    Analysis of the hemagglufinin genes of four avian influenza viruses of H9N2 subtype isolated from northern China

    Four H9N2 avian influenza viruses were isolated from sick chickens in Northern China.The sequences of the hemagglutinin genes of these isolates were determined by RT-PCR and sequencing.The homology of these nueleotide 8e—
    quences and putative amino acid sequences were compared with H9N2 in GenBank.Phylogenetic tree based on different regions in nucleotide sequences ofHA gene cDNA was also analyzed.Although the isolating time of four viruses was different,they were highly homologous and all belong to the same sublineage of H9N2 Eurasian lineage,A/CK/HK/G9/97一like group.Their amino acid sequences that involved in HA cleavage sites,receptor binding sites and potential glycosylation sites were also similar to the above sublineage.However,two of the four isolated viruses,CK SD B1 98 and CK BJ L1 05
    had a deletion of potential glyeosylation site because of a single nucleotide variation on HA gene.

    Bamboo soda pulping waste water produces a great quantity of solid waste in the process of acidification.The main component of the solid waste is lignin.Agricultural udlization of the solid waste was conducted in the lab and pot eX—
    periments.The results showed that rock phosphate could efectively neutralize the free acid in the solid waste to,enhance the pH to a suitable level f0r tl1e agrieultu~ use.Rock phosphate and solid waste should be mixed at the ratio of 1:1 and the mixture should be stored more than 4 d to reach the highest activating efficiency.Further study showed that phosphorous in rock phosphate was predominantly activated by free acid.The result of pot experiment indicated that the nutritional effect of the mixture of solid waste and rock phosphate on the growth of radish(Raphanus sativus L.)and Chinese cabbage (Brassica campestris ssp.chinensis L.)was between rock phosphate and calcium magnesium phosphate.

    This paper discussed briefly the meaning and values ofurban biodiversity as well as the process ofdevelopment of urban biodiversity conservation,analyzed especially approaches to urban biodivemity conservation.As results of the thesis,two standpoints are put forward.One is that landscape eco-planning is the best approach to urban biodiversity conservation.And the other is that the ultimate purpose of urban biodiversity conservation is to establish sustainable cities by constructing sustainable urban landscape ecosystem.

    The 893bp fragment of root-specific promoter of tobacco TobRB7 gene, namely NT1, was cloned by PCR. The expression vector pBI-NT1 : : Pro, driven by tobacco root-specific promoter NT1, was constructed from pBI121 by substituting the CaMV 35S promoter and GUTS gene with NT1 and Pro, a tomato broad-spectrum R gene, respectively. Through freeze and thaw method, the pBI-NT1 : : Pro vector was introduced into Agrobacterium tumefaciens strain EHA105, and the explants of tomato cotyledons were then transformed. PCR test result of NPT H gene indicated that transgenic tomato lines were obtained.

    The effects of the initial pH of medium,nutrition elements including carbon and nitrogen Soul~es,the concentration of metal ions including Mg2+ and Cu2+ on the growth and parasporal crystal production ofBacillus thurlngiensis strain
    YJ-2OO0,which shows no toxicity to the neonate lalwae of the domestic silkworm. Bombyx mori but showed high toxicity to various important lepidopteran species.were studied to define the suitable culture conditions for this strain.The culture conditions,which are the most optimum not only for the high biomass,but also for the highest yield of the parasperal crystals of this strain,were as follow:the initial pH of the medium was 6.5,the medium carbon source was maltose,the medium nitrogen source was peptone plus (NH4)2SO4 with the ratio of 1:1,the concentrations of Mg2+ and Cu2+ in the medium were 1×10-3 mol/L and 0.008×10-3 mol/L, respectively.

    5 ‘UTR of swine fever virus(SFV)was amplified by RT-PCR and cloned into pMD-18 simple T vector to construct a recombinant plasmid pSF,which was used as standard positive template for the fluorescent quantitative PCR(FQ—PCR)to
    detect SFV.The pSF was identified by PCR and DNA sequence analysis.The plasmid Was stable after 6 months storage at 一20℃ .When the plasmids DNA of 1.0×10 9 , 1.0×10 7 and 1.0×10 7 copies/2 μL were used as template,the largest variation coefiqeient of Ct value was 1.55%.and the FQ—PCR provides a broad linear range from 1×10 7 to 1×10 11 copies of DNA per reaction.The pSF was specific and stable for the FQ-PCR.The method can be used for the clinical diagnosis.

