摘要:
基于抗氧化酶活性,以及可溶性糖、丙二醛、脯氨酸含量等生理指标建立的大白菜耐热性评价方程在应用中存在操作复杂、费时费力等问题,亟需建立简便快捷的大白菜耐热性综合评价方法。本研究以33份大白菜种质资源为材料,通过40 ℃处理20 h模拟高温胁迫,测定高温处理后大白菜的叶绿素荧光参数[PSⅡ最大光化学量子产量(Fv/Fm)、初始荧光(Fo)、暗适应后最大荧光产量(Fm)、PSⅡ有效光化学量子产量(Fq'/Fm')、电子传递速率(ETR)、光下最大荧光产量(Fm')、非光化学猝灭系数(NPQ)、叶绿素指数(Chlidx)、总叶绿素含量(Chl)],以及鲜重(FW)和电导率(REC)共11个指标,采用主成分分析和隶属函数分析方法对供试大白菜种质进行耐热性综合评价。结果表明,这2种综合评价方法获得的结果一致,且与高温下白菜的耐热表型结果吻合。最后,通过逐步回归分析建立了基于叶绿素荧光参数的大白菜耐热性综合评价的回归方程,D4=0.464VETR+0.226VChlidx+0.262VChl+0.191VNPQ+0.192Fm'+0.127mFW。该研究建立了一套简便快捷、方便测量的大白菜耐热性综合评价体系,研究结果可为后续大白菜耐热种质筛选、耐热种质选育与耐热机理挖掘提供技术支撑。
