    A simple method for hybrids identification by using SSR molecular markers in barley

    To provide better service to barley breeding and barley hybrids purity test in future, SSR markers were used for hybrids identification in this study. The results showed that a kind of fast, effective and economical method of barley hybrid identification was obtained by the comparison of two kinds of DNA extraction methods and two kinds of detection methods. The method of simple DNA extraction combined with agarose gel electrophoresis could identify barley hy brids rapidly, efficiently and economicly.

    Effects of soybean antigen protein on the growth performance of weaning piglets

    Forty\|eight piglets (Duroc×Large white×Landrace) weaned at 21 days were used to study the effects of soybean antigen protein on the growth performance in a period of 15 days. The results showed that soybean antigen protein significantly decreased AFI by 25.88%(P<0.01)and increased diarrhea rate by 58.77% (P<0.01) compared with the control group. The serum D\|xylose detection indicated that the serum D-xylose concentration was decreased by 62.39%(P<0.01), 68.54%(P<0.01), 67.27%(P<0.01), 67.98% (P<0.01) and 6995%(P<0.01) after 1, 2, 3, 4 h and 5 h respectively compared with the control group. The lengths of the villi within the jejunum and ileum tissues of the test group significantly decreased by 27.56% and 22.43% (P<0.01) and the crypt depth significantly increased by 30.01% and 53.03% (P<0.01) compared with the control group. There was no significant difference between duodenal villi length and crypt length. The activity of NF-κB in jejunum were increased 71.87%(P<0.01). The results indicated that soybean antigen protein in diet exerted a negative influence on growth performance of weaning piglets. Soybean antigen protein could regulate the nuclear factor activity of intestinal mucosa tissue, and affect the morphology and structure of intestinal mucosa tissue, furthermore, affect feed intake, diarrhea rate and growth performance of weaning piglets.

    Spatial distribution patterns of Tomato leaf curl virus and its vector Bemisia tabaci

    The infection status of Tomato leaf curl virus in the early period of tomato planting in 18 winter greenhouses were studied, and the sampling survey of Bemisia tabaci was also conducted in 14 greenhouses. The results indicated that the spatial distribution patterns of Tomato leaf curl virus which was distributed as aggregation distribution was consistent to that of Bemisia tabaci. The Iwao m*-m models of the spatial distribution patterns of Tomato leaf curl virus and Bemisia tabaci were as follows: m*=-0.209060+1.3659m and m*=-0.299134+1.4171m. The sampling equation of tomato leaf curl virus disease and Bemisia tabaci were best fitted n=t2/D2(0.79094/m+0.3659) and n=t2/D2(0.79848/m+0.4171), respectively.

    Current application of large number of commercial asparagus bean cultivars with very high genetic similarity caused more and more disputes on seed quality. In this research, a set of SSR markers for asparagus bean were developed based on a bioinformatics-based approach. Ten of these markers were defined as diagnostic markers by genotyping a core collection of asparagus bean consisting of 44 lines. Based on this, a rapid and precise SSR-based procedure for asparagus bean seed quality surveying was established, which would meet the demand of rapidly developing asparagus bean industry.

    Two\|factor experiment with different nitrogen dosage and planting density were studied to provide further evidence for effect of nitrogen dosage and planting density on the yield and commercial quality of autumn broccoli. The results showed that the days from field planting to squaring of high density planting and less nitrogen treatment were significantly less than that of spaced planting and more nitrogen treatment. The diameter of flower bud significantly decreased in the treatments of middle or high planting density and nitrogen dosage. The broccoli unit yield increased under high planting density and nitrogen dosage treatment. Furthermore, it was beneficial to improving the pass rate of fresh broccoli under the middle level of planting density and nitrogen dosage, and the pass rate significantly decreased under the low level of planting density and nitrogen dosage. According to the field conditions, the planting density should be 2 500-2 800 plants per 666.7m2 and pure nitrogen dosage should be 25-40 kg per 666.7m2 in consideration of high quality, high yield, high effect, and fresh export. In the condition of middle or high fertility, the lower range of plant density and nitrogen dosage was more suitable.

