    Genetic diversity of soybean germplasm resources from Zhejiang revealed by SSR markers

    Genetic diversity of 52 soybean germplasm resources had been evaluated by 47 SSR loci in this study. The results indicated that 411 alleles were detected,and the number of alleles was from 3 to 21 and the mean alleles per locus was 8.75. The distribution of Simpson index was 0.5506-0.9357 and the mean Simpson index was 0.8061,meanwhile,the distribution of Shannon-weaver index was 0.9083-2.8440 and the mean Shannon-weaver index was 1.8386. The range of Nei-Li similarity coefficient was from 0.5547 to 0.8589 with average value of 0.6775. It indicated that these materials had abundant genetic diversity. The results of clustering showed that 52 materials were divided into three groups by setting similarity coefficient at 0.666. GroupⅠincluded 20 materials almost from Hangjiahu,Taizhou,Lishui area;GroupⅡ included 24 materials almost from Shaoxing,Jinhua,Quzhou,Lishui area;Group Ⅲ included 8 materials almost from Jinhua,Quzhou area. It indicated that the clustering of SSR had no dose relationship with origins or eco-type of the materials,but were related to plant height or growth habit

    Accumulation of anthocyanins in Arabidopsis thaliana caused by transformation with 3GT gene from wild potato

    To verify the function of 3GT(UDP-glucose:flavonoid 3-O-glucosyltransferase) gene isolated from wild potato,the 3GT gene was inserted into the plant expression vector pG3GT with CaMV 35S promoter,and then was introduced into Arabidopsis thaliana by Agrobacterium GV3101 through floral dip method. There were four transformants selected for their ability to grow on medium containing 50 mg/L kanamycin. The frequency of transformants was 0.13%. There were three survival transformants conformed by PCR and two transformants conformed by Southern blotting analysis. Only simple copy of 3GT gene had been transferred into the Arabidopsis thaliana genome conformed by Southern blotting analysis. There was one transformant of them whose stem and leaves turned purple,the content of anthocyanin was 7.01 times of the wild type.

    To detect EST-SSR markers in pea,a capillary electrophoresis with fluorescence detection was developed and evaluated based on fluorescence labeled EST-SSR primers and MegaBACE 1000 DNA analysis system. The result showed high precision and reproducibility in determining pea EST-SSR alleles by this detection system. Furthermore,the detection and analysis of 192 samples could be automatically completed within two hours by two different-color fluorescences labelled EST-SSR primers. Therefore,it is suggested that the capillary electrophoresis with fluorescence detection developed in our study with high throughput,efficiency.

    Study on the sensitivity of PCR detection for Bt63 rice and its products

    Comparative studies were performed on the sensitivity of general versus real-time PCR to analyze Bt63 genetically modified rice. Based on the DNA sequences of rice endogenous gene SPS and structure-specific gene Btc,we selected specific primers and fluorescent probes in attempt to identify Bt63 rice. The results showed good specificity which could be capable of identifying various species. In the sensitivity assay,Bt63 rice was serially diluted according to their mass percentage and then DNA was extracted for PCR amplification. As a result,fluorescent quantative PCR displayed tenfold higher sensitivity than general PCR,whose sensitivity values were 0.1% and 0.01%,respectively. Two types of PCR methods were applied to analyze the food samples,both of which turned out to be good for effective amplification of SPS gene,while Btc gene retained negative. The real-time PCR method avoids the process of gel electrophoresis,with higher security and simplicity,therefore,in our actual daily test,real-time PCR detection of Bt63 genetically modified rice and its products was a fast,sensitive and accurate testing method.

    Construction and application of DNA fingerprint database of rice varieties in Zhejiang Province

    A total of 98 major rice varieties in Zhejiang Province from 2002 to 2006 and 155 rice varieties in Zhejiang provincial regional trial from 2007 to 2008 were assayed with a set of DNA fingerprint testing system on rice variety. A DNA fingerprint database containing genotypic data of 279 rice variety times 12 marker loci was constructed. The polymorphism of different markers and genetic similarity of varieties in the DNA fingerprint database were analyzed. Based on the DNA fingerprint database,the distinctness of all rice varieties and consistency of the same varieties in different years were identified. It was found that 11 varieties of 5 groups had no obvious distinctness within groups,and discrepancy existed in the same variety in different years.

    Newcastle disease(ND) is recognized as one of the most important poultry diseases in the world. Along with the extensive use of live vaccine against NDV,newcastle disease virus isolated from the field is often a combination of different newcastle disease. In order to avoid the interference among different strains,it is necessary to purify and distinguish the different NDV strains through the plaque. Newcastle GM2 isolates were purified by plaque-purification in this study,and the biological characteristics of the virus before and after plaque-purification were determined. The result showed that MDT,ICPI,IVPI of the Newcastle GM2 isolates before plaque purification was 50.5,1.78 and 2.41,whereas after plaque-purification MDT,ICPI,IVPI was 47.5,1.89 and 2.65,respectively.

