    Agrobacterum tumefaciens-mediated genetic transformation in melon

    The experimental parameters were investigated in Agrobacteraum—mediated tranformation of melon cotyledons based on the transient expression rate of GUS gene.The result showed that melon cotyledons had lligh tolerance to Kanamyein with concentration of 150 mg/L, which was used to screen the cotyledon ofmelon after transformation.Addition of Ag+ into cocultivation medium was more beneficial for improving transformation frequency,however,acetosyringone (AS) was useless.The different disinfection methods should be taken according to different seasons for saving the melon seeds.

    Glyceraldehyde-3一phosphate dehydIogenase(GAPDH) is a glycoytic enzyme,which is active in all vertebrate tissues and frequently used as a housekeeping gene in expression studies.In this study,the mRNA sequence of glyceraldehyde-3-phosphate dehydrogenasein duck was analyzed by designing a pair of primers and amplified in eastern Zhejiang white geese (a native Chinese geese breed) as a control gene in real time PCR.The mRNA levels of the target gene were measured relatively to their respective templates and were normalized to reference genes,to control the amount of cDNA loaded
    into the reaction.The sequences of genomie DNA and cDNA of goose glyeeraldehydes-3·phophate dehydrogenase were 235bp and 209 bp,respectively.Real time PCR showed a specific Ct using cDNA as template,and the primers showed a single melt peak.So the gene expression study was reliable when glyeeraldehyde-3一phosphate dehydrogenase was used as all
    internal reference gene.

    To clarify the genetic mechanism of percentage of stigma exsertion in rice(Oryza sativa L)and improve stigma exsertion rate of a drought tolerant CMS maintainer Huhan 1B.QTL analysis was conducted in a F2 population of 190 plants derived from a cross between Huhan 1B and Ⅱ一32B.Ten QTLs of rice stigma exsertion rate were identified on four chromosomes.Three QTLs of percentage of single exserted stigma(PSES)were mapped on chromosomes 3,7 and 9 with accumulated contribution of 21.4%;Two QTLs of percentage of dual exserted stigma(PDES)were detected on chromosomes 3 and 9 with accumulated contribution of 15.98% ;Five of percentage of total exserted stigma(PES)were mapped on chromosome 3,4,7,9,including 2QTLs on chromosome 3,whose contribution rates varied from 6.86% to 9.73% and
    the total contribution rates of the five QTLs was 39.42%.Totally,seven QTLs pairs had significant epistatic effect on the percentage of stigma exsertion rate and accounted for 67.39% of the phenotypie variation.Most of them were additive×additive epistatic effects,and some of major QTLs were involved in the interactions.The identification of the QTLs markers may be important for improvement of Huhan 1B in stigma exsertion rate.

    The field experiment with different water treatments and management methods, including traditional flooded cuhivafion(C1),film mulched cultivation(C2)and uncovered cultivation(C3),was conducted to investigate the effects of
    cultivation model and nitrogen application on grain nutritional quality and phytic acid content for japonica rice,with 3 levels of nitrogen fertilizer(N1,0 kg/hm2;N2,124 kg/hm2;N3,150 kg/hm2) being designed for each wate rmanagement treatment.The results showed that C3 treatment(uncovered cultivation)had relatively higher total phosphorus content in rice grain,but much more decreasing phytic acid and in organic phosphorus contents compared with traditional flooded cultivation (C1).For the same nitrogen level,total grain nitrogen and protein components of C3 treatment were lower than those of C2and C3 treatments,implying that a reasonable fertilizer and water management in paddy production could improve grain nutritional quality,especially increase grain protein contents an d decrease grain phytie acids.Additionally,the increasing application of nitrogen fertilizer should cause the remarkably enhancing accumulation of total phosphorus in rice grains.

    The insecticidal effect of camptothecin against Nilaparvata Lugens,Brevicoryne Brassicae and Chilosuppressalis Walker was carried out under laberatory and field tests.A bioassay showed that 0.2% camptotbecin has strong contact toxicity,with LC50 and IC90 values of 0.1—0.6 mg/L and 0.4—5.0 mg/L,respectively.The descending order of activity was:Nilaparvatalugens> Brevicoryne brassicae > Chilo suppressalis.Field tests showed that the survival rate of B.brassieae and N.lugens were between 70% and 95%,which were not signiticantly different from the 10% imidacloprid WP control values of 63% and 98%.The survival and susceptibility of C.suppressalis were 85% and 88%,respectively,which were not significantly different from the 5% Regent SC control values of 93% and 95%, respectively.The results showed that camptotheein has strong insecticidal activity against above three agricultural pests.