    The effects of different concentrations of 6-BA on gas exchange and chlorophyll fluorescence parameters in different cucumber types under low light stress in greenhouse were studied. The results showed that application of optimal concentrations of 6-BA could obviously alleviate the decrease of net photosynthetic rate, stomatal conductance and transpiration rate under low light stress, indicating 10 μmol·L-1 was suitable for cross combination of Huanan type 07QP56 and 2, 5 μmol·L-1 was suitable for cross combination of European mini type 07QP56. Meanwhile, 10 μmol·L-1 6-BA could also alleviate the decrease of Fv/Fo, Fv/Fm in 07QP56 under low light stress as well. It was also indicated that 07QP51 showed higher resistance to low light stress compared to 07QP56, and 07QP51 was more sensitive to the higher concentration of 6-BA.

    Establishment and application of detection methods for 15 pesticide residues in beer barley

    Fifteen pesticides were determined by solid-phase extraction and gas chromatograph-mass spectrum(SPE-GC-MS), and these pesticides were α-HCH, β-HCH, γ-HCH, δ-HCH, 4,4’-DDE, 2,4’-DDT, 4,4’-DDD, 4,4’-DDT, dimethoate, pirimicarb, acetochlor, malathion, triadimefon, triadimenol and deltalmethrin. The precisions of SPE-GC-MS ranged from 1.15% to 9.80% with mostly lower than 5%. The recoveries were 77.78%-112.00% with the detection limits in the range of 0.77-10.34 μg·L-1, which demonstrated it was a good method. SPE-GC-MS was used to determine pesticide residues of barley from Jiangsu, Sichuan, Xinjiang, Yunnan, Heilongjiang, Inner Mongolia, Gansu. All of them didn't exceed its national standard. Fifteen pesticides were classified into two groups, according to content levels. Four pesticides in one group were high in level but lower than national standard level, including 4,4’-DDT (≤ 0.032 mg·kg-1), β-HCH (≤ 0.039 mg·kg-1), triadimefon (≤ 0.093 mg·kg-1) and dimethoate(≤ 0.030 mg·kg-1). Furthermore, residues of dimethoate have only been detected in Jiangsu. The other group including 11 pesticides was much lower than national standard level or even non-detection. The low residues may be related to the high resistance to diseases and pests of barley.

    Alcalase protease was used to hydrolyze the protein extracts of Spirulina to produce polypeptides. The effects of pH, enzyme dosage, hydrolysis time and temperature on degree of hydrolysis were investigated by single factor test respectively, and these factors were optimized by response surface methodology (RSM) which was designed based on Box-Behnken central combination. The optimum hydrolysis conditions were as follows: pH=7, enzyme dosage (ratio of enzyme to substrate) 4 300 U·g-1, hydrolysis temperature 55℃ and time 160 min.The hydrolysates of Spirulina protein were ultrafiltrated by 5 kD hollow fiber ultrafiltration membrane and purified by SephadexG-25 column chromatography with PBS (pH=7) as eluent. Low-molecular-weight peptides were collected, and the molecular weight was from 307 D to 1 450 D.

    The gene fragments of Gα subunits were isolated from Colletotrichum hanaui by using homologue cloning method. The 5′and 3′ flanking sequences of the fragments were cloned by TAIL-PCR method and sequenced. After sequence assembling, the complete senquences of cagA, cagB and cagC were obtained. The structures of the three genes were characterized by comparing genome and CDS sequences. The cagA has an open reading frame (ORF) of 1 380 bp interrupted by 5 introns and putatively encodes a 355aa protein. The cagB has an ORF of 1 283 bp interrupted by 3 introns and putatively encodes a 353aa protein. The cagC has an ORF of 1382 bp interrupted by 4 introns and putatively encodes a 354aa protein. The results of amino acid sequence alignments showed that the predicated proteins were homologous with Gα subunits of other ascomycetous phytopathogens such as Magnaporthe grisea, Gibberella zeae, with the amino acid identity between 75% and 99%, respectively.

