    The effects of different agents on seed germination and growth characteristics of tobacco varieties with different drought tolerance were studied in this paper Seeds of two tobacco varieties with different drought tolerance were primed in different concentration solutions of paclobutrazol, calcium chloride, proline and salicylic acid Then, the germination, seedling growth and physiological changes under drought stress (simulating with 15% polyethylene glycol 6000) were measured The results showed that the germination and seedling growth were inhibited, however, the activities of ascorbate peroxidase (APX), catalase (CAT), peroxidase (POD) and superoxide dismutase (SOD) in tobacco seedlings were enhanced and the malondialdehyde (MDA) concentration were decreased after treated with paclobutrazol (PP333) The germination parameters, root length, seedling length, seedling fresh and dry weight were improved, the activities of APX, CAT, POD and SOD were enhanced and the MDA concentration were decreased in two tobacco varieties after treated with calcium chloride, proline and salicylic acid The treatment of 80 mg·L-1 salicylic acid had the best effects among all treatments, suggesting that this treatment can be used to improve drought tolerance of tobacco seeds and seedlings.

    Eight different genotype female parent materials were induced by three haploid inducers (Stock 6, Gaoyou 1 and MT041) to test their ability of producing haploid through grain color, chromosome number of root tip, stomatal density and field planting performance, and colchicine was used for doubling. The results showed that haploid induction rates of different inducers differed greatly. The haploid induction rate of MT041 was 4.17%, which was higher than the other two inducers. The female genotypes showed significant difference in induction rates. The haploid induction rates of the waxy and ordinary materials were significantly higher than sweet materials. The sweetwaxy corn hybrids were beneficial to increase the sweet corn haploids. The doubling effect of 0.06% colchicine solution was significantly higher than 0.02% colchicine solution by rootsoaking, while seedling percentage in high concentration colchicine treatment was not high. Chromosome doubling is still a bottleneck restricting widespread application of the DH breeding techniques.

    Application of Mutation Surveyor in analyzing heterozygous InDel

    The present study aimed to show the application of Mutation Surveyor in analyzing nucleotide substitutions and InDel with an instance. The polymorphisms in ISG15 5′ flanking sequences were analyzed for 144 Hu sheep from Yuhang Hu sheep farm and slaughterhouse by polymerase chain reaction\|single strand conformation polymorphism (PCR\|SSCP) technique, DNA sequencing and Mutation Surveyor software. PCR\|SSCP analysis showed ISG15 5′ flanking sequences bands were too complex to determine genotype directly. The analysis result of Mutation Surveyor found there were three polymorphisms, 152 C/T, 158 dupC and 239 C/T, in ISG15 5 ′flanking region. The frequency of allele C in loci 152 was 083 showing low polymorphism (PIC = 0.2424). The frequency of allele dupC in loci 158 was 05125 showing mediate polymorphism (PIC = 0.3748). The frequency of allele C in loci 239 was 07675 showing mediate polymorphism (PIC = 0.2932). Except 239 C/T, the gene frequencies and genotype frequencies of Hu sheep 152 C/T and 158 dupC tested have reached the Hardy\|Weinberg equilibrium (P>0.05). Mutation Surveyor software has greater advantage in analysis of nucleotide substitutions, especially InDel sequencing data. So Mutation Surveyor software can be used for SNPs high\|throughput screening and analysis.

    Effect of different rooting agents on cutting propagation of Huiming baicha

    In order to screen out suitable kinds and concentrations of rooting agents, six different rooting agents were used to investigate their effects on rooting and growth of cutting propagation in Huiming baicha. The results showed that all rooting agents had significant effect under suitable concentration, especially the treatment of 800 mg·L-1 ABT1 solution had the best effect on the index of wound healing rate, nodulation rate, rooting rate, survival rate, number of roots, average length and weight of root for cutting seedlings, and the survival rate was 89.5%. The new shoot grew best and the length was 25.3 cm in the treatment of mixed solution of 400 mg·L-1 IBA + NAA (2:1), which had better effect than the single treatment of IBA or NAA.