    A 194 bp region of the porcine parvovirus VP2 gene was amplified by PCR and cloned into pGEM T vector,named as pGEM-VP2.Serial dilutions of plasmid pGEM-VP2 were used as standard templates for PCR to quantify the virus
    genomic copy number.We develoloped a SYBR Green I real-time PCR to detect porcine parvovirus.Sensitivity analysis showedthat the developed SYBR GreenI real-time PCR could detect 1.0×10 2 template/μL.The speeitleity assay exhibited that negative control and the other porcine pathogens,such as PRRSV,PCV,JEV,PRV,HCV and SIV could not be detected by this PCR.It was showed that10 positive results could be observed by the real-time PCR for 10 suspicious positive samples,but 7 positive results by normal PCR.The results suggested that as a result of the good sensitivity and specificity of the assay with a rapid and simple procedure,the real-time PCR method would be useful for the clinical diagnosis of porcine parvovirus infection.

    The authentic 3' end sequences of duck hepatitis virus(DHV)with four strains(ZYM,Z05,Z07 and Z10)were obtained by RACE.The sequence analysis showed that 3' end sequences of the four DHV strains contained 3D region, 3' untranslated region(3' UTR)and a poly(A) tail.The sequencing results showed that the amplified specific sequence was 597nt in length(not including polyA tail),and the sequences of 3' UTR were all 314 nt in length after the terminator codon TGA.The aligning analysis showed that the 3' end sequences of DHV shared high nucleotide acid identity among the four DHV strains rangingfrom 96.3% to 100% while the homology with referenced strain ranged from93.5% to 99.7%.Phylogenetic analysis of 3'end sequences indicated that these strains shared high nucleotide acid identity.

    The explants of Zantedeschia hybrida were disinfected by 0.1% HgCl2.The results showed that 12 min was the best sterilization time for annual tuber bud while 16min was the best steirlization time for biennial and three-year-old tuber
    bud.The best hormone combination was BA 2.0 mg/L+GA3 0.3 mg/L for MS medium,which could induce a higher proliferation multiple of adventitious buds of5.3.Meanwhile,the adventitious buds grew well.The suitable growing medium for plantlet rooting was 1/2 MS medium+ 0.5 mg/L IBA + 10 g/L sucrose.It was benefit to increase the transplantation survival rate when the plantlets were treated with 3 d outdoor acclimation.When transplanted after 80 d,the survival rate of plantlets was 88%.In this experiment,March 10 was the best transplanting date,on which the survival rate of plantlets achieved 100%.

    Effects of different temperatures on activities of three digestive enzymes in Cambarus clarkii were examined.The results showed that the optimum temperatures for amylase in hepatopancreas,intestine and stomach were all 40℃ ;while the optimum temperatures for pepsin were all 35℃.In addition,the optimum temperatures for lipase in hepatopancreas,intestine and stomach were 25,35,25℃ ,respectively,and the optimum temperatures for trypase were 55,65,55℃ ,respeclively.At the oplimum reactive temperature,the activity order of amylase in different organs was stomach > hepatopancreas
    > intestine,meanwhile ,the activity orders of trypase and pepsin were hepatopancreas> stomach > intestine.The activity of lipase in intestine was similar to in hepatopancreas,which was higher than in stomach.

    Effects of drought stress on the chlorophyll fluorescence parameters and photosynthesis rate in leaves of three asparagus bean [Vigna unguiculata ssp.sesquipedalis(L.)Verd.] cultivars with different tolerance to drought stress were investigated.The results showed that the biomass,net photosynthetic rate(Pn)of seedling greatly decreased,and the plasmalemma permeability of seedling leaves significantly increased drought stress.At the same time,the chlorophyll fluorescence parameter(Fo,Fv/Fm,φPSⅡ and qP)were also affected by drought stress,which showed a correlation with drought tolerance.The chlorophyll fluorescence parameters can be used as convenient indexes for evaluation of the drought tolerance of asparagus bean cultivars.

    The effect of phosphorus on root antioxidant enzymes activities and the number and activity of border cells was investigated in buckwheat seedlings under aluminum stress.It was found that addition of 3.0 mmol/L P increased the relative root length and the protein content of root by 22.8% and 18.7% compared with the treatment with Al(200 μmol/L )alone,meanwhile,the activities of superoxide dismutase (SOD) and peroxidase(POD) increased by 20.8%and 26.1%, respectively.Treatment with 1.5 mmol/L P in the presence of Al induced an increase in the border cell number and viability to 243.0% and 187.0% compared with the Al trealment,respectively.It indicated that the alleviation of Al toxicity in buckwheat seedlings with P application could be associated with the enhancement of the Al-resistant ability of the root because of increased activities of the anfioxidant enzymes and number and viability of border cells.