    Analysis on combining ability of grain phytic acid content in indica hybrid rice

    Three\|line indica hybrid rice including 5 indica CMS lines with broad representation and 16 restorer lines, the grain phytic acid content and some important yield trait indexes of the hybrid rice and their parents were determined and analyzed through incomplete diallel cross, meanwhile, General combining ability(GCA) of the parents and specific combining ability(SCA) of hybrid rice for grain phytic acid content were evaluated. The results showed that both female and male parents contributed to grain phytic acid content of hybrids, but F value for variance of female parents was greater than that of male parents. The hybrid rice whose parents had low level of GCA did not always show low level of SCA. Among 80 hybrid combinations, no significantly correlation between grain phytic acid content and yield, plant height, panicle length, effective number of panicles, total number of grains, seed-setting rate or other important agronomic traits was indicated, but there was significantly positive correlation between grain phytic acid content and 1000-grain weight.

    According to OmpA gene of Pasteurella multocida published in GenBank, a pair of primers was designed. Using the primers, full\|length OmpA of C51-2-499, C51-3-500 was amplified by PCR. It was cloned into pMD18-T vector and then sequenced. The result showed that the full-length of OmpA was 1.068 kb containing a 1.068 kb open reading frame. The relative molecular weight of the deduced amino acid sequence (223 amino acid residues) was 37 969.8 kD and the pI value was 8.90. The alignment results showed that the nucleotide sequence homologies were 89.7%-99.1% and the deduced amino acid homologies were 82.9%-98.6% compared with six reference OmpA sequences of Pasteurella multocida published in GenBank.

    In order to provide the technical support to the safe production and dietary risk assessment for pakchoi cabbage, the residue trial and dietary exposure estimation of indoxacarb and chlorpyrifos in pakchoi cabbage were carried out. The results showed that the normal dosage of indoxacarb (40.5 g·hm-2) and chlorpyrifos (540 g·hm-2) were used under the protected field conditions, the primary residues were 388 and 20.6 mg·kg-1, and the half-life (T1/2)were 1.4 and 1.1 d, respectively; After 7 days of spraying, the residue of indoxacarb and chlorpyrifos could meet the limits of CAC, the U.S.A. and Japan. The dietary exposure estimation showed that different age groups had different risk quotient, the risk quotient of 2-10 age group was about 50% higher than the adults’. The theoretically chronic risk quotient of indoxacarb was lower than 100 at the 5 d interval with the normal dosage and 7 d interval with double dosage, the theoretically acute risk quotient value was always lower than 100, which was all acceptable. The theoretically chronic risk quotient value of chlorpyrifos with different intervals was approaching or above 100, the theoretically acute risk quotient decreased to be lower than 100 after 3-day interval. Therefore, the residue trial and dietary exposure estimation of indoxacarb and chlorpyrifos showed that the dietary exposure risk of indoxacarb was low and safe. But the use of chlorpyrifos could cause high risk, and should not be used in the pakchoi cabbage production.

    According to the conserved gene sequences of Duck plague virus(DPV) in GenBank, one pair of specific primer was designed. A rapid diagnostic technique of PCR was established by optimizing the reaction condition. The DNA segments amplified were 1 293 bp. The experiments had proved that PCR possessed a high specificity. And the sensitivity test results indicated that PCR was more sensitive, which could detect DHV with only 15 pg·mL-1 DNA. Detection of field samples from viral diseased ducks also confirmed that the PCR method could identify the infection of DPV, which was specific, rapid and sensitive.

