancy in the research region.

    Effect of high density and high nitrogen fertilizer on resistance to powdery mildew and yield traits of south regional testing barley cultivars was studied. The main results showed as follows: disease severity and disease index of powdery mildew significantly increased at the 0.01 level under high density and high nitrogen fertilizer, while total disease incidence was not significantly affected. As far as yield traits were concerned, 1 000grain weight, panicle length, yield and grain yield per panicle significantly decreased at the 0.05 and 0.01 level; plant height significantly increased at the 0.01 level; panicle number and seed setting rate were not significantly affected. There was great difference in the powdery mildew resistance of barley cultivars under high density and high nitrogen fertilizer, and one was resistant, the other one was sensitive to powdery mildew.

    This experiment was carried out to study the effect of fertilization on yield and nutrients absorption in japonica rice variety Zhejing 22. The results showed that rational combination of chemical fertilizers and manure showed better effect on rice yield, and the efficiency of fertilizers and absorption of nutrients increased. There was a sharply increase in rice yield and economic benefits in the MNPK treatment compared to the control, the single M and combination of chemical fertilizer NK. Compared to the control, the rice yield increased by 53% in the MNPK treatment,38% in the NP treatment, 32.3% in the MNPK’, 27% in the NPK treatment, respectively. The combination of chemical fertilizer and manure could increase the fertilizer efficiency. The absorption amount of N, P, K in grain in the MNPK treatment were 126.66 kg·hm-2, 26.91 kg·hm-2 and 45.59 kg·hm-2. And the absorption amount of nitrogen, phosphorus, potassium were 184.51 kg·hm-2,33.57 kg·hm-2 and 198.40 kg·hm-2 in the MNPK’ treatment. The highest nitrogen, phosphorus harvest index was 43.85% and 36.64% in M treatment, and the highest potassium harvest index was 54.52% in NPK treatment. According to the fertilizer input, nitrogen efficiency in all nitrogen treatments was lower than M treatment, and phosphorus efficiency in phosphorus treatment was lower than M treatment. But potassium efficiency in potassium treatment was higher than M treatment.

    The productivity of transplanting seedlings could not be improved due to the low efficiency of manual picking seedlings that was approximately 45 seedlings per minute in one way working by semi-automatic transplanter. For plug seedling transplanting, the developmental trend of transplanter was automatical-picking instead of manual\|picking. Pick-up seedling device was the end-effector of the automatic picking system, the influence of its different structures and operating conditions on picking success rate were directly related to the operation quality of transplanting mechanism. A deformed sliding needle-type pick-up device was designed, and an orthogonal experiment on the designed machine was used to study the picking success rate, the influencing factors and the best working conditions. The factors selected for this experiment were the vegetable plug seedlings' own physical characteristics, seedling plug moisture content, the angle and the velocity of picking plug seedlings and the number of gripper needles. The experimental results showed that the average picking success rate was 83.61%, and through analysis of ANOVA it revealed that the number of needles and seedling age were two major factors which might affect the success rate of picking operation, the angles was next. The highest picking success rate was obtained as 25-day-old seedling, 4 gripper needles, and pick plug seedlings with 36° angles.

    Effects of four kinds of foliar fertilizers (high performance calcium, boron fertilizer, calcium nitrate and rare earth fertilizer) on fruit quality of Mandarinorange(Citrus suavissima. Hort. ex Tanaka) were studied based on plants of five years old. The results showed that the contents of total soluble solids, soluble sugar and juice yield of fruit increased, while the titratable acid decreased following all foliar fertilizer treatments. Rare earth fertilizer showed the best effect than boron fertilizer and high performance calcium. Treatment of fruit with different concentrations of high performance calcium diluted 600 times showed the highest quality.

    The growing properties of two excellent yogurt fermentation strains were studied in this paper. The results showed that the optimum carbon and nitrogen sources were lactose and peptone respectively, Zn2+ and Mg2+ could promote the growth of the strains. For adding Ca2+, only E2 could be promoted. The optimum culture conditions of E2 were as follows: temperature 41℃, the initial pH 8, culture for 3.5 h, inoculation volume 2%; the optimum culture conditions of D2 were as follows: temperature 43℃, the initial pH 7, culture for 2.5 h,inoculation volume 4%.

This platform established some functions including basic data management, statistical inquiry, thematic mapping, decision support, office online and so on, provided real-time technical support and decision support for agricultural producers and policy makers.

    Foliar spraying with organic selenium on Astragalus sinicus was carried out on the 20 d and 10 d before the period of turning green. The results showed that the selenium content in young shoot of Astragalus sinicus was significantly improved after foliar spraying, moreover, the yield of young shoot also increased. It indicated that the planting benefit of Astragalus sinicus could increase in the production of selenium-enriched young shoot. Furthermore, the optimal amount of organic selenium was 6.75-8.10 kg·hm-2.

    For the complex geomorphic condition of forest districts, the application of Geographic Information System(GIS)in forest monitoring and management system showed great conviniences to daily management such as forest fire positioning, terrain observation, distance measuament and so on. Video monitoring system was one of the most efficient and economical systems used in monitoring and preventing forest fire at home and abroad these days. Thus a GIS integrated with video monitoring system was designed, and the function was developed that 2D map, 3D scene and video monitor screen could display the same target area and could control each other. New system has been successfully applied in BaiLang-Forest of Inner-Mongolia Province. The practice shows that the system is an efficient management platform.

Industry chain is a very complicated system, and the research on the vulnerability of the industry chain is still a very new field. Thus, study on the effect of vulnerability of citrus industry chain on the development of citrus industry was quite important. This paper was based on the brittle model of complex system, and the concept and connotation of the industry chain vulnerability were the entry points. The influencing factors of the internal instability of citrus industry chain were analyzed, and its brittleness factors were refined. Finally, the evaluation index system of citrus industry chain vulnerability were constructed.

    This paper has constructed a model of factor analysis based on 362 questionnaires of vegetable customers, and studied the key products characteristics that influencing the consumers' purchase decision of fresh agricultural products. The results indicated that there were four common factors influencing consumers' purchase decision of fresh agricultural products as follows: the promoting factor, the quality and safety factor, the price factor and convenience factor. The influence degree of these four common factors on the consumers' purchase decision of fresh agricultural products decreased successively. Based on this the authors put forward the corresponding countermeasures.

The reasons were as follows, the farmers and agricultural scientists were lack of intellectual property awareness, the government arranged agricultural technology management, and it's difficult to protect the agricultural intellectual property rights for the characteristics of agricultural technology. So, we should innovate the agricultural technology management system, molulate the plant variety system, and severely punish those who violate the variety law.

    Rice is one of the most important food crops in our country. Two major greenhouse gases, methane (CH4) and nitrous oxide (N2O), play significant roles in global greenhouse effect. This paper reviewed the effects of irrigation, fertilization, rice varieties, cropping patterns, soil\|climatic conditions and other factors on emissions of CH4 and N2O from rice fields. Considering the synthetic greenhouse effect, some measures of reducing greenhouse gas emission in rice fields were also put forward such as varieties and cultivation, fertilization and pesticide application, improving farming system, development of biogas energy technologies, etc. This paper might provide some references for developing low carbon technology in rice field.