    The PCRSSCP was used to study the associations between immune traits(sheep red blood cell, avian influenza and newcastle disease)and polymorphic loci of the Mx gene in Shandong indigenous chicken populations(Luqin chicken, Shi qiza chicken, Langya chicken, Laiwu black chicken and Jining Bairi chicken). The fragment of Mx gene about 150 bp including intron 13 and exon 14 was amplified. The results showed that two single nucleotide polymorphisms were detected in the five indigenous chicken populations by PCRSSCP: 1 892 points of coding sequence of Mx gene G→A (Ser→Asn); 1 911 points G→A, which couldnt lead to amino acid mutation. Through the analysis on the association between the polymorphic loci of the exon 14 and the antibody response, the SNPs on the 1 892 points of coding sequence of Mx gene in Luqin chicken, Langya chicken and Laiwu black chicken was correlated to antibody response of H9 significantly(P<0.05).

    Our current project was conducted by using BMPRIB, BMP15 and GDF9 as candidate genes to study the relationship among polymorphisms of BMPRIB, BMP15 and GDF9 and the fecundity in sheep and goat. PCRRFLP was applied to analyze the relationship between the gene and genotype polymorphisms of BMPRIB, BMP15 and GDF9 in four Fujian goat breeds and their prolific trait. The results showed that there was no same mutation in BMPRIB gene of four goat breeds as that of Booroola Merino. The FecXI, FecXH, FecXB of BMP15 and FecGH of GDF9 were not found. The possibility that the fecundity of four goat breeds were affected by the mutations of BMPRIB, BMP15 and GDF9 gene could be ruled out. As we dont know the whole gene sequence of the three genes in four goat breeds, its hard to determine whether these genes affect the fecundity of them.

    This study was conducted to study the effect of saccharicterpenin on the quality of boar sperm in high temperature season (from July to September, 2011) in Zhejiang province, adding 400 g·t-1 saccharicterpenin in a diet of boar The result indicated that there was an increase in semen volume, density, motility and pH value, but no significant difference (P>0.05) Meanwhile, there was significant increase (P<0.05) in testosterone level In summary, saccharicterpenin can increase the quality of boar sperm in high temperature season.

    To get a better understanding of aroma compounds derived from tea, extracts from Tieguanyin tea were obtained by the methods of water extraction, ethanol extraction, supercritical carbon dioxide extraction, and subcritical extraction, respectively, of which the volatile and semi\|volatile compounds, especially aroma compounds, were determined by the combination of gas chromatography and mass spectrum method (GC\|MS). Additionally, performances of the various extracts on cigarette quality were investigated when the extracts were applied in cigarette filters as well. As the results indicated, compared with the other three methods, supercritical carbon dioxide extraction showed a better contribution to aroma compounds extraction from Tieguanyin tea in this research. According to the assessment of cigarette quality taste, extracts from supercritical carbon dioxide extraction method also had a better significant influence on it. It is concluded that supercritical carbon dioxide extraction is a relatively appropriate method, with high efficiency and economic effectiveness, for preparing cigarette quality-oriented extract.

    The Lycoris bulbs were treated with eight division method of cutting. Effects of different concentrations of nitric oxide (NO) and GGR6 powder on cormel development in five native species of Lycoris were investigated. The results were as follows: The cormel number, cormel diameter, cormel height, cormel fresh weight, root number and root length of the five genus Lycoris were different, meanwhile each index showed different response to experimental treatments. The cormel formation number of L.chinensis and L. rosea was promoted by relatively higher concentration of NO, whose optimal concentration range was 50-75 μmol·L-1. Relatively higher concentration of GGR6 powder was good for cormel development of L. sprengeri and L. albiflora, whose optimal concentration range was 50-75 mg·L-1. The cormel quality of L. radiate was effectively increased by 25 μmol·L-1 NO according to the index of cormel diameter, cormel height and cormel fresh weight.