    A detection method has been introduced for cathay hickory grade based on machine vision in this paper. On the basis of adequate understanding of cathay hickory shape, its digital images were obtained using CCD camera. The width, length and weight of the fruit were extracted through mathematical morphology method. According to these characteristics, the mathematical model was established. The weight for each characteristic was endowed based on some suggestions of jobholders. Comprehensive evaluation standard for cathay hickory grade was established. It was demonstrated that the classification method was objective and reasonable, and the classification results were more accord with peoples consumer psychology and visual habit.

    The effect of different sample preparation conditions on the detection of total SO2-3 in mushrooms was studied. And the optimal ion chromatography detection conditions were selected and the detection method of SO2-3 in mushrooms based on ion chromatography was set up. The detection limit of apparatus was 0.05 mg/kg, the recovery rate from low to high concentration was 86.0%-94.6%, and the RSD value was 1.2%-2.3%. The results provided a method for the further research on the relationships between total SO2-3 and SO2 content of mushroom.

    A hydroponic experiment was conducted to study the effect of low phosphorus (0.5 mg·L-1) on root length, dry weight, nitrogen, phosphorus, potassium uptake and root phosphorus uptake kinetic parameters of japonica rice varieties Zhejing 22, Zhejing 27, Xiushui 110, Xiushui 09, Xiushui 128, Xiushui 63, Jia 991 and Jiahua 1. The results showed that there were significant differences among varieties in root length, biomass, nitrogen, phosphorus and potassium uptake and root phosphorus uptake kinetic parameters (Imax, Km and Cmin). Zhejing 22, Zhejing 27, Xiushui 110, Xiushui 09 and Xiushui 63 showed lower Km and Cmin, but higher Imax values under phosphorus deficiency stress, and they also showed longer root length, higher dry seedling weight and phosphorus utilization efficiency. Meanwhile, Jiahua 1, Jia 991 and Xiushui 128 showed higher Km and Cmin, but lower Imax values and lower biomass, phosphorus utilization efficiency under phosphorus deficiency stress.

    Effect of hypoxia stress on morphological and physiological characters in watermelon and gourd rootstock

    The gourd rootstock and watermelon were planted hydroponically to investigate the effect of anaerobic environment on antioxidant enzymes and metabolites. The results indicated that root activity of gourd rootstock and watermelon both decreased under hypoxia stress, and activities of ADH, SOD, APX, proline content firstly increased and then decreased. The variation range of antioxidant enzymes in gourd rootstock was greater than in watermelon. Moreover, gourd rootstock contained higher content of MDA than watermelon during treatment time. But anaerobic environment could not affect the activity of CAT in rootstock and watermelon.

    Through measuring the amounts and analyzing the components of feces of heifers and cows in the intensive dairy farm in four seasons, the organic matter pollutants producing coefficient (PPC) were calculated and compared between heifer and cow in all seasons. The results showed that the organic matter PPC of the heifer was 1 966.18 g·head-1·d-1, that of the cow was 3 688.74 g·head-1·d-1 in the intensive cow farm in the fourth season, the organic matter PPC of cow was much higher than that of heifer.

    Through field plot experiment,the effect of brackish water irrigation on dynamic change in soil salts as well as growth and yields of spring maize were studied. The results showed that brackish water containing 3 g·L-1 mineralization resulted in accumulation of salt in soil profile, and decreases in leaf area, plant height, leaf chlorophyll content, dry matter and yield of spring maize. The decrease in maize leaf area was faster in later period of maize growth. The decrease in accumulation of dry matter caused lower maize yields. Brackish water irrigation caused 28% loss of maize yields in the dry year, and 10.4% loss in the rainy year. Maize yield with well-canal combining irrigation in the dry year and the rainy year declined 14% and 7.4% respectively.