    Effects of three plant growth regulators(S0D ,Cc and DTA-6)by foliage spray on the photosynthetic characteristics in soybean (Kennong 4)were studied in a field experiment.The results showed that the photosynthetic rate and
    transpiration rate were enhanced from the 5th day to the 30th day after spraying with DTA一6, thus the yield of soybean was markedly increased.However,spraying with SODM could alleviate the decrease of total chlorophyll content.Meanwhile,there was no direct correlation between the water use efficiency and stomatal conductance(Gs)in different treatments and the control.The time-course changes of Gs showed a double-trend performance in soybean ,the first peak appeared on the l0th day,and the second one on the 20th day.On the 30th clay after spraying,the changes of the water use efficiency and Gs with different treatments showed a similar trend, which was SODM>Cc> DTA一6 >CK.The above results indicated that foliage spraying of DTA一6 and SODM could regulate the photosynthetic characteristics and increase the yield in soybean.

    Spinach were packaged with diferent thickness of polyethylene films(0.010,0.025,0.040 and 0.025 mm +anti-condemafion component,with no film packaging as contro1)and stored at(2±0.5)℃.The physiological and biochemical changes in spinach during low temperature storage were studied.The results indicated that spinach packaging with 0.025 mm+ anti—condensation component film could maintain the best quality.It not only reduced the respiration rate,MDA content,and cell membrane permeability,but also kept the level of chlorophyll content and protein content in spinach,which resulting in prolong of the storage period and shelflife.However,there was no clear difference in water loss rate in spinach among packaging with different thickness of films.

    Tobacco seeds of MS Yunyan 87 and MS K326 were primed with gibberellin(GA3),then coated into pelleted seeds.The effects of priming temperature,priming time and GA3 concentration on the vigor of pelleted seed and seedling
    quality were investigated when pelleted seeds were germinated under 25℃ and 10℃ .The results showed that when pelleted seeds were germinated under 25℃, the priming treatment of 100 mg/L GA3 under 25℃ and 20℃ for 36 hours were better than other treatments.The gemination index were increased by 18.58% and 27.91%, vigor index were increased by 30.58% and 29.98%,average germination time was shortened by 0.69 d and 0.85 d,respectively.The seedling length of MS Yunyan 87 was increased by 0.23 cm, seedling dry weight of MSK326 was increased by 0.29 g as compared to the contro1.When pelleted seeds were germinated under 10℃, the treatment with 100 mg/L GA3 under 25℃ for 24 hours was better for MS Yunyan 87 than others, whose germination index was increased by 18.05% and germination time was shortened by 1.84 d.Meanwhile,the treatment with 50 mg/L GA3 20℃ for 24 hours was better for MSK326,whose germination index was increased by 14.58%, germination time was shortened by 1.55 d.The results indicated that suitable GA3 priming treatment could enhance the pelleted seed vigor and seedling quality of tobacco,and enhance the chilling resistance
    of pelleted seeds.

    Aspergillus niger MA was selected as the initial strain for β-mannanase production from a number of strains.After Co 60 mutagenesis of A.niger MA,a genetic stable and β-mannanase overproducing mutant A.niger MA-56 was obtained through primary and secondary screening.The activity of β-mannanase was improved about 64.78% and reached 9.31× 10 4 U/g.The results of enzyme characteristics of β-mannanase showed that the optimum temperature was 70℃ and the optimum pH was 2.5—3.5.Moreover,β-mannanase was also stable at pH 2.5—9.0 at 60℃ .Among different metal
    ions,Cu2+ showed stronger inhibitory effect on the activity of β-mannanase.

    The changes of antioxidant compound contents and antioxidant activities in strawberry during diferent harvest time were studied in the cultivar ‘Zhangji’ and cultivar ‘0362’.The results showed that the contents of total flavonoids,
    total phenolics and assay of DPPH,TEAC,FRAP in ‘Zhangji’ and ‘0362’ fruis significantly increased during the harvest time,which were significantly different between these two cultivars.The significantly positive linear relationships were found between the total phenolics content and antioxidant activities (correlation coefficients for DPPH,TEAC and FRAP were0.8228,0.8812 and 0.8419).Therefore,the changes of antioxidant activities in strawberry fruit during diferent harvest time may be attributed to the content of phenolic compounds.

    A recognition technique of mature pomegranates in natural light conditions was studied.Images of mature pomerganates were enhanced by homomorphic filtering and converted from RGB space to HSI space, then the components of
    hue and saturation were fused to remove the dark backgrounds.Afterwards.images were segmented by the regional areas and morphologic characteristics,then centroids and radius were extracted by Hough transform,and the shape of occlusion fruits was recovered.Finally,image processing program was developed for the recognition of mature pomegranates based on VC++6.0.The results showed that the correct recognition rate is>85%,supplying a good foundation for the implementation in the autonomous harvesting on fruit picking in the future.