    Pyrus pyrifolia Nakai. cv. Eli No.2 was used as material to study the effect of different types of bagging on fruit quality. The results showed that bagging could significantly improve the appearance of fruits with smaller and less fruit dots, and decrease rusty stain index. There was significant linear relation between transparency of bags(x) and the size of fruits(y) with the regression equation of YW=159.61+0.99x. Regression equation and correlation coefficients showed as: brightness (L), YL=74.45-0.71x; colority coordinate value (A), YA=-16.68-0.77x; colority coordinate value (B), YB=52.52+1.86x. The Vc content was higher than that of the control. The SSC/TA and SS/TA of the fruits in bagging treatment increased compared to the controls. According to the results, it was concluded that the most appropriate paper bag types for P.pyrifolia Nakai. cv. Eli 2 were twolayer bags from Xiaolin, South korea, Taiguo and twice bagging was commendatory in the manufacturing of P.pyrifolia Nakai cv. Eli No.2.

    Study on sucrose accumulation and enzyme activities involved in sucrose metabolism in developing seeds of vegetable soybean

    Three cultivars, Zhenong No.6 (high sucrose content), Liaoxian No.1 (middle sucrose content) and Xiafeng 2008 (low sucrose content) were used to investigate the sucrose accumulation and enzyme activities involved in its metabolism in developing seeds of vegetable soybean. The results showed that the growth curves of vegetable soybean seeds showed single sigmoid pattern. The change trends of sucrose metabolism among three cultivars were similar. During the whole periods of vegetable soybean seeds development, sucrose was the major component of soluble sugar, taking about 70% of total soluble sugar contents. The net activities of sucrose\|metabolizing enzymes were the major factor affecting sucrose accumulation. During the early stage of seed development, net activities of sucrose\|metabolizing enzymes were negative value. Sucrose metabolism is in the decomposition direction, when sucrose synthase and acid invertase were key enzymes in sucrose decomposition. However, the net activities of sucrose\|metabolizing enzymes turned to positive value in the middle stage of seed development. Sucrose metabolism was in the synthesis direction, when sucrose synthase and sucrose phosphate synthase were key enzymes in sucrose synthesis. During the late stage of seed development, also the net activities of sucrose\|metabolizing enzymes were positive value, but its value was close to zero, showing that the rate of sucrose decomposition and synthesis was similar and sucrose synthase, sucrose phosphate synthase, acid invertase and neutral invertase played a leading role in sucrose metabolism in this stage.

    The dynamics of fresh weight, color parameter, sugar, organic acid, and total phenolics and flavonoids contents were examined in three white fleshy loquat (Eriobotrya japonica (Thunb.)Lindl) resources The results showed the change trends in fruit weight and color development were similar among four white fleshy loquats The biggest fruit size was Ninghaibai among the four varieties, the size of Big Taipingbai was close to Ninghaibai, Taipingbai was the smallest one The change trends of sugars, organic acids, total phenolics and flavonoids contents were also similar The fructose was the major sugar of these four white fleshy loquat varieties, which accounted for 54%-61% of total sugar content The glucose took the second content component of sugars, which accounted for 35%-37% of total sugar content The highest sugar content was in fruit of Taxiabai, Taipinbai ranked the second, and the lowest sugar content was in fruit of Big Taipinbai The titratable acids and organic acids contents were the highest in fruit of Taxiabai, the content in Taipinbai was the second, Big Taipingbai was the lowest The contents of total phenolics and flavonoids were relatively high in the early stage of fruit development, thereafter quickly declined with fruit development, kept constant in final two weeks before maturation The difference in total phenolics and flavonoids content among four varieties was little The key stage of fruit quality components accumulation began from three weeks before maturation, when the loquat fruit grew quickly, sugars accumulated fast, and organic acid declined rapidly.

    AFLP analysis of genetic diversity of Angelica dahurica

    The 50 germplasmic resources of Angelica dahurica were analyzed by AFLP to determine the genetic diversity. The amplified fragment length polymorphism (AFLP) tags were used to investigate the genetic diversity at the DNA level. Twelve pairs of selective amplification primers were screened out to amplify the bands, which were used as original matrix and analyzed the similarity between different Angelica dahurica germplasm by NTSYS\|PC software calculation and constructed the phylogenetic tree. The results indicated that the similarity of the 50 Angelica dahurica germplasm resources all more than 70%, whose overall phylogenetic relationships were relatively close. The germplasm ZYS002039 and ZYS002045 showed larger difference in genetic background from the others.

