    Study on preparation and immune efftect of PEG conjugate vaccine of ultrasonic inactived Aeromonas hydrophila

    PEG conjugate vaccine of Aeromonas hydrophila was made by the methods of transient high\|temperature, high press and shearing force of ultrasonic, using PEG as ultrasonic medium and vaccine vector. The PEG conjugate vaccine and the traditional vaccine were injected Cryprinuscarpiod intraperitioneally respectively.The percentage of phagocytosis, the index of phagocytosis, antibacterial activity, antibody titer and immune protective efficiency were investigated. The results showed that all the testing indexes of PEG conjugate vaccine were significantly higher than other vaccines. The immune protective efficiency was 80% for PEG conjugate vaccine, which was the highest and 267% higher than that of traditional vaccine, indicating that PEG6000 was a good binder of vaccine preparation for Cryprinuscarpiod against Aeromonas hydrophila

    Degenerate primer pairs were designed according to known anthocyanidin synthase gene sequences in NCBI database. The gene, designated BaANS, was isolated from leaf genomic DNA and cDNA of Chinese kale (Brassica albograbra), respectively. Genomic DNA of BaANS gene is 1 369 bp in length with one intro of 292 bp. Complete coding sequence of BaANS is 1 077 bp in length encoding 358 amino acids. The sequence has been submitted to NCBI with accession number of GU170203. Sequence comparison with Matthiola incana, B. oleracea var. capitata, B. juncea, B. rapa ssp. campestris and Arabidopsis thaliana showed that they shared high homology.

    A recombinant baculovirus expressing VP60 of Rabbit hemorrhagic disease virus (RHDV) was constructed with bac\|to\|bac baculovirus expression system to study the expression and intracellular localization of the structural protein VP60 of RHDV in Sf9 cells. The results of indirect immnuofluorescence assay (IFA) and Western blot analysis showed that the proteins were successfully expressed in Sf9 cells. The results of electron micrographs showed that there were some virus\|like particles (VLPs) formed in Sf9 cells, and further results of confocal microscopy indicated that VP60 proteins were localized at the cellular membrane of Sf9 cells.

    Alpha-toxin gene was amplified from a field strain of Clostridium perfringens by polymerase chain reaction (PCR), PCR product was cleaved with restriction endonucleases EcoR Ⅰ and Nco Ⅰ, and inserted into the EcoR Ⅰ/Nco Ⅰ site of pET\|28a(+)vector. Recombinant pET-28a-CPA plasmid was transformed into BL21 (DE3)cells. The obtained plasmids were studied in detail by PCR, restriction endonuclease analysis and nucleotide sequencing. The recombinant plasmid was transformed into E.coli BL21(DE3)and expressed under the induction of IPTG. Western blot analysis indicated that polyclonal goat-anti-serum against alpha\|toxin had specific reaction with recombinant protein. These results indicated that the target protein had a good immunogenicity.

    Using sterile tomato cotyledons as explants and kanamycin-resistant NPTⅡ as a marker gene, the ThaumatinⅡ gene which encodes a plant sweet protein from Thaumatococcus daniellii has been introduced into the tomato genome by Agrobacterium-mediated transformation. As a result, 10 independent tomato transgenic lines were acquired. The introduction of the foreign gene was confirmed by PCR for the expected specific bands. Furthermore, one transgenic line was shown by two copies of integration on Southern blotting analysis. Taken together, all the above results have indicated that the ThaumatinⅡ gene has integrated into the genome of tomato. This represents the first step to finally obtain marker-free transgenic tomatoes expressing plant sweet protein ThaumatinⅡ.

    Effect of commercial substrates on stock and grafted seedling growth in watermelon

    In this experiment, the effects of four substrates on growth of stock (Lagenaria leucantha Rusby.,L. siceraria Standl.)and watermelon grafted seedling. It was found that the stock seedling was strong with low hypocotyl length, and the leaf growth rate of graft seedling was high when cultivated in ‘Golden 3’ and ‘Golden 1’ substrates, which were ideal substrates. The graft seedling showed low disease rate and high survival rate when cultured in ‘Zhongci 1’ substrate, but it was difficult to manage the water. The seed germination rate of stock in ‘Fafard’ substrate was higher than other three ones, but it was necessary to add fertilizer in middle and late growth stage for the lack of nutrition.