    On the controlled condition of 26℃ and RH 70% ,we investigated the absorption function of the egg pedicel of tobacco whitefly,Bemisia tabaci(Gennadius)(Homoptera:Ahyrodidae).Firstly,we studied the efect of pesticide on development of egg laid on the artifical membrane apparatus and on plant which root was immersed in pesticide.As a result,we found the disadvantage of pesticide on development of whitefly eggs significantly,the eggs on artifical membrane with pesticide were dead entirely,but eggs development well on control,Secondly,we investigated the effect of different
    plants on develolmlent of egg,results showed that there were significant difference on survival and development duration on eggs ovipesit on different plants,and the sequence of plants on which the survival of eggs from high to low was cabbage>cucumber>capsicum>eggplant>corn, the sequence of plants on which development duration of egg from long to short was cabbage>cucumber>eggplant>capsicum>corn.Lastly, we transferred the 1st larva which hatched from the eggs laid on the above different plant to the other common plant,tomato plant,and observed their development,the result showed that the duration sequence of 1st instar from long to short was on apsieum>cabbage>cucumber>eggplant.It is sugested that the substance ofmedium entered into the egg through whitefly egg pedicel,and it affect the development of whitefly egg and the following 1st larva.

    The aim of this investigation was to study the efect of brilliant cresyl blue(BCB)staining on IVM-IVF in cow.For this purpose,oocytes were stained by BCB and selected by IVF examination before mature culture.COCs without BCB staining was considered as control group,and COCs stained with BCB in 26 μmol/L for 90 min as treatment group before mature culture.Treated oocytes were divided into BCB一(eolourless cytoplasm)and BCB+ (coloured cytoplasm).After
    1VM,oocytes were fertilized in vitro(ⅣF).The cleavage rate of eggs at 48 h and the proportion of blastocysts at 8 d after 1VF were compared.The results showed that:(1)The BCB+ oocytes had a signiiicanfly higher proportion of mature rate (80.83%)than control group and BCB—oocytes(60.20% and 51.92%)(P <0.05);(2)The BCB+ooeytes had a signiticanfly higher proportion of blastocysts(32.99%)than control group and BCB-oocytes(17.64% and 3.57%)(P

    Effect of different NPK supply levels on nutrient absorption and transport of super rice

    Mechanisms of dry matter and nutrient accumulation/transport in super rice(Liangyoupeijiu)planted under different levels of NPK supply were studied.The results showed that:(1)The maximal value of dry matter accum ulation in
    super rice emerged in the midge growth phase,which was approximately 50% of the total content.Nitrogen(N)supply was positively correlated with the accumulation of dry matter with R value of 0.8554,however phosphorus(P)and potassium (K) supply had minor effect.(2)Accumulation of N,P and K in super rice had diferent characteristics.Accumulation of N and P increased with supe r rice growing;an d accumulation of K showed a single—peak curve in the whole growing period.Most of N (85.4%) was accumulated in the early and middle growing stages.Both P and K accumulation reached their maximal levels of 58.0% and 64.6% in the middle growth phase.Nutrient accumulation was positively correlated withthe supply ofN,P and K.N and K supply can significantly afected themselves accumulation levels,but the effects of P supply and interaction of N,P and K on nutrient accumulation were not significant.(3)In seeds of super rice,values of accumulated N,P and K contents were 99.2 kg/hm 2,53.2 kg/hm 2 and 23.8 kg/hm 2,respectively.(ratio:4.2:2.2:1),among which 75% of N,70% of P and 100% of K carrie from vegetative mass.Significant negative correlation between N transport rate in vegetative n and N supply level was observed with R value of 0.9610,while efects of P and K were not significant.Increasing P supply level can enhance P transport rate,and the effect of N on P transport rate showed a single-peak curve.With increasing of K supply,transport rate and lose-rate of K elevated .Effect of N on K transport rate also showed a single-peak curve,which suggested that too high or too low N supply could negatively affect the accumulation of P and K.