    Using japonica variety XiuShui 128,field experiments were carried out to investigate the effects of sulfur-polyester resin-coated urea(SPCU)and polyester resin-coated urea(PCU)on rice grain yield and nitrogen use efficiency(NUE). The results showed that:(1)compared to PU 70%(prilled urea)with N content at 147 kg·hm-2,the rice grain yields significantly increased by 5.26% with a single basal application of SPCU 70% under the same N rate which were 8 022.5 kg·hm-2,and the NUE were 49.32% which significantly increased by 38.42%;(2)compared to PU 100% with N content at 210 kg·hm-2,the rice grain yield showed no significant difference but the NUE significantly increased by 33.69% with a single basal application of SPCU 70%;(3)there was no significant difference between rice grain yields of SPCU and PCU; however, NUE of SPCU significantly increased compared to that of PCU.

    A field experiment was conducted to study the effects of different fertilizations on yields of rice and paddy soil microbial biomass. The results showed that eco-fertilizer combined with organic fertilizer could obviously enhance rice yield and improve yield components, and the yield increased by 10.5% compared with organic fertilizer treatment. Yields of rice in plot used chemical fertilizers increased by 8.1%, but yields of mineral powder mingled with organic fertilizer treatment and organic fertilizer treatment were almost equal. The peak value of chlorophyll content (SPAD value) in rice was observed on 15 d after transplanting under the normal planting condition with chemical fertilizer, while it was observed on 80 d after transplanting under organic fertilizer treatment. The mean soil microbial biomass-C in the treatment of NST was higher during whole rice growth stage, CK was the second, and GP showed a decrease compared with CK and its variation coefficient was the highest.

    The relation between carapace color and growth of Procambarus clarkii

    There is a change of carapace coloration from cyan to red during growth and gonad development of Procambarus clarkii. The growth, color of carapace and physiological indexes at different growth stages from young to adult crayfish were observed and measured. The results showed that there were remarkable correlations between carapace color transformation and ontogeny and growth speed of crayfish. The carapace color changed from cyan to red, accompanied with the mature gonad,and the gonadosomatic index and liver/body weight ratio increased,the molt cycle was prolonged and the relative growth speed decreased. The growth speed of cyan shell crayfish was significantly higher than that of red shell crayfish. The tests also indicated that environmental factors, such as temperature, light density, directly affected development and maturation of crayfish with the change of carapace coloration.

    Three stages of colored rice development in China were put forward by rice breeding research combined with historical data. The breeding of colored rice and research progress were reviewed through line breeding, cross breeding, heterosis utilization, biotechnology breeding, space mutation breeding and radiation breeding. Furthermore, the local variety resources of colored rice were evaluated and their genetic characters were studied.     “Zhejiang farmer mailbox” was established and designed according to the general construction planning of “digital Zhejiang” and the principle of “promoting in use” for farmers through developing convenient, practical and manageable system application software in use of internet technology. Based on the principles of users' need and the purposes of convenience and practicability, the farmer mailbox should be registered and used in real name system and was connected with mobile phones, which made it possible for farmers to communicate with each other bi\|directionally on-line or off\|line by means of computer and mobile short message. The farmer mailbox was a public service platform led by government and oriented to “Three Agriculture (Agriculture, Farmer and Village)” with functions such as personal communication, e-commerce, e-government, agricultural technology service, office communication and information integration gathered in internal whole.

    Southern-type solar greenhouse was a newly developed greenhouse with good effect of heat insulation during winter time. Its typical dimension was 50 m in length, 8 m in width, 1.7 m in southern wall height and 3.85 m in northern wall height. The eastern, western and northern walls were constructed with thermal insulated wall composed of supporters, foam boards and films, which could be disassembled after use. The southern side was constructed with roof, supporter, film, outside coverlet and coverlet roller. In order to increase the ventilation capability, the ventilation windows were located on the southern, northern roof and northern walls. The diurnal variation of light intensity, temperature showed relatively lower in the early morning and evening, and higher in the midday, while the relative humidity showed the reverse change trend. Compared to the normal greenhouse, more sunlight entering could be captured in the southern-type solar greenhouse. The inside temperature of southern-type solar greenhouse could be maintained above 5 ℃ during night in winter and the temperature difference between inside and outside could achieve 4.7-12.7 ℃ due to the good heat insulation of external heat preservation quilt. During the sunny days, the inside temperature increased rapidly and might be above 40℃, so ventilation cooling was demanded. Due to the good sealing performance, the relative humidity in southern-type greenhouse was high, and the air exchange management was also necessary. The southern-type solar greenhouse was a good growing environment for tomato in the period of January, February and March, in which the growing and harvesting time could be prolonged for one month, and the yield could increase 20%-50%.