    Effect of NaCl stress on growth and photosynthetic characteristics of eggplant seedlings

    Effects of different concentrations of NaCl on plant growth, photosynthetic characteristics, contents of proline and soluble sugar of eggplant (Solanum melongena L.)seedlings of ‘Huqie 08\|11’ were studied. The results showed that with increasing NaCl concentration, the plant height, stem diameter, shoot fresh weight, root fresh weight and contents of chlorophyll decreased, and net photosynthetic rate (Pn), stomatal conductance (Gs), intercellular CO2 concentration (Ci)and transpiration rate (Tr)decreased significantly; meanwhile, stomatal limitation value (Ls)increased obviously, water use efficiency (WUE)declined after an initial increase, and contents of proline and soluble sugar increased significantly. It was indicated that the growth and photosynthesis of eggplant were inhibited under NaCl stress, and the declined photosynthetic rate was due to stomatal limitation.

    Analysis of damage characteristics of tea longhorn in organic tea garden in Cangnan County of Zhejiang Province

    Cangnan County is one of important tea\|producing areas in Zhejiang Province. The investigation for the damage characteristics of tea longhorn beetle (Aeolesthes induta Newman) was carried out in Wufeng organic tea garden in Cangnan County. The results showed that the damage rate of tea longhorn beetle was about 6.7%-50.0%. Moreover, the damage characteristics of tea longhorn beetle were as follows: the damage occurred near hilltop (altitude 450 m)was more serious than that on the halfway hill (altitude 350 m);the damage occurred in the north side of road was more serious than that in the south side; the damage occurred at about 1 m away from roadside was more serious than that at 100 m away from the roadside. In all, the damage degree of tea longhorn beetle might be influenced by the depth of soil layer, root exposure, trapping light and so on.

    According to the pathological anatomical observation to the grains infected by Ustilaginoidea virens from the field, there were the integrated floral organ including stamen and pistil, which were isolated from the infected grains without forming yellow chlamydospore. The integrated ovaries, which were green and not obviously enlarged because of grain filling, were isolated from the typical infected grains with lots of yellow chlamydospore. Some male\|sterile rice lines were infected by this fungus and resulted in the typical symptoms. It was proposed that the fungus Ustilaginoidea virens infected the pistil before the flowering and absorbed nutrition from the ovary to develop the smut ball.

    The primary core collection of Chinese rice germplasm (including 203 materials) were used to evaluate their resistance to rice blast and bacterial blight. The materials with resistance to leaf blast and neck blast were 41.38% and 78.82%, respectively. Nineteen varieties(9.45%) and nine varieties(4.48%) showed resistance to 80%-90% and 70%-80% races of Magnaporthe grisea, respectively, and 6468% of the tested varieties exhibited resistance to 20%-30% races of M. grisea. There were two(0.98%) and six(2.98%)varieties exhibited resistance to 75% and 62.5% of the isolates of Xanthomonas oryzae pv. Oryzae(Xoo). One hundred and eighty eight varieties (92.61%) showed resistance to less than 50% of the tested isolates of Xoo. Among these varieties, Teqingxuanhui and Huangsiguizhan were highly resistant to both M. grisea and Xoo. It was observed that Jinxibai was highly resistant to POX99, which should be further studied.