    The hybridization experiment was carried out using Rosa primula, Rosa xanthina f. spontanea, Rosa chinensis cv.     According to the flower characters,the cultivars of Chimonanthus praecox were investigated and classified in central and eastern Henan, China. The results showed that 156 cultivars in Henan province were divided into three groups according to the purple speckles of inner tepals including Ch.praecox Concolor Group, Ch.praecox Intermedius Group and Ch.praecox Patens Group and identification key of the cultivars has been compiled.

    Comparison of the RNA isolation methods for Pleurotus ferulae Lenzi hyphostroma

    Total RNA of Pleurotus ferulae Lenzi was extracted by single-step method, STE method and guanidinium isothiocyanate method, whose isolation effects were compared. The results showed that it was difficult to extract total RNA from Pleurotus ferulae Lenzi by single\|step method. Total RNA could be extracted by guanidinium isothiocyanate method, but RNA extracted was immingled by some protein and was not enough for molecular biology experiment. Single\|step method and guanidiniu isothiocyanate method were not suitable for RNA extraction from Pleurotus ferulae Lenzi because of its high amylase content. Total RNA extracted by STE method had high quality, which is the best method of total RNA isolation from Pleurotus ferulae Lenzi. The quality of total RNA was analyzed with agarose gel electrophoresis and nucleic acid and ultraviolet spectrophotometer. Three bright bands were observed in agarose gel electrophoresis as 28S, 18S and 5S. The band of 28S was twice wider than that of 18S, and the value of OD260/OD280 was 18 to 20.These results demonstrated that the quality and purity of RNA obtained by the method of STE could meet the demands of molecular biology experiment.

    For improving forecasting and sustained control damage level, light trap numbers and field dynamic of underground pests (black cutworm, mole cricket and scarab beetle) in Hangzhou district during the years of 2005 to 2011 were investigated The results showed that different pests had obvious differences in population dynamic The black cutworm, Agrotis ypsilon, had several damage peaks (late May, late June and late July etc), and the moth amount of the prophase was relatively high The mole cricket, Gryllotalpa africana, had two damage peaks (late May to early July, early September to middlelate October) The scarab beetle, Anomala corpulenta, had one damage peak (late May to late June) There were periodic changes in total numbers of underground insect pests among years, and the peak period was the year of 2005, 2007 to 2009 and 2011, respectively On this basis, temperature, humidity, precipitation, illumination etc were used as forecasting factors, using the method of stepwise regression, nineteen factors with significant correlation were screened, and occurrence amount forecasting models and occurrence period forecasting models of the three insects were established By using accuracy degree judge model for verification, the score values of occurrence amount and occurrence period forecasting models of the three underground pests were more than 58 and 70, which indicated that the historical coincident rate and prediction accuracy of established forecasting models were good.

    One species of insect nematode (Caenorhabditis sp CNM), which can cause Galleria mellonella death within 4 days, was discovered in the compost of Agaricus bisporus through the Galleria bait method In order to discover the biologicalcontrol potential of CNM in the field application, we investigated its biology characteristics, especially the infectivity and antistress resistance The result of its infectivity to G mellonella showed that the larger of the dosage, the better of its infectivity LD50 of CNM to G mellonella was 46.6 IJs per larva, and the LC50 is 53.8 IJs·cm-2 at 48 h After 45 min, 20 h and 55 h, all of the nematodes died with water bathing of 42℃, 40℃ and 38℃ Therere still 46.0% of the nematodes alive after 12 h under 25℃, 50.5 % RH, and the infectivity decrease was 43.3% Besides, there were re 57.1% of nematode that can move for as far as 3.6 cm under 30℃ So Caenorhabditis sp CNM strain has the biocontrol potential with good infectivity, heat resistance and desiccation.