    Study of farmland disposal volume of biogas slurry and its effect on peppers yield, quality and soil fertility during the production in greenhouse were investigated through a field plot experiment. The results showed that biogas slurry could be a substitute for chemical fertilizer that applied in pepper production system, the yield of pepper in the treatment with biogas slurry amount equivalent to 2-3 times of the chemical nitrogen fertilizer was similar to the yield in the treatment with conventional chemical nitrogen fertilizer. Moreover, quality of pepper was improved, and the chemical fertilizer and cost could be saved. Furthermore, the application of biogas slurry farmland disposal also significantly increased soil organic matter, total nitrogen, available nitrogen, available phosphorus and potassium content. The recommendation amount of biogas slurry was 900 t·hm-2[i.e., nitrogen 530.6 kg·hm-2 (three times more than the conventional mineral nitrogen fertilizer), P2O5 37.8 kg·hm-2, K2O 311.4 kg·hm-2, COD 900 kg·hm-2] in greenhouse pepper production system.

    House keeping gene GAPDH(glyceraldehyde-3-phosphate dehydrogenase) was often used as inner control in quantitative or semi-quantitative PCR test to determine relative expressive amount of target genes. Based on EST monoclones of GAPDH gene obtained from Guzmania full length cDNA library, its full length cDNA sequence was obtained by Primer Walking sequencing. The gene, designated GoGAPDH1, was 1 790 bp in length encoding 421 amino acids. Some characters involving in cDNA sequence, amino acid sequence, conservative domain and genetic relationship were analyzed by Blast, ExPASy, ProtParam, SPOMA, Find Conserved Domains(NCBI), ClustalX and MEGA softwares. The results showed that GoGAPDH1 shared 82% similarity with cytoplasmic GAPDH in maize, which indicated that GoGAPDH1 might be a cytoplasmic GAPDH.

    The genetic diversities of 17 samples belonged to Diospyros were analyzed by ISSR molecular markers to identify the classification of Jinzao persimmon. After previous ISSR-PCR of Diospyros lotus L., 997 bands were amplified with 37 polymorphic primers screened from 100 ISSR primers, and the average number of amplified bands was 26.94 per primer. Among these bands, 973 bands were polymorphic and the polymorphism percentage was 97.59%. The dendrogram showed that the 17 individuals were classified into two groups, among which 6 of the Jinzao persimmon materials made up a big group, and another group was composed of 3 Diospyros species. There was an obvious genetic boundary between the two groups. The results of hierarchical cluster analysis combined with the observation of anatomical characteristics of Jinzao persimmon, indicated that it might be a new species of Diospyros

    Fermentation kinetics of tautomycin production by Streptomyces spiroverticillatus

    In a 15-L fermentor, the batch fermentation kinetics of tautomycin production by Streptomyces spiroverticillatus was studied. During the batch fermentation process, pH, dissolved oxygen (DO) and temperature were obtained on-line. The production of tautomycin, biomass and total sugar were measured off-line by bioassay method, glucoseamine method and dinitrosalicylic acid reagent (DNS) method, respectively. Based on Logistic and Luedeking-Piret equation, the mathematical dynamic models were proposed for cell growth, product formation and substrate consumption. The parameters of the kinetic models were determined with typical experimental data. The fitting degree was 0.9922, 0.9897, 0.9934, respectively. The simulated results agreed well with the experimental data in the evincive experiment and the models showed good prediction ability under the condition as follows: initial total sugar content of 67.2606-74.8759 g·L-1, inoculation quantity of 8% (V/V), the agitation rate of 150 r·min-1 and the ventilation of 1.1 vvm. The models may provide good rationales for the optimization of tautomycin production in the fed-batch fermentation and in scaled up.