    Pathogenicity of tenuazonic acid isolated from Alternaria alternata on Alternanthera philoxeroides was carried out with isolated leaf puncture assay and isolated plant assay. The results suggested that, tenuazonic acid was strongly pathogenic to Alternanthera philoxeroides leaves, increasing spot diameter with the concentration increase from 31.25 to 500.00 μg·mL-1, the spot diameter was 2.41 mm at 500.00 μg·mL-1, and it was more pathogenic to the younger leaves in plant tip than the ones in other plant parts at 20.00 μg·mL-1. Tenuazonic acid inhibited the growth of A. philoxeroides plants, growth length, fresh weight and average root length were significantly inhibited at concentration of 125.00 μg·mL-1, root number was also significantly inhibited at concentration of 250.00 μg·mL-1; at 500.00 μg·mL-1, growth length and root number were both zero, fresh weight decreased 0.77 g. It was concluded that tenuazonic acid had the potential for bioherbicide development for the control of A. philoxeroides

    A bacteriocin-producing strain was isolated from infant feces. With the morphological observation, physiological and biochemical characteristics and sequencing of the 16S rDNA, this strain was identified to be a gram\|positive Lactobacillus plantarum. Bacteriocin activity was degraded by proteolytic enzymes (trypsin, proteinase K etc.), and it was stable at high temperatures (100℃, 30 min), in a range of pH 2-4. The bacteriocin showed antimicrobial activity against Staphylococcus aureus, Escherichia coli, Salmonella species.

    Six immunological properties,including complement C3 content,complement C4 content,alkaline phosphatase(AKP) activity,acid phosphatase (ACP) activity,superoxidde dismutase(SOD) activity and catalase(CAT) activity,were compared between the hybrid snakehead and their parents stocks. The results indicated that all of the immunological traits analyzed in maternal stock Ophiocephalus maculatu were significantly higher than those of paternal stocks Ophiocephalus argus Cantor in CAT activity(P<0.05),and lower in ACP activity(P<0.05),except complement C3,C4 content and AKP activity(P >0.05). While in hybrid stock,almost all properties were between their parents;however,the complement C3 and C4 content were lower than their parents(P >0.05) and the SOD activity was higher than their parents (P<0.05).

    To investigate the relationships between neutral volatile aroma constituents in flue-cured tobacco leaves and ecoregion or genotype, flavor components in cultivar H892 and K326 were assayed by GC/MS. The results showed that the levels of total aroma components in flue-cured tobacco leaves were significantly affected both by ecological condition and genotype, which demonstrated a decrease tendency of Henan>Guangdong>Hubei. The content of flavor components in H892 were much higher than in K326.

    By the method of correlation analysis and factor analysis, total 27 tobacco samples from 9 tobacco\|growing counties scattered in the administrative regions of Hubei Province were analyzed with data from 10 main chemical components in flue-cured tobacco leaves. A comprehensive evaluation was carried out by using the factor scores. The results indicated that the main chemical components in flue-cured tobacco leaves were highly positive-correlated with that of the others. The contribution rate of first-four characteristic roots was 91.400%, four main factors from the main chemical components in flue-cured tobacco leaves were obtained, namely the carbon nitrogen factor, combustion factor, Nit r/ Nic factor and starch factor. Comprehensive analysis can be used in judging the quality of flue-cured tobacco according to the main chemical components in flue-cured tobacco leaves, and it will provide scientific reference to flue-cured tobacco industry usability.

    Using the stepping flue\|curing bam in Henan, effects of prolonging the time of yellowing and leaf drying on neutral aroma constituents of tobacco leaves during curing were studied. The results indicated as follows:① All of the treatments which prolonged the time of yellowing and leaf drying could increase the content of neutral aroma constituents.② The content of phenylalanine products, carotenoid, cemdrenoid, neopytadiene were highest in the treatment of T2 which prolonged the time of yellowing; Compared with normal cured treatment T1, T3 which prolonged the time of leaf drying only increased the content of carotenoid and neopytadiene. The content of maillard reaction products were the highest in the treatment of T4 which prolonged the time of yellowing and leaf drying. Compared with T1, the contents of phenylalanine products, carotenoid, neopytadiene increased and the content of cemdrenoid decreased in treatment of T4.