    Four tomato rootstock varieties( ZJ-1,ZJ-3,ZJ-7 and ZJ-9)were selected as rootstock and Zheza203 was selected as scion to investigate the graft compatibility and the resistance to tomato bacterial wilt.The results showed that ZJ-1 and ZJ-9 were the best rootstock for disease-resistance,which had higher graft compatibility with Zheza203 and whose grafting survival rates were more than 99%.After the inoculation of three Ralstonia solanacearum isolates respectively, the grafting survival rates were 97 .4% and 97 .8% when Zheza203 was grafted with the stocks of ZJ-1 and ZJ-9,while only 75.0% of
    Zheza203 could survive without grafting,showing significant difference.After grafted with ZJ-1 and ZJ-9,the disease plant rates of Zheza203 were lower than 5%,significantly lower than 21.4% of the control Zheza203 without grafting.Meanwhile,the yields of the grated combinations(ZJ1-203 and ZJ9—203)were increased by 92.2% and 101.7% respectively,compared with Zheza203 without grafting.In addition, the grafting seedlings of ZJ1—203 and ZJ9—203 had strong growth and had no effect on the fruit quality.

    Pot experiments with rice were conducted to investigate the nitrogen(N)and potassium (K)distributions in different leaf positions and growth stages.Linear correlation analysis between N,K contents in different leaves,total N,K
    content in leaves and the amount of N,K application wore also studied.The results showed that N content increased from upper leaf position to lower position in jointing stage,while the reVerse was obvious in booting stage.K content decreased from upper leaf positiontolower positionin both jointing and booting stages.The differences of N content among leaf positions were investigated following the increasing amount of N application.However,no significant change was observed in K content among different leaf positions when the amount of N appfication was increased.According to the correlation analysis,the first position of rice leaf was suitable for N diagnosis in jointing stage,and the third or the fourth position of rice leaf was suitable for N diagnosis in booting stage.The first position of rice leaf was suitable for K diagnosis in both jointing and booting stages.

    Optimization of conditions in solid-fermentation of rapeseed meal using response surface methodology

    The solid-state fermentation by microbes was optimized by using response surface analysis(RSA)with JMP staTIStical software.A polynomial regression model including the four major factors(feed to water ratio,cottonseed meal usage,fermentation duration,temperature) was set up.The results showed that the optimal fermentation condition was as follows:cottonseed meal 10.1% ,temperature 30.1℃ ,feed to water ratio at 1:0.8,fermentation for 46 .9 h,in which the crude protein content was increased to 41.7%, exceeding 15.8% than before.

    The genetic diversity of six C.wenyujin populations were analyzed by ISSR.Eleven primers were screened on the template of total DNA of six C.wenyujin populations.A total of 82 bands were amplified,26 of which,31.7% of the total bands were polymorphic bands.Genetic similarity coefficient was obtained by NTSYS-pc(Version 2.10e)software.Nei's genetic identity and genetic distance for six C.wenyujin populations were obtained by POPGENE 1.32 software,and the dendrogram was constructed by MEGA 3.1 software.The results suggested that the genetic divemity of C.wenyujin from these six different geographical populations was low.

    Coat protein gene of Ornithogalum mosaic virus(OrMV)was amplified from a virus infected Narcissus bulbocodium sample from UK by RT-PCR.The coat protein of OrMV was prokatyofic expressed,purified and used in
    preparation of antisenun againstthe virus.OrMV then was detected in local and imported narcissussamples using this antiserum by an indirect ELISA method, and serological relationship between OrMV and other narcissus viruses were discussed.

    An empirical study on contribution rate of agricultural technological progress in Zhejiang Province

    Based on the panel data of eleven cities in Zhejiang province from 1990 to 2005,a model was established for calculation of agricultural technological progress by applying the Translog Production Function,taking account of both partial technological progress and neutral technological progress.The results demonstrated that agriculture development has been in all expanding reproduction stage mainly relying on technology progress but not material extension;the improvement of the quality of labor and land and the development of the market-oriented system of agriculture are the keys to increase the whole agriculture technological progress in Zhejiang province.

    Fuzzy clustering algorithm is widely applied in weather service,geology,pattern recognition and data mining.In this paper,fuzzy clustering was applied in core collection of crop and the traditional fluzzy clustering algorithm was improved.Based on similarity coefficient and distance coefficient, similarity degree has been presented which could describe not only the degree of value similarity,but also the degree of shape similarity.Replaced conventional similarity matrix by similarity degree matrix,the fuzzy clustering algorithm was effective.