    E. coli K88 is the main pathogen of post-weaning diarrhea (PWD). The existing commercial vaccines could effectively control diarrhea caused by E. coli in newborn piglets from immuning sow, while it was unlikely effective to PWD. Oral vaccine could induce antibodies produced in local mucosal, prevent E. coli adhere to intestinal epithelium, thereby prevent PWD. To study the biological characteristics of K88 and its receptors at molecular level was important to develop oral vaccine. The progresses in the basic characteristics, protein subunits and receptors of K88 fimbriae at home and abroad were decribed, furthermore, the application prospect of engineering bacteria vaccine and plant-based vaccine were discussed.

    Using different concentrations of chitosan, methylcellulose and single glyceride, different ratios of chitosan composite membrane coating were made to study the physiological changes of cucumber based on the orthogonal experiment of L9 (33). The results indicated that chitosan composite membrane characteristic was good, which could effectively maintain the cucumber sensory quality, reduce weightlessness lose, respiratory intensity and hardness of cucumber, and could also inhibit the loss of soluble solid and chlorophyll content. The preservation effect of chitosan achieved the remarkable level. Optimized concentration of the chitosan composite film was the ratio of 2.0 % chitosan, 0.2 % single glyceride and 0.2 % methylcellulose.

    Polyploidy in Anoectochilus roxburghii induced by colchicine in vitro

    Explants of Anoectochilus roxburghii were adopted and treated with 300 mg·L-1 colchicine for doubling the chromosome number. As the treating time being prolonged, the toxic effect of colchicine became more evident because the death rate of explants increased. Tetraploid cells were induced from the experiments when the explants were treated with colchicine at this level for 3-19 days. The best chromosome doubling results of 72.7% tetraploid cell frequency and 91% induction rate were obtained when explants were treated for 13 days. The number of chromosome of diploids and tetraploids were 40 and 80, respectively. The stoma size of tetraploids was larger than that of diploids. No difference was found between the stoma size of diploids and mix-ploids.

    Partial region of the PRRSV GP5 gene was amplified by RT-PCR and cloned into pGEM-T Easy vector. Serial dilutions of the recombinant plasmid were used as standard templates for RT-PCR to quantify the virus genomic copy number. A SYBR GreenⅠreal\|time PCR was developed to detect PRRSV. Sensitivity analysis showed that the developed SYBR Green I real-time PCR could detect 10 copy/μL. The specificity assay exhibited that the other porcine pathogens, such as PPV, PCV, JEV, PRV, HCV and SIV could not be detected by this method. Then the established method was used to detect the clinical samples. The results showed that 15 positive samples out of 16 suspicious positive samples could be observed by real-time PCR and 12 positive samples could be detected by normal RT-PCR. All the results suggested that the real-time PCR we established in current study showed the characteristics of sensitivity and specificity, and could be used in clinical diagnosis and epidemiological investigation .

    Correlation and path analysis for 11 agronomic characteristics of beer barley with plot yield, 1 000-grain weight, protein content and α-amylase content. The results showed that: some traits which had prominence correlation with yield were the number of seeds per one plant, grain weight per one plant, the number of seeds and the seed weight in main ear. And 1 000-grain weight, protein content and α-amylase content were significantly affected by the length of main ear, grain weight per one plant, panicles per one plant, the number of main ear, tiller number, basic seedling number and effective panicles number per one acre, in which the h2B of basic seedling number and grain weight per one plant were less than 50%; the heredity effect of the number of main ear and grain weight of main ear was more than other characters, and they had a greater influence on 1 000-grain weight and α-amylase content. Therefore, the number of main ear and grain weight of main ear should be paid more attention in the practice of breeding.