    The genetic diversity and relationship of 44 tomato accessions were analyzed by AFLP (amplified fragment length polymorphism). Fifteen primer combinations selected from 64 primer combinations produced a total of 601 fragments,364 of which were polymorphic with a polymorphism percentage of 60.5%. This results showed that there were abundant diversities in enzyme digestion sites among different tomato varieties. These tomato germplasm resources were clustered into 5 multiple-member groups and 1 one-member group by UPGMA (unweighted pair group method with arithmetic average) based on Nei’s genetic distances. The genetic relationship of 44 tomato accessions was analyzed using the different coefficient.

    The paddy planting manners include direct seeding and mechanical transplanting. According to the characteristics of seeding quality detection technique, there are dynamic monitoring for the metering device performance and static detection for seeds planted in plugs. Dynamic monitoring technology was applied to detect seeding quality of direct seeding, and static detection technology was applied in mechanical transplanting. The main methods of dynamic monitoring and static detection for the paddy seeding quality were suggested, and the main problems existing in the development of the seeding quality detection were discussed. Due to the technology of paddy seeding quality detection was at the level of manpower, the potential methods for the paddy seeding quality detection and the further research approaches were proposed.

    Effects of humic acid on the growth and quality of Citrus medica

    Effect of different concentrations of humic acid on the growth and quality of Citrus medica was studied. The results showed that the leaf area,chlorophyll content,soluble protein content and soluble sugar content in Citrus medica treated four times by 150 mg·L-1 humic acid were significantly higher than the treatment without humic acid and those at other concentrations. Meanwhile,better growth and quality was found in Citrus medica treated eight times by 100 mg·L-1 humic acid. It indicated that,Citrus medica which was treated four times by 150 mg·L-1 humic acid,showed higher vitality and stress tolerance.

    Sediment is a very important treasury stock or releasing source of nutrients in water column,playing a significant role in aquatic ecosystems. The behavior of benthic macro?鄄invertebrate,such as feeding,excretion,burrows,biogenic structure building and so on,could change the clearance rate of sediment,surface composition,ability of deposition,microbial quantity and communities in sediment because of the living benthic macro?鄄invertebrate on the surface or in the middle of sediment;moreover,the process of nitrification and denitrification was stimulated,and the sediment nutrient fluxes were consequently affected. On the basis of a large number of documents at home and abroad,the influence of benthic macro-invertebrate bioturbation on the physical structure of sediments,microbial communities,nitrification and denitrification,stability of organic matter in sedimentsit was elaborated;and the research status of the effect of benthic macro-invertebrate on nutrient fluxes in sediment-water interface,methods of determination and calculation were introduced,in order to provide a better prediction of bioturbation effects on sediment biogeochemistry.

    Silkworm Ping 48 strain( P48 in short) was obtained by transforming of group sex ratio\|controlled letal genes into normal strain Baiyun, which was a near isogenic line of P48 strain. P48 and Baiyun strains were analyzed by amplification fragment length polymorphism (AFLP) in present paper for screening the Z chromosome markers in silkworm sex\|linked balanced lethal strain. C094, one Z linked marker, was found by DNA retrieving, cloning, sequencing, and blasting silkworm (P50) whole genome database on NCBI. PCR primers were designed to perform the difference identification among P48, P50 and their F1 hybridization. The results showed that C094 marker was genetically linked with l1 lethal gene in silkworm balanced sex\|linked lethal strain. This benefits the characterizing and cloning of lethal related genes in silkworm sex\|linked balanced lethal strain.

    Optimal subirrigation quota for cucumber at different growth stages in protected field

    The optimal irrigation quota of sectional subirrigation for cucumber in protected field was studied by measuring the yield and water use efficiency under different soil moisture ratios. The cucumber growth period was divided into two stages as earlier stage and later stage, and the irrigation was based on the upper irrigation control limit and lower control limit for each growth period. The results showed as follows: when the buried depth of subirrigation pipe was 25 cm, the planned wetting soil zone is 15-40 cm(25 cm in depth), the upper irrigation control limit and lower control limit were 6 kPa (soil water suction)and 30 kPa respectively, the optimal soil moisture ratio of planned wetting soil zone for earlier stage of cucumber was 0.37-0.38 (irrigation quota was 54-56 m3·hm-2)and later stage was 0.34-0.37 (irrigation quota was 50-55 m3·hm-2)for sandy clay loam, and at these wet ratios the yield and water use efficiency were optimalizing.

