高温胁迫下杏鲍菇粗多糖对白灵菇菌丝生长的影响

Effect of crude polysaccharides from Pleurotus eryngii on the mycelial growth of Pleurotus tuoliensis under high-temperature stress

摘要: 以杏鲍菇子实体为试材,采用热水浸提法与分级醇沉法制备杏鲍菇粗多糖,获得56%、68%、80% 3种杏鲍菇多糖组分(Pleurotus eryngii crude polysaccharide, PECP),分别探讨3种PECP的体外抗氧化活性,及其对白灵菇菌丝生长和高温胁迫后菌丝恢复的影响。结果表明,3种 PECP组分均具有良好的体外抗氧化活性,其中80%PECP的抗氧化活性最佳。80%PECP在质量浓度为10 mg·mL-1时,羟自由基清除率达 98.85%,总还原力为1.25。质量浓度为 2.5 mg·mL-1 时,80%PECP 对白灵菇菌丝生长的促进作用最显著,菌丝生长速度为 5.37 mm·d-1。此外,添加80%PECP可促进高温胁迫后白灵菇菌丝的恢复,高温胁迫 10 h 后,经80%PECP 处理的菌丝恢复生长速度为 4.23 mm·d-1。同时,与CK-HS相比,80%PECP处理使高温胁迫后白灵菇菌丝体内丙二醛(MDA)含量显著(p<0.05)降低18.55%,过氧化氢(H2O2)含量显著降低43.24%,过氧化氢酶(CAT)、过氧化物酶(POD)活性分别显著提升5.58%和475.00%。除此之外,与CK-HS相比,80%PECP处理使高温胁迫后白灵菇菌丝体内蛋白质和脯氨酸的含量分别显著提高9.44%和16.55%。综上所述,杏鲍菇粗多糖具有良好的体外抗氧化活性,且对白灵菇菌丝生长的促进、抗氧化酶活性的提高及高温损伤的缓解均具有显著作用。

Abstract:

Using Pleurotus eryngii fruiting bodies as raw material, crude polysaccharides were prepared via hot water extraction and fractionated alcohol precipitation, yielding three crude polysaccharide fractions, 56% PECP, 68% PECP, and 80% PECP. The in vitro antioxidant activity of these three PECP fractions, along with their effects on Pleurotus tuoliensis mycelial growth and mycelial recovery after high-temperature stress, were investigated. The results showed that all three PECP fractions exhibited excellent in vitro antioxidant activity, with 80% PECP demonstrating the highest activity. At a mass concentration of 10 mg·mL-1, 80% PECP achieved a hydroxyl radical scavenging rate of 98.85% and a total reductive of 1.25. At a mass concentration of 2.5 mg·mL-1, 80% PECP most significantly(p<0.05) promoted P. tuoliensis mycelial growth, with a growth rate of 5.37 mm·d-1. Furthermore, adding 80% PECP accelerated the recovery of P. tuoliensis mycelia after high-temperature stress. Following 10 hours of 37 ℃ stress, mycelium treated with 80% PECP exhibited a recovery growth rate of 4.23 mm·d-1. Simultaneously, compared with CK-HS, 80% PECP significantly reduced malondialdehyde (MDA) content by 18.55% and hydrogen peroxide (H2O2) content by 43.24% in P. tuoliensis mycelium after high-temperature stress, while significantly enhanced catalase (CAT) and peroxidase (POD) activities by 5.28% and 475.00%, respectively. Additionally, the protein and proline contents in mycelium treated with 80% PECP after high-temperature stress showed significant increased compared with CK-HS, rising by 9.44% and 16.55%, respectively. In summary, crude polysaccharides from P. eryngii exhibit excellent in vitro antioxidant activity and significantly promote mycelial growth, enhance antioxidant enzyme activity, and alleviate high-temperature damage in P. tuoliensis.