    Estrogen receptor as a candidate gene for prolificacy of Hu sheep

    Single nucleotide polymorphism in exon 1 of the estrogen receptor (ESR) gene was detected by PCR-SSCP in high fecundity Hu sheep breed. The results indicated that there were three genotypes (AA,BB,AB) in Hu sheep and the frequency of allele A and B was 0.554 and 0.446,respectively. Comparison with the AA genotype,sequencing revealed a C→G mutation at 363 bp of exon 1 of ESR gene in the BB genotype. The Hu sheep with AB genotype or BB genotype had 0.98 and 1.47 more lambs than those with AA genotype,respectively. These results showed that the estrogen receptor locus was a major gene that controlled the prolificacy in Hu sheep or a mark in close linkage with such a gene.

    The changes of nitrogen and compost temperature by adding different contents of rice chaff during pig manure composting have been studied. The results showed that it could obviously improve fermented temperature during early composting period and could accelerate the maturity of composting by inoculating microorganism agent. Adding different contents of rice chaff could decrease nitrogen loss of pig manure and bad smell emission at different degrees in composting process,and 20% rice chaff was the best additional amount. After compost fermentation,the content of NO3--N decreased,while the content of NH4+-N showed significant difference with different treatments.

    In order to further investigate the antioxidant ability of bayberry juice,four varieties of the fruit from Ningbo,Zhejiang Province were selected as experimental materials in this study. The abilities of scavenging·OH radical were compared among bayberry juice,orange juice,apple juice,and pineapple juice. The ability of scavenging DPPH radical by bayberry juice extracts was in contrast with that of catechel,Vitamin C,and rutin. The inhibitory effects of bayberry juice extracts against lipid oxidization were also investigated. The results indicated that the ability of scavenging·OH radical by bayberry juice was obviously higher than those of other three kinds of juice. The bayberry juice extracts exhibited higher ability in scavenging DPPH radical,with its IC50 value 20.2 mg·mL-1. The juice extracts from all four bayberry varieties could inhibit lipid oxidization to a certain degree,and among them,‘Chida’variety exihibited the highest antioxidant ability with its highest content of total phenols.

    Detection of photoperiod-sensitivity for photo-sensitive genic male sterile lines in rice

    We have developed an identification method for the PGMS (photo-sensitive genic male sterile) lines under the natural day length. A comparison of genetic sensibility of the eight rice genotypes to longday and temperature was performed with the AMMI(additive main effects and multiplicative interaction)model,in which the longday and temperature at sensitive stages of the fertility were divided into 6 and 11 gradients,respectively,the results showed that these eight lines grown under the natural condition in Hangzhou belong to PGMS type,with the following characteristics:10-20 d before heading was the sensitive stage for the fertility transformation,3.0-13.8 h for critical day-length was 13.0-13.8 h,25 ℃ and 21 ℃ for the critical high temperature and critical low temperature of the fertility,respectively.

    Storage characters of tomato fruit ‘Hangza NO.2’

    The dynamic changes in the storage characters of ‘Hangza NO.2’ tomato,compared with ‘Shanghai 903’,were investigated. The index of fruit firmness,weight loss rate,rot rate and so on were examined. The results showed that the dynamic changes of two varieties showed a similar trend. Flavor quality and nutrient quality of ‘Hangza NO.2’ were superior to ‘Shanghai 903’ during the storage period,and the storage time of ‘Hangza NO.2’ was six days longer than that of ‘Shanghai 903’.

    Comparative anatomy analysis of the leaf thickness changes in tomato

    Using the instrument developed by ourselves,the diurnal changes of tomato’s leaf thickness were acquired,which reduced around midday and increased in the morning as well as evening. In order to investigate the reason for this change,the comparison dissection analysis was carried on by the paraffin section with the samples continuously collected at the same time interval. The results indicated that the palisade tissue presented a ‘loose-close-loose’ phenomenon from dawn to dusk. Although there was no obvious change in the arrangement of the spongy tissue,the change of leaf thickness was consistent with the proportion of spongy tissue,which indicated that the spongy tissue’s volume directly affected the value of leaf thickness.

    The temporal-spatial dynamic changes of pesticide residues and the risk evaluation of the vegetable quality and safety in Cixi city was studied. Twenty kinds of pesticide residues in seven kinds of vegetables from the vegetable production base,wholesale market and individual peasant household in Cixi were investigated quarterly during a whole year. The results showed that dynamic change trend of pesticide residues first increased and then decreased in 2008. The exceeding standard rates of legume vegetables,stalk and stem vegetables and leafy vegetables were higher than those of other kinds of vegetables,and the pesticide residues of vegetables from individual peasant household were higher than those from wholesale market and vegetable production base. It was indicated that acephate,fenvalerate,triazophos,cyhalothrin,chlorpyrifos and omethoate were the primary factors affecting the safety of vegetable quality in Cixi from the evaluation of safety index and comprehensive pesticide pollution value.