    Genetic diversity of germplasm resources of Prunella vulgaris Linn based on AFLP marker

    Amplified fragment length polymorphism (An )analysis of eight prunella germplasm resources was carried out to explore a variety of the genetic diversity.Using AFLP markers, 32 pairs of selective amplification primers was screened out.The similarity among the pnmeHa germplasm was analyzed and phylogenetic tree was constructed .The results showed that:(1)genomie DNA extracted from prune~a by SDS method was good to satisfy AFLP analysis;(2)10 m rs of primers with stronger polymorphism,better banding and higher resoluing power were screened out from 32 pairs of selective amplification primers;(3)divided eight germplasms into three categories through constructing evolutionary tree;(4)Through the construction of evdutionary tree,the eight germplasms were divided into three categories.Prunella germplasm resources are rich in genetic diversity,certain morphological characteristics and have relationship with genotype.

    production.Among the ten examined factors of the solution,four statistically significant ingredients,which were selected by Plackett—Bunnan design and analyzed by response surface methodology,were utihzed in order to optimize the process in the Box-Behnken design .The results showed that the production of pigment was double when the components were set as follows:NaCl=0,CaCl2=0.887 g/L,NaHCO3=0.639 g/L,H3BO3=0.058 g/L.

    An information collecting technology based on unmanned gyroplane was trially tested.The characteristics of geometric distortion from acquired images and their correction methods was introduced.The results showed that images from gyroplane exhibited significant geometric distortion,and the precision of the corrected images can satisfy the requirements in precision agriculture.

    This experiment was carried out with browning caused by enzyme reaction of phenolics respiration rate,ethylene production,fruit color,quality and senescence of ’Whangkeumbae' pear fruit to study effects of different 1-MCP concentrations on browning inhibition,and value the effects of modified atmosphere packaging (MAP) on CO2 sensitivity of fruit treated with 1-MCP.The results showed that 0.5 μL/L 1-MCP treatment was more effective on browning inhibition compared to control,and occurred slight browning offresh with 1.0 μL/L 1-MCP treatm ent.This study also showed that two concentrations of 1-MCP combined with MAP lead to browning of flesh and peel at different levels.1-MCP treatment combined with 0.02 mm PE bags was not suitable for storage of pear fruit.O.5 μL/L 1-MCP with unpackaged bag was recommended to storage of pear fruit.

    We investigated the agronomic and physiological characters of the lily bulbs.which were treated with atrazine in different growth phases before sprouting.The results showed that atrazine had different effects on the plants in different
    growth phases.Comparatively,the bulbs treated before sprouting were less significantly affected except some physiological changes.The content of chlorophyll,soluble sugar content and dark respiration rate increased in bulbs of the treated plants,while net photosynthesis rate,special leaf weight and dry matter weight decreased.The result of correlative analysis showed that the content of soluble sugar was positively correlated with dark respiration rate.The increase of soluble sugar content disturbed the photosynthesis of plants to some extent.Thus,it was demoastmted that atrazine could affect the accumulation of photosynthetic products in lily plants through influencing the metabolism and transportation of sugar,and CO2 fixation in the chloroplast.

    An inactivated vaccine with microeapsule shape against fowl cholera was prepared by drying in liquid,and the shape ,distribution of particle size,entrapment eficiency and capability of slow release of the microcapsule were studied.The experiment indicated that the microcapsule were spherical,and distribution of particle size was about 10—80 μm,entrapment efficiency was over 8O%, accumulated release rate was 43.95% when releasing for 72 h in vitro.Because of its high entrapment efficiency,good capability of slow release,good repeatability and simple process of manufacture,the vaccine can be applied to industrial production.

    The drought resistance of four Lysimachia species under drought stress was evaluated with subordinate function, and the physiological and biochemical indexes such as exosmosis rate of cytoplasm,content of malondialdehyde(MDA),activities of superoxide dismutase(SOD)and pemxidase(POD),content of proline,soluble sugar and soluble protein were measures.The results indicated that the order of drought resistance Was-"Lysimachia alfredii> Lysimachia christinae ‘huaye’and Lysimachia nummularia‘Aurea’> Lysimachia congestiflora.This experiment showed that plant drought resistance was a comprehensive index.