    Effect of 60Co-γ irradiation on seed vitality of Yunyan 87 and Hongda

    The seeds of two flue-cured tobacco cultivars, Yunyan 87 and Hongda, were irradiated by the different irradiation doses of 60Coγ-ray, The effect of irradiation treatment on seed vitality of Yunyan 87 and Hongda was studied. The results showed that, in the irradiation treatment of 10-300 Gy at 25℃, the seed vitality for two cultivars were both promoted, among which, 10 Gy was the optimal irradiation dose for two cultivars; The seed vitality for two cultivars were inhibited when treated with dose more than 300 Gy. The results also showed that the seed vitality of Yunyan 87 was better than Hongda, indicating Yunyan 87 had higher adaptability under irradiation treatment.

    The cut fresh branches of Eurya japonica in Zhejiang are processed as a kind of traditional forest products to be exported to Japan. In order to establish a rapid propagation system on shoot tip culture of E. japonica, the effects of different sterilization time of HgCl2 on polluted rate of explants, the effects of different 6-BA concentrations on shoot subculture and multiplication culture, the effects of different IBA concentrations, sucrose concentrations and rooting methods on rooting rate were studied. The results showed that the appropriate conditions for tissue culture and rapid propagation of E. japonica were as follows: 25 min shoot surface-sterilization with 0.2% HgCl2, WPM basal medium with 0.5 mg·L-1 6-BA, 0.05 mg·L-1 NAA, 0.2 mg·L-1 GA, 20 g·L-1 sucrose in proliferation culture, 0.5 mg·L-1 IBA, 30 g·L-1 sucrose and two-step rooting method in rooting culture. Under this condition, bud-induction rate, multiplication coefficient, rooting rate and transplant survival rate of tissue culture seedlings could reach 100%, 2.56-4.85 per time, 98% and >95% respectively.

    In order to study the cell development and maturation patterns of flue-cured tobacco, the changes in cell ultrastructure of variety NC 89 during development and maturation process were studied by electron microscope with the samples obtained in Tangjie Township, Jiaxian County, Pingdingshan City in 2008. The results showed that cell division and increment played a major role from 1 to 11 days of leaf age. From the 21st day of leaf age, the cell division stopped, and the cell volume began to increase rapidly. Cells entered exuberant functional period on the 31st day of leaf age and begun to accumulate assimilation products on the 41st day. The period from 21st day to the 41st day of leaf age was essential to the quality of tobacco leaf. During crudity stage, the cells of immature leaves could accumulate a few of assimilation products. Along with the increasing of maturity, the chloroplast and cell nuclear gradually degraded. In overmature stage, the starch granules begun to degrade.

    Avian infectious bronchitis virus strains that prevailed in 1992-2005 in Shandong Province were collected. The spike protein (S1) gene,matrix (M) protein gene and nucleocapsid (N) gene of these virus were cloned and sequenced. The sequences of S1 gene,M gene and N gene published in GenBank were also collected. The homologies and correlations in nucleotide and amino acid of S1 gene,M gene and N gene were compared and analyzed by DNAStar and SPSS 8.0 software. The results showed the correlation coefficient of nucleotide genetic variation between S1 gene and M gene,S1 gene and N gene,M gene and N gene was 0.483,0.570 and 0.620,respectively,which indicated that the genetic variation among S1M and N genes showed not only synchronism but also independence. The phylogenetic trees based on amino acid sequences of S1 and N genes from 9 Shandong IBV and 17 reference strains indicated that strains of Shandong IBV isolated during 10 years showed close genetic relationship,and the phylogenetic trees based on amino acid sequences of M gene showed that only 3 Shandong IBV had certain relationship. Thus,it was indicated that gene recombination had been observed in IBV.