    Effect of high and low temperature stress on the enzyme activity and anatomy characteristics in the abscission zone of Phaseolus lunatus L.

    The drop of flowers and pods,and the physiological and anatomy characteristics of abscission zone of Phaseolus lunatus L. under high and low temperature stress were studied. Compared with the control,the drop rate of flowers and pods under high and low temperature stress was significantly increased by 262.90% and 202.69%,respectively. The content of soluble protein was significantly decreased by 34.10% and 43.33%,respectively. The activities of cellulase,polygalacturonase and peroxidase in the abscission zone were all significantly increased after high and low temperature stress. And the activity of these enzyme were higher under low temperature stress than high temperature stress,as well as the rate of the drop of flowers and pods. The analysis of anatomy charateristics showed that the volume of abscission zone cell became large and the cavity formed between cells under high and low temperature stress,which was beneficial to the fracture and shedding of the fascicular.

    Effect of simulated acid rain on the bioavailability of heavy metals from the vegetable soil amended with swine manure

    Incubation experiment was employed to study the effects of simulated acid rain on availability concentrations of heavy metals from the vegetable soils amended with metal-rich swine manure. The results indicated that the acid rain affected the availability concentrations of heavy metals by decreasing the pH of soil. The available concentrations of Zn,Cd,Pb increased with the increasing of H+ concentration,whereas the opposite trend was observed for As. Furthermore,vegetable soil amended with metal-enriched swine manure as well as the inorganic heavy metals,would result in significantly higher available concentrations of Cu,Zn,and As. Therefore,the bioavailability of heavy metals from vegetable soil amended with either the manure or the inorganic heavy metal,would be affected by acid rain and exhibit potential risks to environment. As a result,long-term heavy application of metal-rich swine manures to vegetable soils in the southern areas subjected to frequent acid rain deposition would not only improve the nutrient conditions of vegetable soil,but also increase a potential risk of heavy metal contamination to soil environment and vegetables.

    Effects of different substrates on the growth of Vallisneria natans

    In the indoor experiment,the seeds of Vallisneria natans were sown on the different substrates in order to study the sprout,morphology and biomass allocation in V. natans. The results showed that compared with the seeds of V. natans grown in thin layer of sponge and palm,the seeds grown in artificial silk cotton had more new leaves and bigger length of the longest root as well as total length of stem and leaf. However,some of V. natans grown in control group without substrate floated on water surface with wither leaves and they had lower growth parameters than other groups. In spite of the smallest number of roots,V. natans grown in control group had a higher biomass ratio of root to shoot and a larger number of plants with curved stem. The survival rate of V. natans grown in artificial silk cotton was bigger than other groups. And the survival rate of V. natans grown in artificial silk cotton,thin layer of sponge,palm and control group were 91%,88%,69% and 21% respectively. In conclusion,the growth and morphology of V. natans was greatly affected by different substrates. Without substrate,V. natans was likely to float on water surface and die;meanwhile,V. natans grown in the artificial silk cotton with loosen structure grew well. Considering the stability of root adhension and physical-chemical change of substrate submerged in water,the thin layer of sponge was the best substrate for V. natans in the indoor experiment.

    Residual dynamics of chlorpyrifos in cabbage

    In order to definitude the countermeasure for effective control of chlorpyrifos residues in cabbage,the degradation and residual dymanic changes of chlorpyrifos in cabbage under different cultivation methods were studied. The result showed that the half-life of chlorpyrifos in cabbage was 1.650~1.824 d,the residue in cabbage was related to the cultivation methods of cabbage. The cabbage was sprayed once with chlorpyrifos 40% EC using the dose of 25 mL/667 m2, and the final residue in the cabbage under open cultivation was less than 0.1 mg·kg-1 after 7 days. The safety interval of chlorpyrifos in cabbage under protected cultivation before harvest should be extended to an appropriate level.

    In this research,carbonitrides and mineral elements of flue-cured tobacco produced from Junlian,Xingwen,Gongxian,Pingshan were analyzed by employing different statistic methods. The data indicated that nitrogen compounds and mineral elements,such as potassium,zinc and iron in different parts of flue-cured tobacco were significantly different while carbohydrate and mineral elements like chlorine,sulfur and phosphorus were not significantly different. The level of total sugar,reducing sugar,nicotine and potassium varied greatly from year to year. The chemical elements in tobacco varied considerably in different regions.