    Comparison of the modified metagenomic DNA extraction methods of microorganisms from salt lake soils

    In order to construct the metagenomic library of microorganisms from salt lake soils in Yuncheng, the microbial metagenomic DNA extraction method was explored. Six of plant genomic DNA extraction methods were modified to extract the microbial metagenomic DNA. The yield and quality of the metagenomic DNA was detected by ultraviolet spectrophotometry, gel electrophoresis, restriction endonuclease analysis and PCR amplification, and the results showed that the modified method 3 was suitable for extracting the microbial metagenomic DNA from salt lake soils in Yuncheng. For the microbial metagenomic DNA extracted by the modified method 3, the value of OD260/280 was 1.883, the concentration and yield was 18.1 ng·μL-1 and 72.4 ng·g-1 respectively, and the length was about 23 kb with a clear stripe on the agarose gel. Moreover, the metagenomic DNA was completely digested with EcoRⅠand also could be used for PCR amplification.

    The degradation of hexachloroethane (HCE) in the paddy soil and the effect of sulfate on sulfate\|reducing bacteria were investigated through simulating different environmental conditions. Results showed that the degradation rate of HCE increased with the increasing initial HCE concentration in the soil, However, the degradation rate of HCE decreased when it reached a certain quality concentration (≥60 mg·kg-1). The degradation activity of sulfate\|reducing bacteria (SRB) was enhanced and the HCE degradation rate was speeded up with the increasing soil sulfate. But the growth of sulfate\|reducing bacteria and the degradation of HCE were restrained when sulfate was excessive(≥60 mg·kg-1). The soil pH and Eh values were optimal for the growth of sulfate\|reducing bacteria after 21 days with addition of sulfate and HCE, and then the rate of sulfate degration also reached the maximum. The result showed the degradation of HCE was accelerated through controlling the soil pH and Eh values or adding proper concentration of sulfate to soil in order to reduce the risk of HCE in soil.

    With the aid of the 3S technology, the land use change data during 1970s and 2005 in Mongolian Plateau were obtained. The spatial and temporal characteristics of the Mongolian Plateau land use change in the past 30 years was revealed by analyzing the changing process. The results showed that the total area of grassland was significantly reduced, a large area of bare land increased, urban and rural construction land expanded significantly, the area of cultivated land, forest land and water increased in Mongolian Plateau in recent 30 years. Grassland degradation and desertification became the significant spatial and temporal characteristics of landuse change in Mongolian Plateau, and the situation of Inner Mongolia was more serious than Mongolia. The forestry ecological engineering in Inner Mongolia played a positive role in protecting the ecological environment. Population, policy, economic and climate change were the main driving forces of the land use dynamic change in Mongolian Plateau.

    Bayberry Myrica rubra sieb. et Zucc. cv. Lanxi Muye fruits were used as raw material to study the processing of red bayberry wine fermented by whole fruit. Orthogonal test was used to determine the optimal fermentation conditions and the results indicated that fermentation temperature was 24℃, yeast was added in 015% of the fruit weight, sugar addition amount was 14%, and natural pigment was added with 03%. Quality was also evaluated instrumentally for the whole fruit fermented wine. The physical and chemical items determined of the obtained wine are excellent and follow the quality standard of red bayberry fermented wine. The wine has a good smell of bayberry fruit and typical fermented wine with its color in dark red, appearance in clear and transparent, taste in soft and refreshing and flavor in unique. The product contains 14 kinds of amino acids with the total amounts reaching 830 mg·L-1. Seventeen kinds of main aroma components were detected in the four\|month aging wine, which included 4 kinds of esters, accounting for 29.78% of the ingredients identified, 9 kinds of alcohols, representing 47.51% of the total components and the other 4 flavor substances such as ketone, furan,etc.

    The fatty acid composition of the common edible vegetable oil was analyzed,and the significant changed characteristic components in adulterated camellia oil was also determined. By setting different proportions of adulterating contents and using the method of gas chromatography\|mass spectrometry (GC\|MS),the regression equation was established between the characteristic components contents after adulterated of palm oil, rapeseed oil, cotton seed oil, peanut oil and soybean oil in camellia oil and the adulterated amount. The correlation coefficient was greater than 099,and the overall accuracy was 101%. This method can be used for the qualitative identification and quantitative detection of adulterated camellia oil.