    The urban-rural relationship was the most important relationship in the development of humanity economic society. Zhejiang was a pioneer in China's market-oriented rural reform,and it was also a model in the reform and exploration of the coordination construction between the urban and rural areas. On the basis of analyzing and appraising Zhejiang’s great achievements in the coordination construction between the urban and rural areas and the new rural construction,the experience in the process was summarized. The farmers were encouraged to actively find out jobs and employ themselves in the process of urban-rural interaction. The income of farmers were increased in the process of industrial structure adjustment in urban and rural arrangement and the reform of distribution system. The appearance of villages and the new community construction were promoted in the process of town planning and regulative construction predominated by governments. The problems of people's livelihood could be solved by strengthening the basic and public service in countryside. The new rural construction could be promoted in the process of establishing the co-construction mechanism with the leading of party and government and the comprehensive involvement of farmers and society. It was necessary to cultivate the pioneer for the new rural construction,in order to promote the construction of harmony society.

    Occurrence of the key diseases and insect pests in intensive rice system

    The occurrence of the key diseases and insect pests in single crop rice system under intensified cultivation has been investigated. Effects of pesticides on non-target diseases and insect pests when the pesticides were used to control targets in rice field was also studied. The results showed that the populations of brown planthopper (Nilaparvata lugens),whitebacked planthopper (Sogatella furcifera),rice leaf folder (Cnaphalocrocis medinalis) and striped stem borer (Chilo suppressalis) under intensified rice cultivation system were increased by 21.7%,27.19%,57.95% and 200.0%,respectively,while the rice sheath blight (Rhizoctonia solani) disease index was increased by 29.10% compared with that under conventional rice cultivation system. Non-target diseases and insect pests were obviously affected by spraying pesticides such as dimehypo,indoxacarb,validamycin,difenoconazole and propiconazole to control striped stem borer,rice leaf folder and rice sheath blight,and the populations of brown planthopper and whitebacked planthopper were all stimulated and proliferated with the increase of 26%-60% than that in conventional rice cultivation system.

    Grey relative analysis between the egg freshness and its influencing factors

    Freshness is an important index for the measurement of egg’s quality and it is affected by different factors during storage. In this study,the grey relative degrees between the Haugh Unit and influencing factors of egg freshness were calculated based on the grey theory. And then,the importance of influencing factors was ranked according to the grey relative degree. Furthermore,the relations between Haugh Unit and these factors were analyzed theoretically. It was concluded that egg specific gravity was the first important influencing factor,while the egg freshness was affected by the eggshell strength most slightly. With the increase of storage time,the importance of egg-shaped index became weaken,but the egg weight factor was becoming more important. The results suggested that the grey theory presented in this paper might provide a new and effective way for comprehensive analysis of the relationship between egg freshness and influencing factors.

    Technology of grasp fruit and vegetable based on external force control loops

    The open architecture force control system of manipulator grasping was designed based on PC. Grasp force gradually turned small because of the deformation of fruit and vegetable in the process of manipulator grasping. It was necessary to consider the effect of deformation on grasp force,so the external force control loops algorithm was proposed for the control system in experiment. It converted the force error into the speed input command of position control system. The manipulator speed was adaptively decelerated when the grasping force increased in order to reduce force overshoot. Proportional and incremental proportional integral algorithm were adapted to force controller in experiment,both algorithms proved that speed of manipulator would automatically decelerate when grasping force increased. The results showed that it was a good control algorithm by automatic variable speed of manipulator grasping based on external force control loops,which could reduce grasp force overshoot and decrease the damage of fruit and vegetable caused by agricultural robot grasping.

    Traceability of genetically modified food is an important part in food safety. EU had established the management framework of genetically modified food traceability by regulation (EC) NO 1830/2003,and further regulated it by regulation (EC)1829/2003 in managing authorisation and supervision,and labelling of genetically modified food. Moreover,regulation (EC) 65/2004 established a system of unique identifiers for genetically modified food. The genetically modified food traceability reflected the management conception of “from farm to table” in EU. This article introduced the management framework of EU genetically modified food traceability,which was worth using for reference in China.

    RNA silencing is a phenomenon of homologous RNA degradation induced by dsRNA,which works as a natural virus-resistant mechanism in plant. However,in the long co-evolution progress,a number of inhibitory proteins coded by virus in the process of siRNA or miRNA silencing could antagonize and inhibit this mechanism. The discovery,process and inhibition of RNA silencing,as well as the research advances in plant virus-resistance induced by RNA silencing were reviewed. Several plant disease-resistance mechanism were discussed,inclulding the resistance of RNA viruses and DNA viruses mediated by RNA silencing and the virus resistance mediated by virus transgenes. Meanwhile,the problems existed in the research were properly analyzed,and the key points and the research directions of future prospect were discussed.
